Abstract
Compositions having antimicrobial activity against a variety of microorganisms, including vegetative bacteria, bacterial spores, fungi, and fungal spores are particularly useful for microbiocidal treatments of a variety of substances. More specifically, compositions have antibmicrobial activity against microorganisms of the Bacillus cereus group such as Bacillu s cereus, Bacillus mycoides, Bacillus anthracis, and Bacillus thuringiensis ar e particularly useful. Compositions including hydrogen peroxide, a carboxylic acid, and a peroxycarboxylic acid in which the weight ratio of the preroxycarboxylic acid to the hydrogen peroxide is at least 4:1 are effectiv e against microorganims, particularly bacterial spores. Such compositions include a reduced amount of hydrogen peroxide relative to the amount of peroxycarboxylic acid as compared to conventional compositions. Compositions can also inlclude a quaternary ammonium compound, a stabilizing agent, a surfactant, a hydrotrope, or other additives. Methods of using a composition including hydrogen peroxide, a carboxylic acid, and a peroxycarboxylic acid in which the ratio of the peroxycarboxylic acid to the hydrogen peroxide is at least 4:1 are useful for reducing the microbial numbers on a variety of substances contaminated by microorganisms, particularly of the Bacillus cereusgroup. Such substances include foodstuffs, water, general-premise surfaces, specific-equipment surfaces, animal carcasses, soil, and textiles.
Claims (85)
1. A composition having antimicrobial activity against a spore or related spore-forming microorganism, said composition comprising:
(a) an effective antimicrobial amount of hydrogen peroxide;
(b) an effective antimicrobial amount of a carboxylic acid of the formula:
R(COOH)n, wherein R comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3; and (c) an effective antimicrobial amount of a peroxycarboxylic acid of the formula: R(COOOH)n, wherein R comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3, wherein there are about four parts or greater by weight of the peroxycarboxylic acid for each part of hydrogen peroxide.
(a) an effective antimicrobial amount of hydrogen peroxide;
(b) an effective antimicrobial amount of a carboxylic acid of the formula:
R(COOH)n, wherein R comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3; and (c) an effective antimicrobial amount of a peroxycarboxylic acid of the formula: R(COOOH)n, wherein R comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3, wherein there are about four parts or greater by weight of the peroxycarboxylic acid for each part of hydrogen peroxide.
2. The composition of claim1, wherein the composition has antimicrobial activity against spore and related microorganism of the Bacillus cereus group .
3. The composition of claim1, wherein the composition has antimicrobial activity against spore and related vegetative Bacillus cereus, Bacillus anthracis, Bacillus thuringiensis, mixed microorganisms thereof, and associated spores thereof.
4. The composition of claim1, wherein the composition is diluted with water at a ratio of greater than one part by volume of the composition of claim 1 per each 100 parts by volume of diluent.
5. The composition of claim1, wherein the composition comprises hydrogen peroxide in an amount of up to about 2500 ppm.
6. The composition of claim 1, wherein the composition is a concentrate composition, comprising hydrogen peroxide in an amount of between about 0.2 weight percent and about 6 weight percent.
7. The composition of claim 1, wherein the composition comprises the carboxylic acid in an amount of between about 2 ppm and about 27000 ppm.
8. The composition of claim 1, wherein the composition comprises the carboxylic acid in an amount of between about 50 ppm and about 5000 ppm.
9. The composition of claim 1, wherein the composition is a concentrate composition comprising a carboxylic acid in an amount of between about 0.5 weight percent and about 80 weight percent.
10. The composition of claim 1, wherein the composition comprises peroxycarboxylic acid in an amount of between about 1 ppm and about 10000 ppm.
11. The composition of claim 1, wherein the composition is a concentrate composition comprising peroxycarboxylic acid in an amount of between about 0.2 weight percent and about 30 weight percent.
12. The composition of claim 1, wherein the peroxycarboxylic acid comprises peroxyacetic acid, peroxyoctanoic acid, peroxypropionic acid, peroxybutyric acid, peroxyglycolic acid, peroxyglutaric acid, peroxysuccinic acid, peroxylactic acid, peroxycitric acid, peroxydecanoic acid, peroxyformic acid, peroxypentanoic acid, peroxyheptanoic acid, peroxynonanoic acid, peroxyundecanoic acid, peroxydodecanoic acid, or mixtures thereof.
13. The composition of claim 1, wherein the peroxycarboxylic acid comprises an R that is C1-4 alkyl.
14. The composition of claim 13, further comprising a peroxycarboxylic acid comprising an R that is C5-12 alkyl.
15. The composition of claim 14, wherein in the composition the peroxycarboxylic acid comprising R that is C5-12 alkyl is present in an amount of between about 3 ppm and about 500 ppm.
16. The composition of claim 14, wherein the composition is a concentrate composition and the peroxycarboxylic acid comprising an R that is C5-12 alkyl is present in an amount of between about 0.5 weight percent and about 25 weight percent.
17. The composition of claim 14, wherein the composition comprises peroxyacetic acid and peroxyoctanoic acid.
18. The composition of claim 1, further comprising a quaternary ammonium compound, a stabilizing agent, a hydrotrope, a surfactant, or a mixture thereof.
19. The composition of claim 2, further comprising a quaternary ammonium compound, the composition having improved activity against a fungi.
20. The composition of claim 2 wherein the quaternary ammonium compound is present in an amount of between about 4 ppm and about 100 ppm
21. The composition of claim 19, wherein the quaternary ammonium compound comprises N-alkyl dimethyl ethyl benzyl ammonium, di-n-alkyl dimethyl ammonium, di-n-alkyl methyl benzyl ammonium salt, or a mixture thereof.
22. The composition of claim 21, wherein the composition additionally has antimicrobial activity against fungi.
23. The composition of claim 1, further comprising a stabilizing agent in an amount of between about 2 ppm and about 100 ppm.
24. The composition of claim 23, wherein the stabilizing agent is a phosphonic acid or salt thereof.
25. The composition of claim 23, wherein the stabilizing agent comprises 1-hydroxyethylidene-1,1-diphosphonic acid or salt thereof.
26. The composition of claim 1, wherein there are greater than five parts by weight of peroxycarboxylic acid for each part by weight hydrogen peroxide.
27. A concentrate composition having antimicrobial activity against a spore or related spore-forming microorganism, the composition comprising (a) hydrogen peroxide in an amount of between about 0.2 weight percent and about 6 weight percent;
(b) a carboxylic acid of the formula: R(COOH)n, wherein R comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3, the carboxylic acid being present in an amount of between about 0.5 weight percent and about 80 weight percent; and (c) a peroxycarboxylic acid of the formula: R(COOOH)n, wherein R
comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3, the peroxycarboxylic acid being present in an amount of between about 0.2 weight percent and about 30 weight percent, wherein there are four parts or greater by weight of peroxycarboxylic acid for each part by weight hydrogen peroxide.
(b) a carboxylic acid of the formula: R(COOH)n, wherein R comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3, the carboxylic acid being present in an amount of between about 0.5 weight percent and about 80 weight percent; and (c) a peroxycarboxylic acid of the formula: R(COOOH)n, wherein R
comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3, the peroxycarboxylic acid being present in an amount of between about 0.2 weight percent and about 30 weight percent, wherein there are four parts or greater by weight of peroxycarboxylic acid for each part by weight hydrogen peroxide.
28. The composition of claim 27, wherein the composition has antimicrobial activity against a spore or related spore-forming microorganism of the Bacillus cereus group.
29. The composition of claim 28, wherein the composition has antimicrobial activity against a spore or related spore-forming microorganism comprising Bacillus cereus, Bacillus anthracis, Bacillus thuringiensis, mixed microorganisms thereof, and associated spores thereof.
30. The composition of claim 27, wherein there are greater than five parts by weight of peroxycarboxylic acid for each part by weight hydrogen peroxide.
31. The composition of claim 27, wherein there are greater than six parts by weight of peroxycarboxylic acid for each part by weight hydrogen peroxide
32. The composition of claim 27, wherein the composition comprises hydrogen peroxide in an amount of between about 0.5 weight percent and about 5 weight percent.
33. The composition of claim 27, wherein the composition comprises the carboxylic acid in an amount of between about 10 weight percent and about 70 weight percent.
34. The composition of claim 27, wherein the composition comprises the peroxycarboxylic acid in an amount of between about 2.5 weight percent and about 25 weight percent.
35. The composition of claim 27, wherein the peroxycarboxylic acid comprises peroxyacetic acid, peroxyoctanoic acid, peroxypropionic acid, peroxybutyric acid, peroxyglycolic acid, peroxyglutaric acid, peroxysuccinic acid, peroxylactic acid, peroxycitric acid, peroxydecanoic acid, peroxyformic acid, peroxypentanoic acid, peroxyheptanoic acid, peroxynonanoic acid, peroxyundecanoic acid, peroxydodecanoic acid, or mixtures thereof.
36. The composition of claim 27, wherein the peroxycarboxylic acid comprises an R that is C1-4 alkyl.
37. The composition of claim 36, further comprising a peroxycarboxylic acid comprising an R that is C5-12 alkyl.
38. The composition of claim 37, wherein the peroxycarboxylic acid comprising an R that is C5-12 alkyl is present in an amount of between about 0.3 weight percent and about 5 weight percent.
39. The composition of claim 37, wherein the composition comprises peroxyacetic acid and peroxyoctanoic acid.
40. The composition of claim 27, further comprising a quaternary ammonium compound, a stabilizing agent, a hydrotrope, a surfactant, or a mixture thereof, the composition having activity against a fungi.
41. The composition of claim 27, further comprising a quaternary ammonium compound in an amount of between about 0.08 weight percent and about 7 weight percent.
42. The composition of claim 41, wherein the quaternary ammonium compound comprises N-alkyl dimethyl ethyl benzyl ammonium, di-n-alkyl dimethyl ammonium, di-n-alkyl methyl benzyl ammonium salt, or a mixture thereof.
43. The composition of claim 27, further comprising a stabilizing agent in an amount of between about 0.1 weight percent and about 5 weight percent.
44. The composition of claim 27, wherein the stabilizing agent is a phosphonic acid or a salt thereof.
45. The composition of claim 43, wherein the stabilizing agent comprises 1-hydroxyethylidene-1,1-diphosphonic acid or a salt thereof.
46. composition of claim 27, wherein the composition is diluted with water to provide a use solution, the use solution comprising:
(a) hydrogen peroxide in an amount of up to about 2500 ppm;
(b) the carboxylic acid in an amount of between about 2 ppm and about 27000 ppm; and (c) the peroxycarboxylic acid in an amount of between about 1 ppm and about 10000 ppm.

47 The composition of claim 46, wherein the composition is diluted with water at a ratio of about one ounce to six gallons of water.
48 A method of reducing the concentration of spore forming vegetative bacteria and related bacterial spores, the method comprising contacting a surface of a substance contaminated by vegetative bacteria and bacterial spores, the composition comprising:
(a) an effective antimicrobial amount of hydrogen peroxide;
(b) an effective antimicrobial amount of a carboxylic acid of the formula:
R(COOH)n, wherein R comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3; and (c) an effective antimicrobial amount of a peroxycarboxylic acid of the formula: R(COOOH)n, wherein R comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3, wherein there are greater than four parts by weight of peroxycarboxylic acid for each part by weight hydrogen peroxide.
(a) an effective antimicrobial amount of hydrogen peroxide;
(b) an effective antimicrobial amount of a carboxylic acid of the formula:
R(COOH)n, wherein R comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3; and (c) an effective antimicrobial amount of a peroxycarboxylic acid of the formula: R(COOOH)n, wherein R comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3, wherein there are greater than four parts by weight of peroxycarboxylic acid for each part by weight hydrogen peroxide.
49 The composition of claim 48, wherein the composition has antimicrobial activity against a spore or related spore-forming microorganism of the Bacillus cereus group.
50. The method of claim 48, wherein the vegetative bacteria and bacterial spores comprise Bacillus cereus, Bacillus mycoides, Bacillus anthracis, Bacillus thuringiensis, or mixtures thereof.
51. The composition of claim 48, further comprising a quaternary ammonium compound, the composition having improved activity against a fungi.
52. The method of claim 48, wherein the substance contacted is in a health-care environment.
53. The method of claim 48, wherein the health-care environment comprises a human hospital, an animal hospital, a human clinic, an animal clinic, a nursing home, or a day-care facility.
54. The method of claim 48, wherein the substance contacted is in a food environment.
55. The method of claim 48, wherein the food environment comprises a restaurant, a food-processing plant, a food-storage plant, or a grocery store.
56. The method of claim 48, wherein the substance contacted is in a hospitality environment.
57. The method of claim 48, wherein the hospitality environment comprises a hotel, a motel, a resort, or a cruise ship.
58. The method of claim 48, wherein the substance contacted is in an education environment, the education environment comprising a school or a university.
59. The method of claim 48, wherein the substance contacted comprises a general-premise surface,
60. The method of claim 48, wherein the general-premise surface comprises a floor, a ceiling, furniture, or a mixture thereof.
61. The method of claim 48, wherein the substance contacted comprises a specific-equipment surface,
62. The method of claim 48, wherein the specific-equipment surface comprises a hard surface, manufacturing equipment, processing equipment, or a mixture thereof.
63. The method of claim 48, wherein the substance treated comprises wood or cellulose-based systems.
64. The method of claim 48, wherein the substance treated comprises foodstuffs.
65. The method of claim 48, wherein the substance comprises soil, the soil being present on a cooling tower, a flume, a hard surface, a foodstuff, or a mixture thereof.
66. The method of claim 48, wherein the substance comprises water.
67. A method of reducing the concentration of vegetative bacteria and bacterial spores on a textile, the method comprising contacting a textile with a composition, the composition comprising:
(a) an effective antimicrobial amount of hydrogen peroxide;
(b) an effective antimicrobial amount of a carboxylic acid of the formula:
R(COOH)n, wherein R comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3; and (c) an effective antimicrobial amount of a peroxycarboxylic acid of the formula: R(COOOH)n, wherein R comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3, wherein there are greater than four parts by weight of peroxycarboxylic acid for each part by weight hydrogen peroxide.
(a) an effective antimicrobial amount of hydrogen peroxide;
(b) an effective antimicrobial amount of a carboxylic acid of the formula:
R(COOH)n, wherein R comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3; and (c) an effective antimicrobial amount of a peroxycarboxylic acid of the formula: R(COOOH)n, wherein R comprises hydrogen, alkyl, alkenyl, alicyclic group, aryl, heteroaryl, or heterocyclic group, and n is 1, 2, or 3, wherein there are greater than four parts by weight of peroxycarboxylic acid for each part by weight hydrogen peroxide.
68. The composition of claim 67, wherein the composition has antimicrobial activity against a spore or related spore-forming microorganism of the Bacillus cereus group.
69. The method of claim , 67 wherein the spore or related spore-forming microorganism comprise Bacillus cereus, Bacillus mycoides, Bacillus anthracis, Bacillus thuringiensis, or mixtures thereof.
70. The composition of claim 67, further comprising a quaternary ammonium compound, the composition having improved activity against a fungi and fungal spore.
71. The method of claim 67, wherein the textile comprises cotton, wool, silk, synthetic fabric, or a mixture thereof.
72. The method of claim 67, wherein the textile comprises personal clothing.
73. The method of claim 67, wherein the textile comprises textiles found in a health-care environment, a hospitality environment, or a food environment.
74. The method of claim 67, wherein the textile is from a health-care environment, the textile comprising a towel, bedding, a lab coat, a hospital gown or apron.
75. The method of claim 73, wherein the textile is from a hospitality environment, the textile comprising bedding or a towel.
76. The method of claim 73, wherein the textile is from a food environment, the textile comprising a napkin or a tablecloth.
77. A method of improving antimicrobial activity of a first composition, the first composition comprising an amount of a carboxylic acid, an amount of a peroxycarboxylic acid, and an amount of hydrogen peroxide at a first equilibrium, the method comprising:
modifying the first composition by reducing the amount of hydrogen peroxide relative to the amount of the peroxycarboxylic acid such that the ratio of the peroxycarboxylic acid to the hydrogen peroxide in the composition is at least 4:1 to form a modified composition, wherein an improvement of the modified composition over the first composition is quantifiable by a comparative laboratory method with statistically significant results.
modifying the first composition by reducing the amount of hydrogen peroxide relative to the amount of the peroxycarboxylic acid such that the ratio of the peroxycarboxylic acid to the hydrogen peroxide in the composition is at least 4:1 to form a modified composition, wherein an improvement of the modified composition over the first composition is quantifiable by a comparative laboratory method with statistically significant results.
78. The method of claim 77 wherein the comparative method comprises:
(a) contacting a surface of a first substance being contaminated with microorganisms with the modified composition to reduce the number of microorganisms, (b) determining a first log 10 reduction of the number of microorganisms that resulted from the contacting step with the modified composition;
(c) contacting a surface of a second substance identical to the first substance with the first composition to reduce the number of microorganisms;
(d) determining a second log 10 reduction of the number of microorganisms that resulted from the contacting step with the first composition; and (e) comparing the first log 10 reduction to the second log 10 reduction, wherein the improvement of the modified composition comprises the first logo reduction being statistically significantly greater than the second log reduction.
(a) contacting a surface of a first substance being contaminated with microorganisms with the modified composition to reduce the number of microorganisms, (b) determining a first log 10 reduction of the number of microorganisms that resulted from the contacting step with the modified composition;
(c) contacting a surface of a second substance identical to the first substance with the first composition to reduce the number of microorganisms;
(d) determining a second log 10 reduction of the number of microorganisms that resulted from the contacting step with the first composition; and (e) comparing the first log 10 reduction to the second log 10 reduction, wherein the improvement of the modified composition comprises the first logo reduction being statistically significantly greater than the second log reduction.
79. The method of claim 77, wherein the modifying step comprises minimizing the amount of hydrogen peroxide.
80. The method of claim 79, wherein the amount of hydrogen peroxide is minimized by contacting the composition with a hydrogen-peroxide destroying agent.
81. The method of claim 80, wherein the hydrogen-peroxide destroying agent comprises catalase, peroxidase, or platinum metal.
82. The method of claim 77, wherein the A composition having antimicrobial activity against a spore or related spore-forming microorganism of the Bacillus cereus group.
83. The method of claim 82, wherein Bacillus cereus group of a spore or related spore-forming microorganism comprise Bacillus cereus, Bacillus cereus, Bacillus mycoides, Bacillus anthracis, Bacillus thuringiensis, or mixtures thereof.
84. The method of claim 77, further comprising contacting a surface of a first substance being contaminated with the modified composition to reduce the number of microorganisms after the modifying step.
85. The method of claim 77, wherein the modified composition has a ratio of the peroxycarboxylic acid to hydrogen peroxide is at least 5:1.
