Abstract

发明背景
本发明通常涉及药物和其它试剂的释放。更具体地说,本发明涉及使用口香糖制品释放的药物和试剂。
当然,已知可为了各种目的而向个体提供试剂。这些试剂可用于治疗疾病并且本身典型地称之为药物或药剂。同样,药物或药剂可用于预防目的。甚至已知为了各种非医疗目的而向个体提供试剂,这些目的包括增强行为或者保持或引发机敏。
已有大量这类试剂存在。这些试剂包括了从例如咖啡因的兴奋剂到例如止痛剂、安定药、心血管产品、胰岛素等的药物的全部。这些试剂按照所需基础服用,同时个体必需定时间隔地服用其它试剂。
典型地,不经肠道或经肠道使用药物(药剂)。当然,不经肠道使用为药物直接经静脉注射进入血流的给药。经肠道是指将药物给到胃肠道中。在这两者情况任一中,使用药物的目的是将药物从给药位置移动到体循环中。
除了静脉注射给药外,药物在到达体循环之前必需穿过几个半透细胞膜。这些膜起生物屏障作用,阻止药物分子通过。据信有四种方法将药物穿过生物屏障:被动扩散、促进扩散、主动运输和胞饮作用。
被动扩散是穿过细胞膜的运输,其中移动用的驱动力是溶质的浓度梯度。当口服药物时,该吸收发生在小肠内。促进扩散据信是以在细胞膜外部与底物分子可逆地组成的载体组分为基础。该载体底物复合物快速扩散通过该膜,并在内表面释放该底物。主动运输需要细胞的能量消耗,并似乎限制了与正常体组分的结构相似的试剂。这些试剂在小肠内经常从特定位置吸收。胞饮作用是指通过细胞卷入颗粒或流体。据信在药物运输中起次要作用。Merck Manual,第16版第2598-2599页。
当测定使用药物治疗疾病的药物功效和有效性时,关键是药物吸收。药物吸收是指药物从给药位置到体循环移动的过程。
口服药物是至今为止最普通的方法。当口服时,由于细胞在胃肠道内通过上皮细胞膜的运输,因此经常发生药物吸收。口服后的吸收被许多因素阻隔。这些因素包括沿消化器官烛煤(cannel)在以下方面的差异:腔的pH;表面积/腔体积;组织灌注、胆汁、和粘液流动;以及上皮膜。参见Merck Manual第2599页。
影响口服药物吸收的另一方面是药物的形式。大多数口服的药物为片状或胶囊状。这主要是为了方便、经济、稳定和患者接受性起见。因此,这些胶囊或片剂在能够发生吸收之前必需崩解或溶解。有各种因素能够改变或阻碍固体剂量形式崩解。而且,有各种因素影响溶解速度并因此决定了药物吸收的可获得性。参见Merck Manual第2600页。
非肠道给药允许直接将药物放在血流中。这样经常确保了剂量完全释放到体循环中。然而,通过要求药物通过一个或多个生物膜转移到达血流的路线的给药预先排除了所有药物最终被吸收的保证。甚至进行非肠道给药时,由于毛细血管趋于高度多孔,因此灌注(血流/克组织)为吸收速度的主要因素。因此,注射位置能够显著影响药物的吸收速度;例如,地西泮注射型IM进入血流差的位置的吸收速度可能比进行口服时慢得多。参见Merck Manual第2601页。
在药物释放过程中不仅药物吸收是关键,而且药物的生物利用性也是关键。生物利用性定义为活性部分(药物或代谢物)进入体循环,由此到达作用位置的速度和程度。生物利用性取决于许多因素,包括药物产品是如何设计和生产的,其生物化学性能,和与患者生理和病理有关的因素。参见Merck Manual第2602页。
当药物从药物产品中快速溶解并准备通过膜时,从大多数给药位置的吸收趋于结束。这不总是口服给药的情形。在到达静脉腔之前,药物必需沿消化道移动并通过肠壁和肝,他们都是药物代谢的常规位置。因此,药物在其可以在体循环中测出之前可以被代谢。这种药物输入的减少称之为第一通道影响。大量药物因过度的第一通道代谢而呈现出低的生物利用性。降低生物利用性的其它两个最常见的原因是在胃肠道中的时间不足和存在竞争反应。参见Merck Manual第2602页。
就口服药物而言最常见的是考虑生物利用性。生物利用性的差异意味着临床重要性。
尽管非肠道给药的确提供了一种消除大量口服时存在的变量的方法,但是非肠道给药并不是优选路径。典型地,非肠道给药需要使用医疗人员并且对大多数试剂和药物如止痛剂的给药既没授权也不可行。甚至当需要时因患者关心舒适、感染等,以及涉及的设备和成本,因此不优选非肠道给药。然而,尽管进行了最好尝试,但是某些治疗仍然需要非肠道注射的药物。例如,几十年的研究集中在通过非肠道方式将胰岛素释放给个体。尽管有这些努力,但是今天胰岛素仍然经静脉注射给药。
因此需要一种改进方法将药物和试剂释放到个体中。
发明简述
本发明涉提供了向个体释放药物和试剂的改进方法。为此,提供了含有药物或试剂的口香糖,特别是涂布过的口香糖产品。药物或试剂存在于基本上包围糖芯(水溶性部分和/或不溶性胶基部分)的涂层或外壳中。已发现,通过咀嚼该涂布过的口香糖,或者在某些情况下甚至将该涂布过的口香糖放入嘴中,该药物或试剂从口香糖中释放。据信连续咀嚼口香糖,在口腔内产生一压力迫使试剂或药物通过口腔中所含的口腔粘膜直接进入个体的体循环。这大大提高了药物在体循环中的吸收以及药物在体循环中的生物利用性。
本发明还提供了包括药物和试剂的改进制品。
为此,本发明提供了一种含有糖芯的涂布口香糖组合物。该糖芯包括水溶性部分和不溶于水的部分。涂层基本上包围了糖芯,该涂层包括至少50wt%的口香糖产品。该产品包括药物或试剂。
在一个实施方式中,该涂层包括足够量的掩蔽剂,从而改善含有药物的涂层的口感性能。该掩蔽剂可以选自:三氯半乳蔗糖、葡萄糖酸锌、乙基麦芽酚、甘氨酸、乙酰舒泛-K、阿斯巴甜、糖精、果糖、木糖醇、喷雾干燥的甘草、甜根(glycerrhizine)、葡萄糖、葡萄糖酸钠、葡糖酸δ-内酯、乙基香兰素、香兰素、正常和高效甜味剂和各种适宜风味剂。
在一个实施方式中,该涂层含有高强度甜味剂。在另一实施方式中,该高强度甜味剂选自:阿斯巴甜、三氯半乳蔗糖和乙酰舒泛-K。
在一个实施方式中,糖芯含有约30-约90wt%的水不溶性胶基。
在一个实施方式中,根据该药物,该制品在唾液中产生至少5ppm-约66wt%药物的唾液含量。
在一个实施方式中,该涂层含有高达75wt%的口香糖组合物。
在一个实施方式中,该涂层为一再结晶颗粒涂层。
在一个实施方式中,该涂层为非晶形涂层。
在一个实施方式中,该涂层为一粉末涂层。
在一个实施方式中,将该口香糖咀嚼至少2分钟。
在一个实施方式中,该药物选自:止痛剂、肌肉松弛剂、制酸剂、抗组胺剂、减充血剂、抗炎剂、抗生素、抗病毒剂、精神治疗剂、胰岛素和心血管药。
在一个实施方式中,将含有药物的口香糖每天咀嚼至少2次。
在一个实施方式中,一次咀嚼两片口香糖。
在本发明的另一个实施方式中,提供了一种释放药物的方法。该方法包括步骤:提供含有占口香糖至少50wt%的涂层的口香糖,该涂层含有基本上围绕糖芯的药物;咀嚼该口香糖,使药物从口香糖组合物中释放到咀嚼者的口腔中;并连续咀嚼该口香糖,由此产生使药物通过口腔中所含的口腔粘膜进入咀嚼者体循环的液压。

本优选实施方式的详细说明
本发明提供了向个体释放药物和其它试剂的改进方法以及含有这些药物和试剂的改进制品。
根据本发明,将药物或试剂含在围绕口香糖制品的涂层中。该涂层占整个口香糖产品重量的至少50%。当咀嚼口香糖时,药物或试剂释放到唾液中。在连续咀嚼过程中,因咀嚼产生的压力,然后迫使唾液中的药物或试剂通过口腔中的口腔粘膜。口腔粘膜具有一薄的上皮并富含血管。因此,口腔粘膜易于吸收药物。与典型口服咽下的药物相反,其中溶液接触过于简短而无法通过口腔粘膜感知吸收,据信在咀嚼期间,试剂和/或药物保持在口腔中并被迫通过口腔粘膜。还出人意料地发现,与常规口服相比,使得药物吸收增加,并使药物的生物利用性也增加。已发现药物或试剂的吸收比常规口服吞咽时相比要快得多。事实上,吸收接近非肠道给药,并且生物利用性也比口服大得多。
参照图1,描述了本发明口香糖组合物10的一个实施方式。正如所述的,该口香糖组合物10含有糖芯12。该糖芯可以为本领域已知的任意口香糖制品。尽管正如下面所述的,优选该糖芯的水不溶性胶基含量比常用的高。根据本发明,围绕该糖芯12为一涂层14。该涂层14含有药物或其它活性剂。
现在参照涂层14,涂层14占口香糖组合物重量的至少50%。优选该涂层占口香糖组合物重量的约50%-约75%,并且在一优选实施方式中,该涂层占产品重量的约67%。可以利用各种涂层。例如,该涂层可以为软的非晶形涂层。或者,该涂层可以为再结晶颗粒涂层。正如下面所讨论的,在一优选实施方式中,该涂层以糖浆/粉末组合物使用。
优选,该涂层含有掩蔽剂。就此而言,可以使用高强度甜味剂和适宜风味剂掩蔽因药物或试剂存在的任意异味。已发现,就可能具有涩味或苦味的某些药物或试剂而言,通过向该制品中加入掩蔽剂,可以提供含有药物的更可口制品。就此而言,甚至药物,例如其粉末形式可能苦或者具有令人不快的味道,但是用作本发明的涂层的基质当含有该掩蔽剂时,将赋予产品可接受的口感性能。例如,已出人意料地发现,通过将药物粉末化基质和掩蔽剂溶解,该掩蔽剂掩盖药物或试剂产生的苦味和不良风味的能力增加。通过以药物所产生的不良风味或异味为基础选择特定的掩蔽剂,可以提供可口的制品。
例如,如果人们想掩盖例如阿斯匹林的涩味,人们可以使用经发现能有效抗涩味的掩蔽剂例如果糖和高强度甜味剂,例如糖精、阿斯巴甜和乙酰舒泛-K。当为苦味适中的活性组分如咖啡因时,人们应使用组分例如甘氨酸、乙基麦芽酚、葡萄糖酸锌、甘草粉末、高强度甜味剂等。当为不良味非常重的活性组分如对乙酰氨基酚时,已发现薄荷的作用非常好,但是可能需要增加使用高强度甜味剂如阿斯巴甜。
在一个实施方式中,这些掩蔽剂选自:三氯半乳蔗糖、葡萄糖酸锌、乙基麦芽酚、甘氨酸、乙酰舒泛-K、阿斯巴甜、糖精、果糖、木糖醇、麦芽糖醇、异麦芽糖、盐、喷雾干燥的甘草、glycerrhizin、葡萄糖、葡萄糖酸钠、蔗糖、葡糖酸δ-内酯、乙基香兰素和香兰素。
在本发明的一个实施方式中,在涂层中使用足够的掩蔽剂,从而提高并赋予口香糖产品可接受的口感性能。本文所用的赋予“可接受的口感性能”意思是该口香糖制品具有足够令人愉快、或者至少没有令人不快的味道,以使消费者能够将口香糖咀嚼至少2分钟。掩蔽剂是否必需和/或掩蔽剂的量将根据异味或试剂而变化。当然,如果需要的话,可以使用多种掩蔽剂,令人葡萄糖酸锌和甜味剂或风味剂。在一个实施方式中,掩蔽剂可以占涂层制品重量的约30%-约99%。
在一优选实施方式中,涂层含有例如阿斯巴甜、三氯半乳蔗糖和乙酰舒泛-K的高强度甜味剂。优选该高强度甜味剂占涂层重量的约0.5%-约5%。
如上所述,涂层将含有药物或试剂。还出人意料地发现,与个体通常口服的相比,可以将更少的药物或试剂放入口香糖中并且可以到达同样的生物当量。事实上,已出人意料地发现,在一定情况下,就至少某些药物和试剂而言,甚至与非肠道给药相比,使用口香糖通过口腔给药药物或试剂能够提供增强的效果。
例如,通常使用咖啡因作为兴奋剂减少瞌睡。它在肝中几乎完全代谢掉了,因此被列为低清除、流动独立型药物。这意思是失活速度不受向肝中释放的影响并且仅能够通过改变肝脏酶活性来改进。
咖啡因的药动力学已为公知,并且在口腔和静脉给药之间没有明显差异。然而,数据详述于下,揭示了当咖啡因通过口香糖给药时其吸收速度常数(Ka)显著增加。这意思是咖啡因以显著快的速度移到体循环中。还注意到开始动态响应时的相似变化,例如机敏和行为。
另外还出人意料地发现,就至少某些试剂而言,将该试剂放置于口香糖中可以对可能存在于体循环中的该试剂有触发效果。例如,已发现,就口腔咽下的咖啡因而言,在咽下一定量咖啡因并过去一段时间之后,咖啡因进一步消化对个体的影响忽略不计。然而,通过咀嚼含有咖啡因的口香糖,已观察到显示出与体循环中的咖啡因产生协同效果的触发效果。据信该触发效果也存在于其它试剂如止痛剂中。
假定各种不同的药物和试剂可用于该涂层中。例如,这些试剂包括(尤其是)例如咖啡因的兴奋剂。一般说来,这些药物包括(尤其是)止痛剂、抗生素、抗病毒剂、抗组胺剂、抗炎剂、减充血剂、制酸剂、肌肉松弛剂、精神治疗剂、胰岛素和心血管药。根据该药物,假定所得口香糖可用于治疗,尤其是:咳嗽、感冒、运动病、变态反应、发烧、疼痛、发炎、咽喉痛、感冒痛、窦问题、腹泻、糖尿病、抑郁症、焦虑以及其它疾病和症状。特定试剂/药物包括,例如但不限于:咖啡因、阿斯匹林、对乙酰氨基酚、布洛芬、羟基柠檬酸、啶甲酸铬、磷脂酰丝氨酸、尼古丁、胰岛素、紫色松果菊、锌、维生素C、人参、可乐果、kaua kaua和春黄菊。
优选在口香糖制品的涂层中含有约50μg-500mg的试剂或药物。具体含量取决于活性组分。例如,如果啶甲酸铬为一个实施方式中的活性组分,那么它将以50μg/份(2.8g涂布口香糖)的量存在;阿斯匹林将以325mg/2.8g份存在。药物或试剂在口香糖制品涂层中的含量经选择,以便当咀嚼口香糖时在唾液中产生足够高浓度的药物或试剂。
例如,当试剂为例如尼古丁或咖啡因的兴奋剂时,兴奋剂在口香糖涂层中的含量应是当将口香糖咀嚼2分钟时使唾液含有约15-440ppm的兴奋剂。在此水平,足够量的兴奋剂供应到咀嚼者,从而产生本申请所述的效果。如果将药物作例如药用(例如止痛剂),那么在口香糖涂层中应存在足够的药物,以便当将口香糖咀嚼2分钟之后使唾液含有约1700-约4400ppm。就植物性药材(例如春黄菊、卡法根、可乐果、坚果、人参和松果菊属)而言,试剂应以足够量存在,从而当将该口香糖咀嚼2分钟时使唾液含有约85-1100ppm。就代谢物而言,例如啶甲酸铬和羟基几丁酸,当咀嚼至少2分钟时试剂应以产生约0.5-约900ppm的唾液含量存在。如果试剂为维生素或矿物质(例如磷脂酰基丝氨酸、维生素C和锌),那么试剂应以咀嚼2分钟时使唾液含有约10-约250ppm该维生素或矿物质的量存在。
根据本发明,剂量机理随试剂或药物变化。例如,如果药物为止痛剂,口香糖将以所需基础服用。当然,与口服止痛剂相似,应限制口香糖的片数,例如每隔4小时咀嚼经常不超过1条,每天不超过4-5次。
如果试剂为例如咖啡因的用于增强行为的兴奋剂时,在优选实施方式中应在行为之前10分钟或更少时咀嚼口香糖。正如下面试验中所述的,出人意料地发现,另一5分钟咀嚼含咖啡因的口香糖条达到高水平的机敏。
可以使用各种方法构建口香糖涂层。典型地将涂层以三阶段操作施加于口香糖。在这方面,第一阶段是添加交替施用糖浆的粗涂层并使用粉末。接着的第二阶段是所谓的磨光涂布,其中使用较细的粉末和较长的滚光产生光滑的磨光。最后进行以虫胶处理和抛光的第三阶段,从而提供光泽度高的光滑磨光。在一个优选实施方式中,不使用第二阶段并任选第三阶段。如上所述,与常规涂布口香糖产品相反,本发明的产品含有至少50-75wt%的涂层。仅使用本方法的第一阶段,可以将这样大百分比的涂层以实际的时间-框架涂布到产品上。
在一个实施方式中,涂层含有约10-约30wt%的糖浆和约70-约90wt%的粉末。例如,在一个优选实施方式中,涂层含有20%糖浆和80%粉末。
在构建该涂布口香糖的一个实施方式中,首先将糖浆分散在糖芯上。然后将一部分粉末撒在上面以将糖浆干燥。加入其余量的糖浆并涂布粉末。连续该过程,直到将必需量的糖浆和粉末涂布的口香糖的外部,例如涂布10-20层涂层或更多。在涂层中起着重要作用的掩蔽剂可以包括以下物质的组合:糖、玉米糖浆、或者当为无糖产品时各种糖醇的组合、单体、和聚合物。
已发现,使用这类增加涂层过程,实现了本发明含有药物的产品的优点。无论在粉末中或糖浆中是否含有药物,这都是可行的。因此,如果需要的话,药物可以含在糖浆中,而不是含在粉末中。
根据本发明,涂布产品可以不含虫胶或其它磨光或有光泽层。已发现,涂层可以仅含有无光泽磨光层,其功能不仅令人满意,而且具有一些优点。就此而言,在带有虫胶层的涂层上留有水分的常规涂布产品因涂层中的水分可能降解,因此不会有长的储存期。当具有本发明的厚涂层时这尤为突出。这些厚涂层吸收的水分比较薄的涂层吸收的水分要多。如果利用无光泽磨光层,尽管该厚涂层可以吸收水分,但是该无光泽涂层允许水分加入涂布层和从该涂布层出来。因此这防止了产品降解。因此,本发明提供了具有厚涂层并且储存期增加的产品。
另外该无光泽磨光层不仅允许使用厚涂层,而且允许使用高度吸水的组分。由于采用无光泽磨光层,可以使用高度吸水的药物,但不会使产品降解。
在涂层的一个实施方式中,葡萄糖或蔗糖或其组合作为主要组分。在一优选实施方式中使用葡萄糖,该葡萄糖占涂层的约50-约90%。涂层中的活性组分或药物可以占到高至涂层的30%,直到非常少的量,只要该药效有效。在一优选实施方式中,风味剂为粉末化风味剂并且可以为0.1%-约5%。也可以将高强度甜味剂如阿斯巴甜、三氯半乳蔗糖和乙酰舒泛-K用于该涂层中,并且占涂层的约0.5-约5%。如上所述,这些高强度甜味剂为优良的掩蔽剂。
含有药物或试剂的涂层可以包围各种不同的糖芯组合物。就现在所述的糖芯,根据本发明,该糖芯可以各种已知的不同口香糖为基础。例如,糖芯中的水分可以或低或高,有糖或无糖,含蜡或不含蜡,热值低(经过高基质或低热值填充剂),和/或可以含有牙药。
口香糖通常由水不溶性胶基、水溶性部分和风味剂组成。咀嚼过程中水溶性部分和口香糖中一部分风味剂分散。胶基部分在整个咀嚼过程中都留在嘴中。
不溶性胶基通常含有弹性体、树脂、脂肪和油、软化剂和无机填充剂。胶基可以含有蜡或者可以不含。典型地,胶基占口香糖产品的约20-约40%。然而,由于在本发明中使用如此高含量的涂层,因此糖芯异常地小;否则整个涂布口香糖片消费时太大。如果将常规量的胶基用于小糖芯中,那么将获得一咀嚼用的咀嚼物。因此,在本发明中,基料量比常规高。不溶性胶基可以占口香糖的约30wt%-约90wt%,在一个实施方式中,胶基占口香糖的50%。
在一个实施方式中,本发明的口香糖胶基含有约20wt%-约60wt%的合成弹性体、约0wt%-约30wt%的天然弹性体、约5wt%-约55wt%的弹性体增塑剂、约4wt%-约35wt%填充剂、约5wt%-约35wt%的软化剂和任选少量(约1wt%或更少)的各种组分如色素、抗氧化剂等。
合成弹性体可以包括,但不限于,GPC重均分子量为约10,000-约95,000的聚异丁烯、异丁烯-异戊二烯共聚物(丁基弹性体)、苯乙烯-丁二烯、苯乙烯-丁二烯之比为约1∶3-约3∶1的共聚物、GPC重均分子量为约2,000-约90,000的聚乙酸乙烯酯、聚异戊二烯、聚乙烯、月桂酸乙烯酯含量为共聚物重量的约5%-约50%的乙酸乙烯酯-月桂酸乙烯酯共聚物、及其组合。
优选GPC重均分子量为50,000-80,000的聚异丁烯和苯乙烯-丁二烯以1∶1-1∶3结合的苯乙烯-丁二烯,GBC重均分子量为10,000-65,000并且典型用于泡泡糖胶基的高分子量聚乙酸乙烯酯的聚乙酸乙烯酯,以及月桂酸乙烯酯含量为10-45%的乙酸乙烯酯-月桂酸乙烯酯。
天然弹性体可以包括例如烟熏或液态胶乳的天然橡胶以及例如节路顿胶、香豆胶乳(lechi caspi)、香豆树胶、香豆胶乳(sorva)、钱铁子树胶(massaranduba balata)、钱铁子树胶(massaranduba chocolate)、尼斯佩罗胶(nispero)、山榄胶、糖胶树胶、马来树胶、及其组合。这些优选的合成弹性体和天然弹性体的浓度根据使用该胶基的口香糖是否有粘性或常规、泡泡糖或规则口香糖而变化,下面将讨论。优选的天然弹性体包括节路顿胶、糖胶树胶、香豆胶乳(sorva)和钱铁子树胶(massaranduba balata)。
弹性体增塑剂可以包括,但不限于,天然松香树脂如甘油酯或部分氢化的松香、聚合松香甘油酯、部分二聚松香甘油酯、松香甘油酯、部分氢化的松香季戊四酯、松香甲酯和部分氢化的松香甲酯、松香季戊四酯;合成物例如得自α-蒎烯、β-蒎烯、和/或d-柠檬烯的萜烯树脂;和前述的任意适宜组合。这些优选的弹性体增塑剂也随特定应用和所用弹性体的种类变化。

除了水不溶性胶基部分之外,典型口香糖组合物包括水溶性填充剂部分和一种或多种风味剂。水溶性部分可以包括散装甜味剂、高强度甜味剂、风味剂、软化剂、乳化剂、色素、酸化剂、填充剂、抗氧化剂、以及赋予所需特性的其它组分。
为了使口香糖的咀嚼性和口感最佳化,向口香糖中加入软化剂。这些软化剂,还已知为增塑剂,通常占口香糖重量的约0.5%-约15%。软化剂可以包括甘油、卵磷脂及其组合。也可以将例如含有山梨糖醇、氢化淀粉水解物、玉米糖浆及其组合的甜味剂水溶液作为软化剂和粘合剂用于口香糖中。
散装甜味剂包括有糖和无糖组分。填充甜味剂典型地占口香糖重量的约5%-约95%,更典型地约20%-约80%,更常见是约30%-约60%。有糖甜味剂通常包括口香糖技术领域中公知的含糖组分,包括但不限于,蔗糖、葡萄糖、麦芽糖、糊精、干燥转化糖、果糖、左旋糖、半乳糖、玉米糖浆固体等,单独或组合。无糖甜味剂包括,但不限于,糖醇如山梨糖醇、甘露糖醇、木糖醇、氢化淀粉水解物、麦芽糖醇等,单独或组合。
也可以单独或与上面的组合使用高强度人工甜味剂。优选的甜味剂包括,但不限于,三氯半乳蔗糖、天胺甜精、乙酰舒泛盐、阿斯巴甜、糖精及其盐、环拉酸及其盐、glycerrhizinate、二氢查耳酮、祝马丁、莫尼林等,单独或组合。为了赋予较长持续的甜味和风味感觉,理想地可包封或以其它方式控制至少部分人工甜味剂的释放。可以将这些技术如湿造粒、蜡造粒、喷雾干燥、喷雾冷却、流化床涂布、凝聚和纤维扩展用于实现所需释放特性。
可以将有糖和/或无糖甜味剂的组合用于口香糖中。另外,软化剂也可以赋予另外的甜味例如用糖或糖醇水溶液。
如果希望低热值口香糖,可以使用低热值填充剂。低热值填充剂的例子包括:聚葡萄糖;Raftilose、Raftilin、果糖低聚糖类(NutraFlora);巴糖低聚糖;瓜尔胶水解物(Sun Fiber)或不能消化的糊精(Fibersol)。然而,可以使用其它低热值填充剂。
如果需要的话,也可以使用各种风味剂。风味剂可以口香糖重量的约0.1-约15%使用,优选以0.2-约5%使用。风味剂可以包括香精油、合成风味剂或其混合物包括,但不限于,得自植物和水果的油,例如柑橘油、水果香精、欧薄荷油、绿薄荷油、其它薄荷油、丁香油、冬青油、茴香油等。也可以使用人工风味剂和组分。可以将天然和人工风味剂以任意感觉可接受的方式组合。
可以使用本领域中已知的各种不同方法和机械制备该糖芯。例如,可以使用sigma叶片混合器将制品混合。也可以通过本领域已知的连续加工设备制备该糖芯制品。可以使用常规成片和划线机械形成和划线这些芯,或者可以在一包含落下框的成型机上制备这些芯,并且可以使用氮气冷却制备成球形、椭圆形和其它形状。

与安慰剂口香糖比较,在20个对象中通过单剂量、安慰剂控制、随机化、双向交叉研究评价积极和消极情绪影响的效果。健康对象18-65。评价平均咖啡因消费、烟草药物和酒精状态的筛选调查表。将服药后具有CNS影响的任意对象排除在该研究之外。间隔至少24小时将两种情况下评价约2小时。
剂量:将1x条咖啡因口香糖咀嚼30分钟。该口香糖具有上表咖啡因口香糖中所述的配方。
这些对象被指示他们在试验之前至少8小时内必需食用咖啡因、酒精或其它药物。在试验之前的至少2小时内没有食用烟草产品。对象在开始试验之前的至少8小时但不超过16小时必需是醒的且在活动。对象在以下时间点(开始咀嚼口香糖之前的20、10、5小时和开始咀嚼口香糖之后的2、5、10、15、20、30、40和1小时)需要完成10份调查表。适当分析比较每一项试验,并成组分析积极和消极效果。
以每一时间点的基线数据(-5分钟读数)校正数据。评价所有时间点的积极组和安慰剂组的平均值和标准偏差。
分析结果列于图2。图2图示了机敏与时间的关系。这些结果证实经过5分钟对象报告他们相当机敏。该机敏反应是以参照Panas感觉和情绪等级为基础的。试验2
对6个健康、成年、不吸烟男性对象进行随机、单剂量、双向交叉研究。在每次研究过程中在整夜禁食之后给予单一100mg剂量的咖啡因。试验处理是2个50mg咖啡因口香糖片(条),将其咀嚼15分钟并除去。参照处理是一100mg可咀嚼的No-Doz_片,将其咀嚼并吞咽。每一阶段进行一个处理;给药顺序是按照剂量随机机理。在处理之间有7天冲刷。
在给药前和每一剂量之后15小时内收集血样。通过全验证色谱步骤测定咖啡因的血浆浓度。就这些含量对来自具有可测定给药前含量咖啡因的对象的样品进行校正。由调整过的数据计算药动力学参数并进行统计分析以比较试验和参照处理。临床步骤A.对象选择
参与该研究的6个对象为健康男性,年龄在25-35岁,15%的理想体重如方案中所述。
所有对象都是以医疗史、体检和临床实验室评价上没有任何临床重大发现为基础进行选择的。大于正常范围之外10%的任何实验室值或生命征测量被调查者单独评价。对该研究中注册的那些对象进行测定都没有临床显著性。在开始给药的28天内进行筛选评价。B.药物供应
制品:
试验(A)-两个50mg口香糖条,Amurol ConfectionsCo.(Lot#ALRT7/9/19/96,没有试验日期)
参照(B)-一个100mg No-Doz_咀嚼片,Bristo-Myers产品(Lot#601041,试验日期10/98)。
给药:在整夜禁食之后对象接受试验和参照。对试验为随机的对象首先饮用240ml室温自来水。然后将口香糖片咀嚼15分钟,并放在贴有标签的小瓶中。对参照为随机的对象咀嚼No-Doz_片,然后饮用240ml室温自来水。处理给药的顺序按照随机机理。

在限制期间(第1天),在给药之后4、10和14小时时给予标准化、无咖啡因的膳食和小吃,如第3节中发现的活动日程表和菜单上所述。在每次研究期间食用相同菜单。对象消费至少95%所要求的所有食品和饮料。在每次研究期间在给药之后大约2200小时将对象从临床设备中释放。一7天的冲刷将给药隔开。
限制:在签到研究之前,这些对象被指令在开始给药之前至少14天和整个研究过程中不要服用规定药物。在每一研究期间在给药之前的72小时不允许使用不需处方可以出售的药物。除了所给的药物之外在限制期间不允许服用药物。对象还被指令在给药之前的48个小时和整个每一限制期间远离所有含酒精或咖啡因的产品。对象没有一个报道在所述时间计划内服用任何受限制的物质。
在该研究的限制期间,除了用水(240ml)服药之外,在给药前1小时将对水进行限制,直到给药后1小时。在所有其它时间对水没有限制。在每次给药之后对象保持直立坐着或站着持续2小时,除了研究过程需要之外。在限制期间不允许有兴奋的体力活动。
安全性:在每次签到时进行尿药筛选,检查酒精、大麻和可卡因代谢物。
在每次给药之前测定血压(坐着)、脉率、呼吸速率和口腔温度。调查者考虑所有临床上可接受给药的对象的测量。
在每次给药之后约1小时和每次研究期间释放之前获得血压和脉率测量(坐着),从而监控这些对象的健康状况。如果临床批准的话重复测量。
在研究最后抽样时收集血样用于血细胞比容测定。所有血细胞比容值都在正常范围(41-50%)的10%之内。
相反事件:在任何相反试验的研究中监控这些对象。他们被鼓励报道体征、症状和研究护理的健康的任意变化。在该研究过程中没有一个对象报道任何相反事件。
药动力学样品:在每次期间,在给药之前和给药之后以下述名义时间下收集血样:0.25、0.5、1、1.5、2、2.5、3、4、6、8、10、12和15小时。在收集时用唯一的6位数编号对这些样品做标签。在给药前30分钟内收集给药前样品。所有血浆样品在-18℃至-20℃之间冻藏,直到将其转移到实验室分析,只有1天例外。
对象完成:总共6个对象进入研究,所有对象完成1个研究。分析过程A.质量控制
标准和对照:制备掺加有咖啡因(USP Reference,Lot I)的无干扰、肝素化人血浆池的校准标准。从洲际血库(Memphis,TN)获取该血浆。将标准制备含有0.050、0.100、0.200、0.500、1.00、2.00和5.00μg/ml的咖啡因。
将这些咖啡因标准和对照分成2.5ml等分试样并在实验室中贮藏在冷冻到至少-19℃的聚丙烯雷管中。
研究前操作时的变异系数为0.915%-2.54%。评价步骤的相对准确性,通过比较对照样品的测定浓度平均值和其理论浓度值,发现咖啡因的平均值为99.5%。
运行接受标准:使用电子积分器监控色谱法峰反应和峰反应比。检查可接受停留时间的每次色谱跟踪,将峰形状、图形分辨率和峰反应比之前的积分(分析-内在标准)输入计算机。
通过将最低浓度标准(0.050μg/ml)的平均反应与水空白、基质空白和零标准中观察的任意干扰响应相比较,评价运行量子化的可接受限度。运行量子化限度定义为信噪比为至少2的浓度。
内部标准峰响应与内部标准的平均运行内峰响应(对所有标准和对照计算的)偏离超过±25%的样品、标准和对照为不合格。
每个保持非零标准的响应比作为浓度的函数画图。使用(1/CONC)2作为加权因子通过最小二乘方法计算线性回归(R/S 1,4.3版)。用该校准线确定每个标准样品的计算浓度。与其理论值相差超过±25%的任意标准都将排除在回归之外,并重新计算该回归。由校准线使用等式:(比率-截矩)/斜率评价样品和对照的分析浓度。
如果6个对照中有4个通过评价接受标准并且至少1个对照样品在每个浓度范围内可接受的话,那么该分析运行认为可以接受。如果低对照值与其理论值相差不超过±20%并且高对照值与其理论值相差不超过±15%的话,认为分析运行的对照可以接受。图示对照浓度,从而允许肉眼确定可接受性并确定趋势。B.样品分析

将来自6个报道有咖啡因浓度的对象的所有可获得的数据用于药动力学分析。几个对象具有含可测定浓度的咖啡因的给药前样品。对这些对象每一个的数据而言,通过减去从给药前含量减少预知的时间内的咖啡因含量校正每次抽样时间下测定的浓度。使用与衰减常数相同的时间内观察到的消除速度构建该衰减曲线。调整之后,每个给药前的含量为0.0μg/ml,因此每个给药后的浓度降低。
使用收集样品的实际时间而不是预定时间计算药动力学参数(面积、到达峰值的时间、以及消除速度和半衰期)。单个对象结果的绘图显示也使用样品收集的精确时间。平均结果的绘图显示使用样品收集的预定时间。
在收集的0-15小时期间峰浓度(Cmax)是观察到的最大值(如果需要的话,收集给药前的含量)。到达峰浓度(Tmax)的时间为观察到Cmax的时间(或者如果存在多个峰的话为第一个观察到的)。
以最终log线性浓度-时间值的回归线的斜率的绝对值评价表观一阶消除速度(Ke)。在回归分析中包含的这些值是通过测定浓度的自然对数相对时间的单个对象图确定的。消除半衰期(t-l/2)以0.693/Ke计算。
通过线性梯度法计算至最后非零浓度(Ct)的时间的曲线(AUC)下的面积。通过将量:Ct/Ke相加外推AUC计算至无限时间(AUCinf)的面积。B.统计分析
使用SAS统计程序的综合线性模型(GLM)步骤进行统计分析。在α=0.05下进行治疗效果的假定测定。该统计模型含有后续的主要影响、后续内的对象、周期和治疗。相对后续对象的III型均方项测定后续影响。相对均方误差项测定所有其它主要影响。

咖啡因口香糖片比No-Doz_咀嚼片呈现更快的吸收速度。该口香糖的面积和峰浓度低于No-Doz_的一半,即使该胶基释放的咖啡因为该片的1半。并且到达该口香糖的峰的时间比该片早30分钟。

为了测定口腔的吸收性,进行以下试验。
分析保持机敏的肉桂风味的咖啡因口香糖(Lot 713176)样品的咖啡因,发现每片含有53.44±0.52mg。征募2名对象(S1和S2)咀嚼口香糖。每个对象将1条口香糖咀嚼20分钟,将所有唾液吐在一容器中。咀嚼之后,每个用10ml水(总共20ml)冲洗2次,将冲洗水加入收集的唾液中。用蒸馏水使该溶液的体积到达75ml。S2用一条新的口香糖重复该提取过程。
通过气相色谱法分析口香糖咀嚼物和唾液的咖啡因。(还进行表明从口香糖咀嚼物和唾液中的回收率分别为99.25%和103.50%的掺加研究。当它们实际上与100%没有显著区别时不要校正这些回收的测定咖啡因水平。)试验结果示于表4。

最初的芯糖块重量是0.956克。口香糖被涂敷得到的最终糖块重量是1.46克,得到34.5%的涂层。涂层糖浆1第一次涂敷涂层的60%,使糖块重量为1.26克。使用涂层糖浆2涂布至最终的糖块重量。对5个糖块进行分析,各个糖块产生26.1mg的咖啡因。对于两块的剂量,咖啡因含量是52.2mg。
将该口香糖产品用于咖啡因吸收研究,以比较咖啡因从口香糖和饮料的释放和吸收摄取。当通过血浆咖啡因测定,与饮料中同样含量相比时,该试验结果显示出口香糖是咖啡因较快的释放载体。在通过口香糖释放后,与同样剂量的咖啡因通过咖啡、可乐和茶释放之后相比,试验对象的血浆中咖啡因摄取较快。
在口香糖和三种饮料之间,咖啡因释放的比较,通过统计证明,在下述一种或多种参数方面明显不同:
1.对于口香糖与饮料相比,在释放咖啡因之后的10至30分钟内,血浆咖啡浓度明显较大。这种与较快摄取有关。
2.对于口香糖与一种或多种饮料(2)相比,血浆吸收速率常数(A-速率)较大。对于口香糖与一种或多种饮料(2)相比,血浆吸收半存留期(吸收半存留期)较小。峰值咖啡因血浆的时间。
进行临床试验研究,其中有六个对象参与其中,抽取血液,并分离血浆。事先采集血样,且在预定的时间间隔后通过试验释放载体释放50-55mg的咖啡因。完成5个不同的研究:口香糖(吞咽唾液,G2),口香糖(咳出唾液,G3),咖啡(咽下COF),可乐(咽下COK)和茶(咽下T)。收集5ml的血样,并分离、贮存血浆部分,并提取和分析。对于流体中咖啡因的提取和分析,研究出一种方法,其报告结果作为血浆中咖啡因的浓度。
对来自于六个参与研究的对象的数据进行收集、分析,并用在特定时间间隔测定的平均血浆咖啡因浓度作出曲线。基于平均值进行差异分析(ANOVA)以确定统计显著性。
通过Wagner1967年的Residuals方法使用药动力学软件包确定药动力学参数。通过对线性回归的图的吸收相分析,也确定了吸收速率常数和吸收半存留期,这是因为吸收相遵循着零级动力学。结论如下:
1.与通过咖啡和可乐释放同样剂量咖啡因相比,通过口香糖释放,在服药10至25分钟(T10-T25)的早期时间间隔内,血浆中的咖啡因摄入较快。例如,与咖啡0.186μg/ml和可乐0.236μg/ml相比,在咀嚼口香糖后的血浆咖啡因的平均含量(在T=10分钟时)是0.545μg/ml。换言之,从3种不同的载体释放同样剂量的咖啡因,在T10时,在咀嚼口香糖之后血浆中的咖啡因超过饮用咖啡3倍多,超过饮用可乐2倍多。茶研究的结果变化太大,这是由于仪器问题和对样品反复冷冻/解冻。它们没有包括在计算内。
2.在该研究中,典型的药动力学参数、T-max,A-速率常数、吸收半存留期不能解释在这段感兴趣时间间隔(T10-T25)快速摄取的真相。这部分是由于使用了Residuals方法计算的结果。该方法起源于使用典型药动力学曲线,在数据上该曲线没有太多的波动,其中药浓度(通常每隔1小时测定)增长达到峰T-max,接着降低,没有任何波动。与之相比,数据含较小的波动,这最有可能是以下因素的组合:每隔5分钟而不是每隔一刻钟至1小时测量一次血浆浓度,咖啡因与血浆蛋白结合,发现舌下吸收和肠吸收的组合。该血浆咖啡因浓度遵循典型的药动力学曲线同样的趋势,不同之处在于浓度增长到了一个宽阔T-max,接着降低,而曲线中的一些点上下波动。
A-速率常数和吸收半存留期也通过线性回归测定。在该平均值中没有发现明显不同,然而注意到了一个趋势:对于对象1-4口香糖研究(G2)的A-速率大于咖啡和可乐的,而G2的吸收半存留期研究小于咖啡和可乐的。例如,对于对象1-4,G2的吸收半存留期平均是13±4分钟,对于对象5和6是是28±2分钟,说明对象1-4吸收较快。对于对象1-4舌下的咖啡因吸收量是21±7mg,而对于对象5和6是10±1mg,这可以解释为对象1-4中A-速率增加,而吸收半存留期降低。将对象1-4和对象5和6分别进行ANOVA,显示出统计学上对象1-4的可乐的吸收半存留期大于G2的吸收半存留期(p=0.10),而统计学上G2的A-速率大于可乐和咖啡的A-速率(p=0.05)。
3.通过口香糖的释放,显示出舌下吸收的大量咖啡因直接进入了血流。这证明了经过了唾液被咳出的咖啡因化的口香糖测试。尽管唾液被咳出,却有20-50%的咖啡因通过口腔被吸收。这解释为早期摄入血流中。试验6

最初的芯糖块重量是0.995克。口香糖被涂敷得到的最终糖块重量是1.52克,得到34.5%的涂层。涂层糖浆3第一次涂敷涂层的60%,使糖块重量为1.30克。涂敷涂层糖浆4得到最终的糖块重量。对5个糖块进行分析,单独的糖块产生20.0±0.8mg的咖啡因。对于两块的剂量,咖啡因含量是40.0mg。
将该口香糖产品用于咖啡因吸收研究,以比较咖啡因从口香糖和丸剂的释放和吸收摄取。当通过血浆咖啡因测定,与丸剂中同样含量相比时,该试验结果显示出口香糖是咖啡因较快的释放载体。在通过口香糖释放后,与同样剂量的咖啡因通过丸剂释放之后相比,试验对象的血浆中咖啡因摄取较快。
对来自于六个参与各项研究的对象的数据进行收集、分析,并用在特定时间间隔测定的平均血浆咖啡因浓度作出曲线。基于平均值进行差异分析(ANOVA)和学生t-试验以确定统计显著性。使用药动力学软件包进行药动力学参数测试。测试的口香糖是试验5的颗粒,在涂层中含所有的咖啡因,在咀嚼两颗口香糖(标记为G2、G4或50mg颗粒)后,释放约50mg咖啡因,而在试验6,在涂层和糖芯中含咖啡因,在咀嚼两颗口香糖(指定为G5或40mg颗粒)后,释放约40mg咖啡因。所述的两种口香糖与Pro-PlusTM 50mg丸剂进行比较,该Pro-PlusTM50mg片剂由授权厂商制造,该厂商是PP Products,40 Broadwater Road,Welayn Garden City,Harts,AL7 Bay,UK。以每丸剂48.3mg±1.4mg咖啡因进行咖啡因分析(平均n=5)。
基于如下所示,作出口香糖比丸剂在血流中咖啡因摄取较快的结论:
1.与通过丸剂(50mg)释放同样剂量咖啡因相比,通过口香糖摄取,在服药5至50分钟(T5-T50)之后的早期时间间隔内,血浆中的咖啡因摄入较快。例如,同样剂量的咖啡因通过两种不同的载体释放,基于平均值,在咀嚼口香糖(0.205μg/ml)后,T5的血浆咖啡因的平均含量是丸剂的30多倍。平均血浆咖啡因含量明显大于在T5的a=0.01,和T10的a=0.005的丸剂。
2.通过50mg颗粒口香糖(试验5)释放咖啡因与通过50mg丸剂释放相比,典型的药动力学参数、T-max(峰值血浆咖啡因浓度的时间)和吸收半存留期(吸收度半存留期,咖啡因浓度是峰值1/2的时间)明显不同。与丸剂相比,通过口香糖释放,证明吸收的血浆咖啡因较快,这是由于口香糖的平均血浆吸收半存留期和平均血浆T-Max比丸剂明显小。对于50mg颗粒口香糖,平均吸收半存留期=12.84分钟,而平均T-Max=36.5分钟,而50mg的丸剂,其平均吸收半存留期=24.47分钟(丸剂明显地大于口香糖,a=0.0075),平均T-Max=73.67分钟(丸剂明显地大于口香糖,a=0.0075)。而平均T-Max=73.67分钟(丸剂明显地大于口香糖,a=0.005)。换言之,与口香糖释放同样的咖啡因剂量相比,在咽下丸剂之后,需要花费更长的时间达到1/2的峰值血浆咖啡因浓度和峰值血浆咖啡因浓度。
3.与50mg丸剂相比,50mg颗粒口香糖(试验5)吸收速率常数(速率是咖啡因吸收进血流的速率)明显大,表明以口香糖释放之后的咖啡因吸收速率大于以丸剂释放同样剂量咖啡因的速率。对于50mg的颗粒口香糖,平均吸收速率常数=0.060,而50mg丸剂的平均吸收速率常数=0.031(口香糖明显大于丸剂,a=0.005)。
4.该试验也表明,与通过丸剂释放50mg咖啡因相比,通过试验6的产品-40mg颗粒口香糖释放,在服药10至30分钟(T10-T30)后的早期时间间隔内,血浆咖啡因的摄取较快。显著水平在a=0.05至a=0.20。例如,与丸剂0.034μg/ml相比,在咀嚼40mg颗粒口香糖后的血浆咖啡因的平均含量(在T=10分钟时)是0.228μg/ml(差异略为显著,a=0.2)。换言之,使用两种不同的载体释放咖啡因,在T10时,在咀嚼试验6咖啡因口香糖产品之后,血浆中的咖啡因超过服用丸剂6.7倍多,即使丸剂释放约50mg咖啡因,而试验6产品释放约40mg。在T5时,在咀嚼试验6口香糖之后,血浆中的咖啡因平均超过服用丸剂13倍多。
5.通过试验6的40mg颗粒口香糖产品释放咖啡因与通过50mg丸剂释放相比,典型的药动力学参数、T-max和吸收半存留期明显不同。与丸剂相比,通过试验6口香糖产品释放,证明血浆咖啡因摄取较快,这是由于口香糖的平均血浆吸收半存留期和平均血浆T-Max比丸剂明显要小。对于50mg试验5的口香糖,平均吸收半存留期=18.33分钟,而平均T-Max=45分钟,而50mg的丸剂,其平均吸收半存留期=24.47分钟(丸剂明显地大于口香糖,a=0.05),而平均T-Max=73.67分钟(丸剂明显地大于口香糖,a=0.15)。即使试验6产品释放40mg咖啡因与通过丸剂释放50mg相比,丸剂比口香糖需要花费更长的时间达到1/2的峰值血浆咖啡因浓度和峰值血浆咖啡因浓度。
6.基于如下所述,可以判定所有咖啡因都配制在在颗粒涂层中的口香糖其释放到血浆中咖啡因的速度,比咖啡因掺加在涂层和糖芯中配制的口香糖的更快:
对于50mg颗粒口香糖和试验5的口香糖,典型的药动力学参数T-max和吸收半存留期比丸剂大,尽管对于50mg颗粒口香糖(试验5)(分别是a=0.0075,a=0.005)显著差异水平大于试验6的产品(a=0.05,a=0.15)。对于50mg颗粒口香糖或试验6的产品,丸剂吸收速率常数明显低。而且,对于50mg颗粒口香糖(试验5)a=0.005,显著差异水平大于大于试验6产品的0.20。
7.综合两个完整的咖啡因研究的结论,显示出测试的通过多种释放载体的血浆中咖啡因摄取速率遵循以下模式:
所有咖啡因在涂层中的颗粒口香糖>咖啡因在涂层和糖芯中的颗粒口香糖=饮料咖啡/可乐>丸剂
选择咖啡因作为药物释放试验的模型,这是因为它被核准为食用级的、药理学活性剂,它容易在血浆中以宽范围药剂量被发现。通过许多释放载体,包括液体(咖啡、可乐和丸剂),被广泛食用。通过不同的摄取载体服药,即液体糖浆和丸剂这样两种口服释放载体。咖啡因化的饮料和丸剂与咖啡因化的口香糖相比进行试验,将揭示出以液体或涂敷的丸剂与涂敷的口香糖服用同样药物,会发生怎样的变化。
应理解的是,本领域技术人员对本文所述的本优选实施方式的各种改变和改进是显而易见的。这些改变和改进可以在不背离本发明精神和范围并在不消除其意料优点的情况下进行。因此所附加的权利要求书包括这些改变和改进。
Claims (54)
1.一种向个体释放药物的方法,包括步骤:
提供含有糖芯和涂层的口香糖,该涂层基本上围绕糖芯,该涂层占口香糖重量的至少50%,并且该涂层含有药物;
咀嚼该口香糖,使药物从口香糖组合物中释放到咀嚼者的口腔中;和
连续咀嚼该口香糖,由此产生使药物通过个体的口腔粘膜进入个体的体循环的液压。
2.如权利要求1的方法,其中该涂层含有高强度甜味剂。
3.如权利要求1的方法,其中该高强度甜味剂选自:阿斯巴甜、三氯半乳蔗糖、糖精和乙酰舒泛-K。
4.如权利要求1的方法,其中该涂层是通过在糖芯上交替形成粉末和糖浆层生产的。
5.如权利要求1的方法,其中糖芯含有至少50wt%的水不溶性胶基。
6.如权利要求1的方法,该药物选自:止痛剂、肌肉松弛剂、抗生素、抗病毒剂、抗组胺剂、减充血剂、抗炎剂、制酸剂、精神治疗剂、胰岛素、维生素、矿物质和心血管药。
7.如权利要求1的方法,其中该涂层为无光泽磨光层。
8.如权利要求1的方法,其中该涂层不含虫胶层。
9.一种口香糖,含有:
糖芯,它包括水溶性部分和不溶于水的部分;和
含有药物的涂层,它包围了糖芯,该涂层占口香糖产品重量的至少50%。
10.如权利要求9的口香糖,其中该药物选自:止痛剂、肌肉松弛剂、抗生素、抗病毒剂、兴奋剂、抗组胺剂、减充血剂、抗炎剂、制酸剂、精神治疗剂、胰岛素、维生素、矿物质和心血管药。
11.如权利要求9的口香糖,其中该涂层包括足够量的味道掩蔽剂,从而赋予可接受的口感性能。
12.如权利要求11的口香糖,其中该味道掩蔽剂选自:葡萄糖酸锌、乙基麦芽酚、甘氨酸、乙酰舒泛-K、阿斯巴甜、糖精、果糖、木糖醇、异麦芽糖、麦芽糖醇、喷雾干燥的甘草、甜根、葡萄糖酸钠、葡糖酸δ-内酯、乙基香兰素、葡萄糖、三氯半乳蔗糖、香兰素和乙基麦芽酚。
13.如权利要求11的口香糖,其中该味道掩蔽剂占涂层重量的约30%-约99%。

涂层,它至少基本上包围了糖芯并含有药物和高强度甜味剂,该涂层占产品重量的至少50%。

提供含有糖芯和基本上围绕糖芯的涂层的口香糖,该涂层占口香糖重量的至少50%,该涂层含有典型地通过个体吞咽达到特定效果的试剂,该口香糖含有低于通过个体吞咽达到效果的典型量的试剂;
将该口香糖咀嚼,由此使试剂释放到个体唾液中;和
连续咀嚼口香糖,迫使试剂通过个体口腔中的粘膜。
28.如权利要求27的方法,其中该试剂为药物。
29.如权利要求27的方法,其中该药物选自:止痛剂、肌肉松弛剂、抗生素、抗病毒剂、抗组胺剂、减充血剂、抗炎剂、制酸剂、精神治疗剂和心血管药。

32.一种增强个体行为的方法,包括步骤:
