发明背景
已经鉴定了与血液凝固过程有关的多种因子,包括因子VII,一种血浆糖蛋白。血管壁损伤后暴露于循环血的组织因子(TF)与循环中存在的FVIIa之间复合物的形成启动止血(haemostasis),FVIIa的含量相当于总FVII蛋白量的约1%。FVII在血浆中主要以单链酶原存在,其被FXa切割成两条链的活化形式——FVIIa。已经研制了重组活化因子VIIa(rFVIIa)作为一种止血(pro-haemostatic)剂原。

作为一种蛋白质,修饰的因子VII分子易于被物理降解,包括变性和聚集,如形成可溶的或不可溶的二聚体、寡聚体和多聚体形式的聚合体,或者易于被化学降解,包括例如水解、脱酰胺和氧化。总的后果包括:修饰的因子VII分子的活性丧失,形成毒性和免疫原性的降解产物,在注射降解的修饰因子VII分子后造成血栓形成的严重危险,注射用针头的堵塞,和非均一性的危险。因此,含有修饰的因子VII的药物的安全性和有效性与修饰的因子VII的稳定性直接有关。
因此,含有修饰的因子VII分子的组合物需要稳定。具体而言,需要贮存和处理含有修饰的因子VII的药物,而不需要冰箱,其中组合物在使用前可以长期贮存,如至少6个月。
希望的是制备修饰的因子VIIa的施用形式,适于贮存和递送。理想地,药物产品作为一种液体贮存和施用。此外,药物产品也可以冻干,即冷冻干燥,然后在患者使用之前才添加适当的稀释剂重建。理想地,药物产品具有足以长期贮存(即6个月以上)的稳定性。
决定制成的药物产品是以液体形式保存还是冻干保存通常以这些形式的蛋白质药物的稳定性为基础。蛋白质稳定性能够受如离子强度、pH、温度、重复冻融循环和暴露于剪切力等因素影响。由于物理不稳定性,包括变性和聚集(可溶的和不可溶的聚合体形成),以及化学不稳定性,包括如水解、脱酰胺和氧化等,活性蛋白质可能丢失。关于蛋白质药物稳定性的综述,参见,例如:Manning等人,PharmaceuticalResearch 6:903-918(1989)。
尽管可能发生蛋白质的不稳定性已经普遍知道,但是预测一种具体蛋白质的具体的不稳定性问题是不可能的。所有这些不稳定性都能导致蛋白质副产物或衍生物的形成,它们具有降低的活性、提高的毒性和/或提高的免疫原性。实际上,蛋白质沉淀可以导致血栓形成,剂型和剂量的不均一性,以及注射器堵塞。此外,翻译后修饰,如N端某些谷氨酸残基的γ羧化,和碳水化合物侧链的加入,也提供了在贮存后容易修饰的潜在位点。因此,一种蛋白质的所有药物组合物的安全性和有效性与其稳定性直接有关。以液体剂型保持稳定性一般不同于冻干剂型,因为分子运动的可能性大大提高,因此分子相互作用的可能性提高。以浓缩形式保持稳定性也是不同的,因为在升高的蛋白质浓度时倾向于形成聚合体。
研制一种液体组合物要考虑许多因素。短期(即不到6个月)液体稳定性通常依赖于避免总体结构改变,如变性和聚集。这些方法在关于大量蛋白质的文献中有描述,并且存在稳定剂的许多例子。众所周知,可有效稳定一种蛋白质的试剂实际上可以使另一种不稳定。一旦蛋白质对于总体结构改变稳定,研制一种具有长期稳定性(例如6个月以上)的液体组合物依赖于针对该蛋白质特异的降解类型进一步稳定蛋白质。可能包括更多的具体降解类型,例如二硫键不规则、某些残基氧化、脱酰胺、环化。尽管不总是能够确定个别的降解种,但是发展了测定法监测细微的改变,以监测具体赋形剂独特地稳定目的蛋白质的能力。
除了稳定性考虑之外,人们通常选择全世界多家医药管理机构批准的赋形剂。组合物接近等渗、组合物在注射/输液后pH处于生理适当范围内是希望的,可能导致患者的其它疼痛和不适。

修饰的因子VII现在能够在液体制剂中提供,需要在-80℃下冻存。
因此,本领域需要提高修饰的因子VII多肽的稳定性和提供适于在2-8℃下长期贮存6个月以上的液体组合物的方法。因此,本发明的一个目的是提供修饰的因子VII多肽的含水组合物,它将可接受地控制降解产物。
发明概述
我们发现,修饰的因子VII或其类似物(“修饰的因子VII多肽”),当与适于将pH保持在约4.0-约8.0的试剂、抗氧化剂和钙盐一起在水溶液中配制时,在物理和化学上是稳定的。
一方面,本发明提供一种液体含水组合物,其含有:(i)一种修饰的因子VII多肽;(ii)一种试剂,其适于将pH保持在约4.0-约8.0范围;(iii)一种抗氧化剂;和(iv)一种试剂,其选自:钙盐、镁盐或其混合物。
在不同实施方案中,pH保持在约4.0-约7.0,如约4.5-约7.0,约5.0-约7.0,约5.5-约7.0,或约6.0-约7.0。

在不同的实施方案中,组织因子介导的反应与选自下列的疾病有关:全身性炎症反应综合征(SIRS),急性呼吸系统疾病综合征(ARDS),多器官功能衰竭(MOF),HUS和TTP。
发明详述
根据本发明的组合物可用作稳定的、优选地即用的修饰的因子VII多肽的组合物。当贮存于2-8℃时,该组合物至少稳定6个月,优选地可达36个月。当在2-8℃至少贮存6个月时,该组合物是化学和/或物理稳定的,特别是化学稳定的。
“稳定的”是指组合物在2-8℃贮存6个月后,至少保留其最初生物活性的50%,其测定是使用基本如WO 92/15686(实施例III)所述的竞争性凝血测定法,或如本说明书中所述的测定法5-8中的一种或多种(参见下文“测定法”部分)。优选地,稳定的组合物在2-8℃贮存6个月后,至少保留其最初活性的80%。
术语“物理稳定的”是指一种组合物,其目视保持澄清。组合物的物理稳定性的评价方法是,该组合物在不同温度下贮存不同时间后,通过目视检查和根据浊度评价。组合物的目视检查在暗背景和锐聚焦光线下进行。一种组合物当目视可见混浊时,被分类为物理不稳定的。

在此使用时,指定的量理解为±约10%,例如,约50mM包括50mM±5mM;例如,4%包括4%±0.4%,等等。
当指溶解于溶液中的固体和混合到溶液中的液体时,百分数是(重量/重量)。例如,对于Tween,是100%原液的重量/溶液的重量。
术语“等渗的”含意是“与血清等渗”,即,约300±50毫渗透分子/千克。张力是溶液在施用前的重量摩尔渗透压浓度的量度。

使用方法:
根据本发明的制剂,其含有修饰的因子VII多肽,生物活性大大低于野生型因子VII,可以作为抗凝剂,例如对接受血管成形术或其它手术操作的患者使用,这些操作可以增加血栓形成或血管阻塞的危险,如再狭窄中所发生的。抗凝剂适用的其它医学适应证包括但不限于:深静脉血栓形成,肺栓塞,中风,弥散性血管内凝血(DIC),与革兰氏阴性内毒素血症有关的组织(例如肺和/或肾)中的血纤蛋白沉积,心肌梗死;急性呼吸窘迫综合征(ARDS),全身性炎症反应综合征(SIRS),溶血性尿毒性综合征(HUS),MOF和TTP。
根据本发明配制的因子VII多肽:

当使用如测定法1-4(参见下文“测定法”部分)所述的一种或多种凝血测定、蛋白水解测定或TF结合测定法检测时,生物活性比野生型因子VIIa大大降低的修饰的因子VII多肽,包括变体,显示低于同一细胞型产生的野生型因子VIIa的比活性的约25%,优选地低于约10%,更优选地低于约5%,最优选地低于约1%。

在优选实施方案中,可以在因子VII催化三联体的氨基酸序列内进行氨基酸置换,在此被定义为含有有助于因子VIIa催化部位的氨基酸的区域。催化三联体中的置换、插入或缺失通常位于或邻近构成催化部位的氨基酸。在人和牛因子VII蛋白中,构成催化“三联体”的氨基酸是Ser344、Asp242和His193(数字编号表示在人野生型因子VII中的位点)。来自其它哺乳动物种的因子VII的催化部位可以用目前可用的技术确定,包括蛋白质分离和氨基酸序列分析。催化部位也可以如下确定:将一种序列与其它丝氨酸蛋白酶(特别是胰凝乳蛋白酶)的序列进行比对,后者的活性部位以前已经确定(Sigler等人,J.Mol.Biol.35:143-164(1968),在此引用作为参考),从而根据这种比对确定类似的活性部位残基。
为了防止或以其它方式抑制因子VIIa对因子X和/或IX的激活,进行氨基酸置换、插入或缺失。但是,这样修饰的因子VII应当也保留与未修饰的(authentic)因子VII和/或因子VIIa在血凝级联中竞争结合组织因子的能力。例如,利用如此处所述的凝血测定法,或者使用例如含有细胞表面组织因子的细胞系(如人膀胱癌细胞系J82)的竞争结合测定法(Sakai等人,J.Biol.Chem.264:9980-9988(1989)),可以容易地测定这种竞争。
构成因子VII的催化部位的氨基酸,如人和牛因子VII中的Ser344、Asp242和His193,可以被置换或缺失。优选地只改变一个氨基酸,从而使提高分子抗原性或抑制结合组织因子的能力的可能性最小化,然而,可以进行两个或多个氨基酸的改变(置换、添加或缺失),也可以进行置换、添加和缺失的组合。在人和牛因子VII的一个优选实施方案中,Ser344优选地被置换为Ala,但是Gly、Met、Thr或其它氨基酸也能够被置换。优选地用Glu置换Asp,用Lys或Arg置换His。通常选择尽可能小地破坏蛋白质三级结构的置换。人们可以向人、牛或其它种的适当因子VII序列的催化部位中引入如上所述的残基改变,如此处所述检测获得的蛋白质的希望的催化活性抑制水平和产生的抗凝活性。
在人和牛因子VII的优选实施方案中,活性部位残基Ser344被修饰,被置换为Gly、Met、Thr,或者更优选地Ala。这种置换能够分开进行,或者与催化三联体的其它位点(包括His193和Asp242)处的置换组合进行。
因子VII多肽的生物活性:
因子VIIa在血液凝固中的生物活性来源于其(i)结合组织因子(TF)和(ii)催化因子IX或因子X的蛋白水解切割,产生活化的因子IX或X(分别为IXa或Xa)的能力。
对于本发明,因子VII多肽的生物活性(“因子VII生物活性”)可以如下定量:如美国专利号5,997,864或WO 92/15686所述,使用缺乏因子VII的血浆和促凝血酶原激酶,测定一种制品促进血液凝固的能力。在该测定中,生物活性表示为凝血时间相对于对照样品的减少,通过与含有1U/ml因子VII活性的合并人血清标准对比,将其转化为“因子VII单位”。此外,因子VIIa生物活性也可以如下定量:
——在一个含有包埋于脂膜中的TF和因子X的系统中,测定因子VIIa或因子VIIa相当物产生活化的因子X(因子Xa)的能力。(Persson等人,J.Biol.Chem.272:19919-19924,1997);
——在一个水系统中测定因子X的水解(“体外蛋白水解测定法”,见下文);
——使用一种以表面胞质团共振为基础的仪器,测定因子VIIa或因子VIIa相当物与TF的物理结合(Persson,FEBS Letts.413:359-363,1997);和
——测定因子VIIa和/或因子VIIa相当物对一种合成底物的水解(“体外水解测定法”,见下文);和
——在一个不依赖于TF的体外系统中测定凝血酶的产生。
适于测定因子VII多肽的生物活性的测定法:
根据本发明有用的因子VII多肽可以通过适当的测定法选择,这些测定能够象简单的体外预试验那样进行。因此,本说明书公开了一种测定因子VII多肽活性的简单试验(称为“体外水解测定法”)。
体外水解测定法(测定法1)
可以测定天然(野生型)因子VIIa和因子VII多肽(下文均称为“因子VIIa”)的比活性。它们也可以平行测定,以直接比较它们的比活性。该测定在微孔板(MaxiSorp,Nunc,Denmark)上进行。在含有0.1MNaCl,5mM CaCl2和1mg/ml牛血清白蛋白的50mM Hepes,pH7.4中,向因子VIIa(终浓度为100nM)中加入终浓度为1mM的显色底物D-Ile-Pro-Arg-对硝基苯胺(S-2288,Chromogenix,Sweden)。用SpectraMaxTM 340读板仪(Molecular Devices,USA)连续测量405nm的吸光度。在20分钟温育过程中产生的吸光度,在减去不含酶的空白孔的吸光度后,用来计算因子VII多肽与野生型因子VIIa的活性之比:
比=(A405nm因子VII多肽)/(A405nm因子VIIa野生型)。
据此可以鉴定活性低于、相当于或高于天然因子VIIa的因子VII多肽,如因子VII多肽活性与天然因子VII(野生型FVII)活性之比高于约1.0的因子VII多肽。
因子VII多肽的活性也可以用一种生理底物测定,如浓度适当地为100-1000nM的因子X(“体外蛋白水解测定法”),其中在加入适当的显色底物(例如S-2765)后测定产生的因子Xa。另外,活性测定可以在生理温度下进行。
体外蛋白水解测定法(测定法2)
天然(野生型)因子VIIa和因子VII多肽(下文均称为“因子VIIa”)平行测定,以直接比较它们的比活性。该测定在微孔板(MaxiSorp,Nunc,Denmark)上进行。因子VIIa(10nM)和因子X(0.8μM)在含有0.1M NaCl,5mM CaCl2和1mg/ml牛血清白蛋白的100μL 50mMHepes,pH7.4中温育15分钟。然后加入含有0.1M NaCl,20mMEDTA和1mg/ml牛血清白蛋白的50μL 50mM Hepes,pH7.4,终止因子X的切割。加入终浓度为0.5mM的显色底物Z-D-Arg-Gly-Arg-对硝基苯胺(S-2765,Chromogenix,Sweden),测定产生的因子Xa的量。用SpectraMaxTM 340读板仪(Molecular Devices,USA)连续测量405nm的吸光度。在10分钟期间产生的吸光度,在减去不含FVIIa的空白孔的吸光度后,用来计算因子VII多肽与野生型因子VIIa的蛋白水解活性之比:
比=(A405nm因子VII多肽)/(A405nm因子VIIa野生型)。
据此可以鉴定活性低于、相当于或高于天然因子VIIa的因子VII多肽,如因子VII多肽活性与天然因子VII(野生型FVII)活性之比大约高于1.0的因子VII多肽。
因子VIIa或因子VII多肽产生凝血酶的能力也能够用一种测定法(测定法3)测定,其中包括生理浓度的所有有关的凝血因子和抑制剂(当模拟血友病A条件时,除去因子VIII)和活化的血小板(如Monroe等人,(1997)Brit.J.Haematol.99,542-547,543页所述,在此引用作为参考)。
因子VII多肽的活性也可以用基本如WO 92/15686或US5,997,864所述的一步凝血测定法(测定法4)测定。简言之,待测样品用50mM Tris(pH7.5)稀释、0.1%BSA稀释,100μl稀释样品与100μl缺乏因子VII的血浆和200μl含有10mM Ca2+的促凝血酶原激酶C温育。测定凝血时间,与使用参考标准或一组连续稀释的柠檬酸化正常人血浆获得的标准曲线进行对比。
竞争测定法:FVIIa/sTF酰胺分解活性的抑制(测定法5)
修饰的因子VII对FVIIa-TF催化的酰胺分解活性的抑制使用可溶性TF(10nM)、重组人FVIIa(10nM)和浓度提高的修饰的因子VII检测。不同浓度的修饰的因子VII与10nM sTF和10nM FVIIa在BSA缓冲液(50mM Hepes,pH7.4,100mM NaCl,5mM CaCl2和1mg/ml BSA)中室温预温育60分钟,然后加入底物S2288(1.2mM,Chromogenix)。在405nm下连续测量显色30分钟。酰胺分解活性表示为mOD/min。可以计算修饰的因子VII抑制FVIIa/TF酰胺分解活性的IC50值。
FXa产生的抑制(测定法6)
脂化TF(10pM)、FVIIa(100pM)和修饰的因子VII(0-50nM)在BSA缓冲液(参见测定法4)中室温温育60分钟,然后加入FX(50nM)。加入1/2体积的终止缓冲液(50mM Hepes,pH7.4,100mM NaCl,20mM EDTA)10分钟后终止反应。加入底物S2765(0.6mM,Chromogenix),并连续测量405nm的吸光度10分钟,测定产生的FXa的量。可以计算修饰的因子VII抑制FVIIa/脂化TF介导的FX激活的IC50值。
依赖TF的凝血测定法(测定法7):
该测定在ACL300 Research凝血装置(ILS Laboratories)上进行。修饰的因子VII用50mM咪唑,pH7.4,100mM NaCl,0.1%BSA稀释,与25mM CaCl2以2∶5比例混合,加至该凝血装置的样品杯中。来自人、大鼠、兔、狒狒或猪的促凝血酶原激酶用咪唑缓冲液稀释,使得不含修饰的因子VII的样品达到约30秒的凝血时间,将该稀释液置于试剂容器2中,人、大鼠、兔、狒狒或猪血浆置于试剂容器3中。在分析过程中,将70μl修饰的因子VII和CaCl2混合物转移到25μl促凝血酶原激酶试剂中,预温育900秒,之后加入60μl血浆,测定凝血时间。最大凝血时间设为400秒。用稀释的促凝血酶原激酶作标准曲线,将凝血时间转化为相对于未加修饰FVII的对照的TF活性。
修饰的因子VII对FVIIa/细胞表面TF催化的FX激活的抑制(测定法8):
表达人TF的细胞单层,例如人肺成纤维细胞WI-38(ATTC No.CCL-75)、人膀胱癌细胞系J82(ATTC No.HTB-1)、人角质细胞系CCD 1102KerTr(ATCC no.CRL-2310)、人胶质母细胞瘤细胞系U87或人乳腺癌细胞系MDA-MB231,用作FVIIa/TF催化的FX激活中的TF来源。在24、48或96孔板中铺满的细胞单层用缓冲液A(10mMHepes,pH7.45,150mM NaCl,4mM KCl,和11mM葡萄糖)洗涤一次,用缓冲液B(添加1mg/ml BSA和5mM Ca2+的缓冲液A)洗涤一次。向细胞中同时添加溶于缓冲液B的FVIIa(1nM)、FX(135nM)和不同浓度的修饰的因子VII。此外,在添加rFVIIa和FX之前,细胞也与修饰的因子VII预温育15分钟。FXa的形成在37℃下进行15分钟。从每孔中取出50μl等份,加入50μl终止缓冲液(添加10mMEDTA和1mg/ml BSA的缓冲液A)。通过将50μl上述混合物转移到微孔板的孔中,并向孔中加入25μl Chromozym X(终浓度为0.6mM),测定产生的FXa的量。连续测量405nm的吸光度,利用FXa标准曲线将最初的显色速度转化为FXa浓度。
修饰的因子VII多肽的制备和纯化:
适于根据本发明配制的修饰的因子VII分子及其生产已经在WO92/15686、WO 94/27631、WO 96/12800和WO 97/47651中描述。
通常,纯化的人因子VIIa优选地通过DNA重组技术制备,如Hagen等人,Proc.Natl.Acad.Sci.USA 83:2412-2416,1986所述,或如欧洲专利号200.421(ZymoGenetics,Inc.)所述。
因子VII也可以利用Broze和Majerus,J.Biol.Chem.255(4):1242-1247,1980,和Hedner和Kisiel,J.Clin.invest.71:1836-1841,1983所述的方法生产。这些方法产生的因子VII不含可检测的量的其它凝血因子。以另外一步凝胶过滤作为最终的纯化步骤,可以获得进一步纯化的因子VII制剂。然后利用已知的方法,例如使用几种不同的血浆蛋白,如因子Xlla、IXa或Xa,将因子VII转化为活化的因子VIIa。此外,如Bjoern等人(Research Disclosure,269September 1986,pp.564-565)所述,因子VII的活化方法也可以是通过离子交换层析柱,如MonoQ(Pharmacia fine Chemicals)等,或者在溶液中自活化。
因子VII多肽,无论它是分离的还是重组产生的,然后可以如下所述化学修饰,例如:WO 92/15686,WO 94/27631,WO 96/12800和WO 97/47651,或Sorensen等人.J.Biol.Chem.272:11863-11868,1997(FFR-rFVIIa:活性部位用D-Phe-L-Phe-L-Arg-氯甲基酮封闭的FVIIa)。
因子VII变体可以通过野生型因子VII的修饰或者利用重组技术产生。与野生型因子VII相比氨基酸序列改变的因子VII相当物可以如下产生:使用公知的方法,例如位点特异的诱变,改变编码天然因子VII的核酸中的氨基酸密码子或除去某些氨基酸密码子,修饰编码野生型因子VII的核酸序列(参见,例如:Cunningham和Wells,1989,Science 244:1081-1085)。
从来源细胞中分离多肽可以用本领域公知的任何一种方法实现,包括但不限于:从贴壁细胞培养物中取出含有希望的产物的细胞培养基;离心或过滤,除去非贴壁细胞;等等。
任选地,修饰的因子V多肽可以进一步纯化。纯化可以用本领域公知的任何一种方法完成,包括但不限于:亲和层析,例如使用抗因子VII抗体柱(参见,例如Wakabayashi等人,J.Biol.Chem.261:11097,1986;和Thim等人,Biochem.27:7785,1988);疏水相互作用层析;离子交换层析;大小排阻层析;电泳方法(例如制备等电聚焦(IEF),差示溶解度(例如,硫酸铵沉淀),或提取,等等。通常参见:Scopes,《蛋白质纯化》(Protein Purification),Springer-Verlag,New York,1982;和《蛋白质纯化》,J.C.Janson和Lars Ryden编著,VCH Publishers,New York,1989。纯化后,制剂中所含的来自宿主细胞的非因子VII多肽优选地不到约10%重量,更优选地不到约5%,最优选地不到约1%。

实施例1
A.测定方法
聚合体的含量通过非变性大小排阻HPLC测定。氧化形式的含量通过RP-HPLC测定。酶降解形式的含量通过RP-HPLC测定。
非变性大小排阻层析在7.5×300mm的Waters Protein Pak 300SW柱上进行,使用0.2M硫酸铵,5% 2-丙醇pH7.0作为流动相。流速:0.5ml/min。检测:215nm。上样:25μg FVIIa。
反相HPLC在颗粒大小为5μm、孔径为300的专利4.5×250mm丁基键合硅石(butylbonded silica)柱上进行。柱温度:70℃。A-缓冲液:0.1%v/v三氟乙酸。B-缓冲液:0.09%v/v三氟乙酸,80%v/v乙腈。该柱在30分钟内用从X到(X+13)%B的线性梯度洗脱。调节X,使得FVIIa以大约26分钟的保留时间洗脱。流速:1.0ml/min。检测:214nm。上样:25μg FVIIa。
实施例2
含有甲硫氨酸作为抗氧化剂的Phe-Phe-Arg氯甲基酮-灭活的因子VII(FFR-rFVIIa)水制剂的化学稳定性
制备两种不同的制剂。制剂的组成如下:
FFR-rFVIIa 2mg/ml
NaCl 2.8-2.9mg/ml
CaCl2,2H2O 1.4-1.5mg/ml
甘氨酰甘氨酸 1.3mg/ml
甲硫氨酸 0或1mg/ml
pH 7.0
该制剂由含有FFR-rFVIIa、NaCl、CaCl2和甘氨酰甘氨酸的液体FFR-rFVIIa本体(bulk)溶液制成。甲硫氨酸溶解于水中。混合FFR-rFVIIa本体溶液和甲硫氨酸溶液,然后调节溶液的pH为7.0。过滤该制剂(0.2μm),装到小瓶中(每个小瓶2.2ml溶液)。这些小瓶贮存于35℃。取出样品,在下表所述的时间点(通过RP-HPLC)分析氧化形式的含量。该表显示氧化形式的含量(%)。

Claims (31)
1.一种液体、含水组合物,其含有:
(i)一种修饰的因子VII多肽;
(ii)一种适于将pH保持在约4.0-约8.0范围内的试剂;
(iii)一种抗氧化剂;和
(iv)一种选自:钙盐、镁盐或其混合物的试剂。
2.根据权利要求1的组合物,其中pH保持在约4.0-约7.0,如约4.5-约7.0,约5.0-约7.0,约5.5-约7.0,或约6.0-约7.0的范围内。
3.根据权利要求1或权利要求2的组合物,其中抗氧化剂(iii)选自:L-或D-甲硫氨酸、甲硫氨酸类似物、含甲硫氨酸的肽、甲硫氨酸同系物、抗坏血酸、半胱氨酸、高半胱氨酸、谷胱甘肽、胱氨酸和胱硫醚。
4.根据权利要求3的组合物,其中所述抗氧化剂是L-甲硫氨酸。
