Nach Ablauf der Zeit wird die SPME-Nadel vorsichtig durch das Septum gestochen und der aktivierte Fiber für genau 30 Minuten in den Gasraum der Flasche ausgefahren (Exposition). Nach Ablauf dieser Zeit wird der Fiber zurückgezogen und die SPME-Nadel aus der Flasche gezogen. Nun wird die SPME-Nadel durch das Septum des Injektors gestochen, der Fiber in den Splitliner des Gaschromatographen (GC) (3 mm ID, ohne Verjüngung) ausgefahren und das GC-Chromatogramm gestartet (Desorption). Nach ca. 3 Minuten kann der Fiber zurückgezogen und die Nadel aus dem Injektor gezogen werden. Der Fiber kann sofort für die nächste Messung eingesetzt werden.
