Die Kultivierung von Mikroorganismen im industriellen Maßstab hat in den letzten Jahren vielseitige Anwendungen gefunden. So werden Mikroorganismen gezüchtet um Biomasse zur Stromgewinnung oder zur Produktion von Biodiesel herzustellen. Im Zuge der Bemühungen den weltweiten Kohlendioxid(CO2)-Ausstoß zu reduzieren, werden zur Züchtung auch Abgase verwendet, aus denen photosynthetisch aktive Mikroorganismen CO2 entnehmen, das sie in Kohlenwasserstoffe umwandeln. In Bioreaktoren kann das Wachstum der Mikroorganismen hingegen genau überwacht werden. Die Mikroorganismen werden in einer geeigneten Nährlösung kultiviert, die Wasser, eine Kohlenstoffquelle sowie gegebenenfalls eine Energiequelle und Ergänzungsstoffe wie Mineralien oder Spurenelementen enthält. Die Zusammensetzung richtet sich dabei nach den Anforderungen der Mikroorganismen. Photosynthetisch aktive Algen oder Cyanobakterien brauchen neben Wasser häufig lediglich CO2 als Kohlenstoff- und Sonnenlicht als Energiequelle. Bei der Kultivierung von Mikroorganismen werden vier Wachstumsphasen unterschieden, Anlaufphase, exponentielle Phase, stationäre Phase und Absterbephase. Bei der industriellen Kultivierung von Mikroorganismen sollte sich die Kultur stets in der exponentiellen Wachstumsphase befinden, weil die Zellteilungsrate darin am höchsten ist. Surface receptors to which magnetizable particles bind allow a particularly specific loading of the microorganisms. Polysaccharides, in particular mixtures of different polysaccharides, on the other hand, form highly branched molecular networks which enable a particularly efficient loading of the microorganisms.
In einer bevorzugten Ausführungsform wird die Bildung des Oberflächenmoleküls durch eine Veränderung mindestens einer Kultivierungsbedingung induziert. Dadurch sind keine Zugaben von Chemikalien notwendig, welche die Mikroorganismen gegebenenfalls schädigen könnten oder nach dem Abtrennen der Mikroorganismen wieder entfernt werden müssten. In einer besonders bevorzugten Ausführungsform ist die Kultivierungsbedingung ausgewählt aus der Gruppe bestehend aus Lichtexposition, Lichtwellenlänge, Temperatur, pH-Wert, Salzkonzentration und Zusammensetzung des Mediums, da diese Kultivierungsbedingungen ohne großen Aufwand verändert werden können. Die Temperatur des Mediums und die Lichtexposition kann beispielsweise durch Änderung der Beschattung des Photobioreaktors verringert oder erhöht werden. Alternativ kann die Bildung des Oberflächenmoleküls auch durch den natürlichen Nachtzyklus induziert werden. Die Wellenlänge des einfallenden Lichts kann verändert werden, indem das auf dem Bioreaktor eintreffende Sonnenlicht durch einen optischen Filter geleitet wird. Der pH-Wert des Mediums kann über die Menge an CO2, welches den Mikroorganismen als Kohlenstoffquelle zugeführt wird, reguliert werden. Des Weiteren kann die Zusammensetzung, beispielsweise der Salzgehalt der Nährlösung, welche den Mikroorganismen ständig zugeleitet wird, verändert werden. Es können auch zusätzliche Nährstoffe wie beispielsweise Zuckermoleküle zugeführt werden, die geeignet sind die Bildung von Oberflächenmolekülen zu induzieren. Die Verwendung eines Induktionsreaktors ist insbesondere vorteilhaft wenn die Bildung der Gallerte von Zuckermolekülen abhängig ist und durch diese induziert wird. Steht während der Kultivierung der Mikroorganismen im Photobioreaktor als einzige Kohlenstoffquelle CO2 zur Verfügung, wird eine effiziente CO2-Fixierung gewährleistet. Gleichzeitig bilden die Mikroorganismen kaum Gallerte, weil ihnen hierzu die essentiellen Zuckermoleküle fehlen. Im Induktionsreaktor werden dann Zuckermoleküle zugegeben und die Mikroorganismen so zur Bildung der Gallerte veranlasst. Werden die Mikroorganismen anschließend mit den magnetisierbaren Partikeln gemischt, lagern sich diese in die Gallerte ein. Der Begriff "magnetisierbare Partikel" bezeichnet kleine Teilchen beispielsweise in Form von Pulver oder Späne, die ferromagnetische Elemente enthalten und deshalb die Eigenschaft besitzen, sich in einem Magnetfeld auszurichten. Als magnetisierbare Partikel besonders bevorzugt sind Magnetitpartikel. Diese binden effizient an Mikroorganismen bzw. lagern sich in deren Gallerte ein. Dabei wirken die magnetischen Kräfte der Partikel aufeinander, wodurch die Agglomeratbildung der Mikroorganismen verstärkt wird. Sobald die mit Magnetit beladenen Mikroorganismen durch ein Magnetfeld geführt werden, richten sich die Magnetitpartikel in diesem aus und bewegen sich zu dem Magnetpol hin, wobei sie die Mikroorganismen mitziehen. Ein weiterer Vorteil von Magnetit liegt darin, dass es auf dem Rohstoffmarkt gut verfügbar und daher kostengünstig ist. Darüber hinaus ist Magnetit nicht giftig, sodass eine vollständige Entfernung der Partikel nach dem Trocknen der Mikroorganismen nicht zwingend notwendig ist. Der Induktionsreaktor kann ferner mit einer Einfüllvorrichtung für die magnetisierbaren Partikel ausgebildet sein. Dabei reicht die zur Kultivierung notwendige Bewegung des Mediums aus, um die Mikroorganismen gleichmäßig mit den magnetisierbaren Partikeln zu beladen, sodass kein zusätzliches Mischen notwendig ist. Alternativ kann die Einfüllvorrichtung auch in einer Transportleitung, die vom Induktionsreaktor zum magnetischen Separator führt, angeordnet sein.
