Abstract
Vorrichtung und Verfahren zur Extraktion von Nukleinsäuren, umfassend einen Hohlkörper, vorzugsweise eine Pipettenspitze, durch den eine Flüssigkeit geleitet wird, dadurch gekennzeichnet, dass in diesem Hohlkörper ein Material mit strukturierter Oberfläche so angeordnet ist, das es von einer Flüssigkeit umspült werden kann. Nach Lyse der Probe und Einstellung notwendiger Bindungsbedingungen für die Adsorption der Nukleinsäuren an das Trägermaterial wird der Ansatz mittels eines Pipettiervorganges am vertikal in der Pipettenspitze verbrachten nukleinsäurebindenden Material mehrmals „vorbei pipettiert“. Das Verfahren offenbart noch einen weiteren Vorteil. Bisher eingesetzte Materialien zur Anbindung von Nukleinsäuren waren entweder poröse Ionenaustauscher-Materialien sowie poröse silicatische Träger bzw. poröse Gläser. Überaschenderweise zeigte sich Folgendes. Wenn man eine Nukleinsäure enthaltende Proben mit Extraktionsreagenzien von kommerziell verfügbaren Kits (z.B. innuPREP Blood DNA Kit/IPC16, Analytik Jena AG; DNeasy Blood & Tissue Kit; Fa. Qiagen) bearbeitet und man anstelle eines mineralischen Trägermaterials ein Polymermaterial (z.B.Polypropylen) einsetzt und dieses in Kontakt mit der in Bearbeitung befindlichen Probe bringt, bindet Nukleinsäure an das Plastikmaterial. Die nachfolgenden Schritte des Waschens können mit bekannten alkoholhaltigen Waschpuffern oder auch nur mit einem Alkohol durchgeführt werden, ebenso können für die Elution bekannte Niedrigsalzpuffer oder auch Wasser eingesetzt werden. Wesentlich dabei ist, dass das Plastikmaterial keine glatte Oberfläche besitzt, sondern die Oberfläche strukturiert bzw. aufgeraut ist. Dabei reichen schon einige Rillen im Material aus, um das Material als ein geeignetes Material zur Bindung von Nukleinsäuren zu nutzen. Als Extraktionsreagenzien können klassische Kitreagenzien unterschiedlicher Art verwendet werden. Ebenso arbeitet das Extraktionsprotokoll nach dem bekanntem Schema Lysieren-Binden-Waschen-Eluieren. Verbringt man solche neuartigen Materialien vertikal in eine Pipettenspitze, dann können diese Materialien in exzellenter Weise für die Extraktion von Nukleinsäuren eingesetzt werden. Das erfindungsgemäße Verfahren läuft damit wie folgt ab:
- 1. Lysieren einer Nukleinsäure enthaltenden Probe (oder Bereitstellen einer Nukleinsäure enthaltenden Probe). Dazu wird die Probe mit einem klassischen Lysepuffer oder einem Puffer versetzt, die man bisher aus Verfahren zur Isolierung von Nukleinsäuren kennt. Diese Puffer können chaotrope Salze oder nichtchaotrope Salze oder Mischungen dieser beiden Gruppen enthalten. Darüber hinaus können diese Puffer weitere Komponenten enthalten wie Chealtbildner, Trispuffer, Netz- und Dispergiermittel etc. Das Verfahren funktioniert aber auch mit Puffern, welche keine Salze enthalten und z.B. nur aus einem Detergenz, Tris und EDTA bestehen. Zusätzlich können auch noch proteolytische Enzyme eingesetzt werden.
- 2. Zugabe eines Bindungspuffers, welcher eine organische Komponente (vorzugsweise Alkohole oder Ketone) und ggf. weitere Zusätze enthält.
- 3. Aufziehen dieser Mischung mit einer Pipettenspitze, in welcher sich vertikal angeordnet das nukleinsäurebindende Material befindet (z.B. Plastikmaterial, dessen Oberfläche angeraut ist). Mehrmaliges Auf und Abpipettieren (die Flüssigkeit bewegt sich dabei am Material vorbei). Nach letztem Auspipettieren ist die erfindungsgemäße Pipettenspitze leer.
- 4. Entfernen der erfindungsgemäßen Pipettenspitzes aus der Probe
- 5. Eintauchen der erfindungsgemäßen Pipettenspitze in eine Waschpufferlösung. Mehrmaliges Auf und Abpipettieren (die Flüssigkeit bewegt sich dabei am Material vorbei). Nach letztem Auspipettieren ist die erfindungsgemäße Pipettenspitze leer.
- 6. Trocknung durch z.B. multiple Pipettierschritte zur Entfernung des restlichen Alkohols vom Waschpuffer. Auch ist eine mögliche Trocknung mittels erhöhter Temperatur möglich.
- 7. Ablösen der Nukleinsäure mit einem Elutionspuffer (Niedrigsalzpuffer oder Wasser). Dazu mehrmaliges Auf und Abpipettieren (der Elutionspuffer bewegt sich dabei am Material vorbei). Nach letztem Auspipettieren befindet sich die Nukleinsäure in der flüssigen Phase.

Dabei umgeht die erfindungsgemäße Pipettenspitze die bekannten Nachteile aus dem Stand der Technik, welche durch die bisherige konstruktive Anordnung bzw. Mittels eines Lysepuffers (Lysis Solution CBV) sowie von Proteinase K wurden die Zellen bei 60°C für 15 min lysiert. Die Lyse erfolgte in einem 2.0 ml Reaktionsgefäß. Nach Lyse wurde der Ansatz mit 400 µl Isopropanol versetzt. Nachfolgend wurde die modifizierte Pipettenspitze eingesetzt und mittels einer Pipette wurde der Ansatz 20 × auf- und abpipettiert. Danach wurden 3 weitere 2 ml- Reaktionsgefäße mit den alkoholischen Waschpuffern (Washing Solution LS, 80%iger Ethanol, 80%iger Ethanol) befüllt. Die Pipettenspitze wurde dann sukzessive in die jeweiligen Waschpuffer getaucht und es wurde jeweils 5 × auf- und abpipettiert. Nach dem letzten Waschschritt wurde die Spitze getrocknet und damit der restliche Ethanol entfernt. Die Elution der gebundenen Nukleinsäure erfolgte mit 100 µl Elution Buffer. Dieser wurde wiederum in ein 2 ml- Reaktionsgefäß gegeben. Es wurde 30 × auf- und abpipettiert. Nach Entfernung der Pipettenspitze liegt die isolierte Nukleinsäure im Reaktionsgefäß vor. Die automatisierte Extraktion wurde mit dem Extraktionsautomaten InnuPure C16 (Analytik Jena AG) durchgeführt. Dieser Extraktionsautomat basiert auf einer Magnetpartikel-Nukleinsäureextraktion. Für die Durchführung einer Nukleinsäureextraktion nach dem erfindungsgemäßen Verfahren wurden die Pipettenspitzen, die für den Extraktionsautomaten genutzt werden, so geändert, dass sie dem erfindungsgemäßen Mittel entsprechen. In die Pipettenspitzen wurde in das untere Drittel vertikal, eine aus einem angerauten Polymer hergestellte Scheibe eingebracht, die das Lumen nicht verschließt und somit die Pipettierfunktion der Pipettenspitzen erhalten bleibt. Es wurden dabei angeraute Scheiben aus verschiedenen Materialien eingesetzt. Für die Nukleinsäureextraktion wurden jeweils 5 × 105 NIH 3T3 Zellen eingesetzt.
