2. Diagnostisches Mittel zur Bestimmung von Hydroperoxiden bzw. Substanzen, die unter Freisetzung
von Hydroperoxid reagieren, enthaltend Peroxidase oder eine peroxidatisch wirksame Substanz
und ein Chromogen, dadurch gekennzeichnet, daß als Chromogen Substanzen der allgemeinen Formel I gemäß Anspruch 1 enthalten sind.
3. Diagnostisches Mittel zur Bestimmung von Hämoglobin oder anderen peroxidatisch wirksamen
Substanzen, enthaltend ein Hydroperoxid oder Hydroperoxid bildende Substanzen und ein
Chromogen, dadurch gekennzeichnet, daß als Chromogen Substanzen der allgemeinen Formel I
gemäß Anspruch 1 enthalten sind.

60 Die Erfindung betrifft ein Verfahren zur Bestimmung von Hydroperoxiden, bzw. von Substanzen,
die unter Freisetzung von Hydroperoxid reagieren, sowie von Peroxidase bzw. peroxidatisch wirksamen
Substanzen mit Hilfe eines Chromogens unter Verwendung der Reaktion der Hydroperoxide mit Peroxidase
bzw. den peroxidatisch wirksamen Substanzen durch Auswertung der Verfärbung. Die Erfindung
betrifft ferner diagnostische Mittel zur Durchführung derartiger Verfahren.
Von besonderem diagnostischem Interesse ist der Nachweis von Glukose im Harn, Blut und Serum bei
Diabetes sowie der Nachweis von peroxidatisch wirkenden Substanzen, wie Hämoglobin im Harn und
Blut, und der Nachweis von Hydroperoxiden z. B. in der Milchwirtschaft, Kosmetik und Polymerchemie.
Es sind eine Reihe von Verbindungen bekannt, die von Wasserstoffperoxid und Peroxidase als Katalysator
zu einem Farbstoff oxidiert werden (vgl. schweizerische Patentschrift 339 755 und britische Patentschrift
1035 911). Die Substanzen, die bevorzugt verwendet werden, sind z. B.: Benzidin, o-Dianisidin,
o-Tolidin, Guajakol usw. Diese Verbindungen sind aber teilweise nicht sehr stabil und können nach den
neuesten Erkenntnissen gesundheitsschädlich sein, so daß ihre Handhabung nicht ungefährlich erscheint.
Der Erfindung liegt deshalb die Aufgabe zugrunde, Substanzen zu finden, die als Chromogene für ein
Verfahren der obengenannten Art geeignet und zugleich stabil und unschädlich sind.
Es wurde überraschenderweise eine Substanzgruppe gefunden, die zum Teil bessere Indikatoreigenschaften
als die bisher verwendeten Benzidinkörper zeigt, aber dabei absolut beständig ist und sich demzufolge
besonders gut dazu eignet, Wasserstoffperoxid und andere Hydroperoxide sowohl im optischen Test als
auch mit Hilfe von Reagenzpapieren zu bestimmen. Die Substanzen sind physiologisch unschädlich und
besitzen die allgemeine Formel I

mit R3 einen gegebenenfalls substituierten Ring bilden können.
Die erfindungsgemäße Aufgabe wird somit dadurch gelöst, daß man in einem Verfahren der eingangs
genannten Art als Chromogene Substanzen der allgemeinen Formel I verwendet. Die Bestimmung von
Hydroperoxiden nach dem erfindungsgemäßen Verfahren ist für gekoppelte und ungekoppelte Enzymreaktionen
geeignet, wie z. B. für die Bestimmung von Glukose, Galaktose, Aminosäuren, Harnsäure, Peroxiden,
Hämoglobin, Peroxidase oder anderen peroxidatisch wirksamen Substanzen sowie Enzymaktivitäten.
Bei der Bestimmung von Glukose beispielsweise wird diese von Glukoseoxidase zu Glukonsäure
oxidiert, wobei der Luftsauerstoff zu Wasserstoffperoxid reduziert wird. Das Wasserstoffperoxid oxidiert
dann mittels Peroxidase oder einer peroxidatisch wirksamen Substanz den erfindungsgemäß verwendeten
Indikator zum entsprechenden Farbstoff. Weitere Beispiele für derartige analytisch brauchbare Enzymsysteme,
die unter Freisetzung von Wasserstoffperoxid reagieren, sind L-Aminosäureoxidase + L-Aminosäuren,
D-Aminosäureoxidase + D-Aminosäuren, Uricase + Harnsäure, Xanthinoxidase + Hypoxanthin
bzw. Xanthin, Glycinoxidase + Glycin, Monoaminooxidase + Monoamin (wie Adrenalin, Mescalin usw.),
Diaminoxidase + Diamine (wie Histamin), Luciferase + Luciferin, D-Asparaginsäureoxidase + D-Asparaginsäure,
Leber-Aldehydoxidase + Aldehyd, Galaktoseoxidase + Galaktose, Edsons Flavinenzym
+ Milchsäure.
Die Auswertung der Verfärbung erfolgt beispielsweise durch optische Ausmessung im Spektralphotometer
oder bei Verwendung von Testpapierstreifen durch Vergleich der Farbstärke mit Blindproben
bekannter Zusammensetzung bzw. Farbtafeln.
Das erfindungsgemäße Verfahren und zu seiner Durchführung geeignete diagnostische Mittel werden
an Hand der folgenden Beispiele näher erläutert.

7,5 mg Peroxidase und 143 mg Glukoseoxidase werden in 100 ml eines 0,1-m Citrat- oder Phosphatpuffers
vom pH 5,6 gelöst. Mit dieser Lösung imprägniert man Filterpapier (2316 der Firma Schleicher &
Schüll). Danach werden 0,30 g 2,3-Dihydroxybenzal-(vanilloyl)-hydrazon
in 100 ml Alkohol gelöst und das Filterpapier noch einmal imprägniert. Beim Eintauchen
in glukosehaltigen Harn zeigt das Papier ab etwa 50mg-% eine Rosafärbung, die je nach
Intensität den Glukosegehalt angibt.

7,5 mg Peroxidase, 143 mg Glukoseoxidase und 200 mg alginsaures Natrium werden in 100 ml 0,4-m
Citrat- oder Phosphatpuffer von pH 5,6 gelöst. Mit dieser Lösung wird ein Filterpapier (23 S der Firma
Schleicher & Schüll) imprägniert. In einem zweiten Imprägniervorgang wird dasselbe Papier mit einer
Lösung von 0,316 g o-Vanillin-(vanilloyl)-hydrazon in 100 ml Alkohol getränkt. Danach wird das Papier
in einem dritten Imprägniervorgang mit einer Lösung von 3 g Paraffin in 100 ml Petroläther imprägniert.
Bringt man auf ein so hergestelltes Papier einen Tropfen Blut mit 50mg-% Glukose, läßt 1 Minute
einwirken und wäscht das Blut mit Wasser ab, so zeigt das Papier eine deutlich violette Färbung. Die Farbintensität
hängt von der Glukosekonzentration im Blut ab.

Es werden Tabletten folgender Zusammensetzung gepreßt:
o-Vanillin-(vanilloyl)-hydrazon 0,002 g
Peroxidase 0,001 g
Glucoseoxidase 0,002 g
sek. Natriumphosphat 0,0069 g
prim. Natriumphosphat 0,013 g
Eine solche Tablette wird in 5 ml Alkohol/Wasser (1:1) gelöst, mit 0,2 ml der zu messenden Glukoselösung
versetzt, mit Alkohol auf 10 ml aufgefüllt und bei einer Wellenlänge von 520 nm colorimetrisch
gemessen. Aus der gemessenen Extinktion wird der Glukosegehalt auf Grund einer Eichkurve ermittelt.
7,5 mg Peroxidase werden in 100 m! eines 0,1-m Citrat- oder Phosphatpuffers von pH 5,6 gelöst und
damit ein Filterpapier (2316 der Firma Schleicher & Schüll) imprägniert. Danach werden
0,31 g o-Vanil!in-(o-vanilloyl)-hydrazon in 100 ml Alkohol
gelöst und das Filterpapier nochmals imprägniert.
Beim Eintauchen in Lösungen von Wasser-Stoffperoxid verschiedener Konzentrationen werden
die Papiere, entsprechend der Konzentration, verschieden stark violett gefärbt.

Je 1 mMol der in nachstehender Tabelle aufgeführten Indikatoren wird in 1 ml Methanol gelöst bzw.
suspendiert, mit ! ml einer Lösung von 143 mg Glukoseoxidase, 7,5 mg Peroxidase in 100 ml Phosphat- oder Citratpuffer (pH 5,6) sowie 1 ml einer
Glukoselösung versetzt. Man erhält die in der Tabelle angeführten Farbreaktionen:

Ein Filterpapier (2316 der Firma Schleicher & Schüll) wird mit einer Lösung von 0,316 g o-Vanillin-(vanilloyl)-hydrazon
in 100 ml Methanol imprägniert.
500 ml Akazien pflanzenschleim, der durch Kochen von 200 g Akazien in 500 ml Wasser hergestellt wurde,
und 25 ml Cumolhydroperoxid werden zusammengegeben und 5 Minuten im Mixer heftig gerührt.
Weiter werden in 1,51 Wasser 163 g Natriumeitrat und 37 g Zitronensäure gelöst. In dem auf diese Weise
erhaltenen Citratpuffer werden 3,5 g Gelatine gelöst, worauf man die obengenannte Lösung zugibt. Die
vereinigten Lösungen vermischt man mit 5 ml einer l%igen wäßrigen Emulsion einer peroxidhaltigen
Stärke und — nachdem wieder gut durchgemischt wurde — mit 50 ml Natriumlaurylsulfat und rührt
5 Minuten weiter. Die so erhaltene Emulsion wird dann 5 Minuten in einen Homogenisator mit
100 kg/cm2 Druck gegeben. Zu der homogenisierten Mischung gibt man 250 ml des wie oben hergestellten
Citratpuffers. Die mit dem Indikator imprägnierten Papiere werden in die erhaltene Emulsion getaucht
und 16 Stunden bei 5Ö°C in einem Umlufttrockenschrank getrocknet. Die so erhaltenen Testpapiere
ergeben beim Eintauchen in einen bluthaltigen Urin eine violette Farbreaktion.
Claims (1)
1. Verfahren zur Bestimmung von Hydroperoxiden bzw. von Substanzen, die unter Freisetzung
von Hydroperoxid reagieren, sowie von Peroxidase bzw. peroxidatisch wirksamen Substanzen mit.
Hilfe eines Chromogens unter Verwendung der Reaktion der Hydroperoxide mit Peroxidase bzw.
den peroxidatisch wirksamen Substanzen durch Auswertung der Verfärbung, dadurch gekennzeichnet,
daß man als Chromogene Substanzen der allgemeinen Formel I
