Description
a) Saccharose mit Mesityler.s-jifonylchlorid umsetzt,
b) aus dem so erhaltenen Reaktionsgemisch ar-D-Glucopyranosyl-l.e-di-O-mesitylensuIfonyl-^-D-fructof-ο
uranosid durch Chromatographie an Kieselgel gewinnt,
c) sämtliche Hydroxylgruppen der gemäß Verfahrensstufe b) gewonnenen Verbindung acetyliert und
d) den gemäß Verfahrensstufe c) erhaltenen Ester mit Lithiumchlorid in Dimethylformamid umsetzt.
4. Verfahren zur Herstellung der Verbindung nach Anspruch 2. dadurch gekennzeichnet, daß man 6-Chlor-6-desoxy-dr-D-glucopyranosyl-l,6-dichIor-l,6-didesoxy-/?-D-fructofuranosid
mit Sulfurylchlorid in einem inerten Lösungsmittel in Gegenwart einer Base bei einer Temperatur von —40 bis 0° C umsetzt
Die Erfindung betrifft die in den Patentansprüchen 1 und 2 genannten chlorsubstituierten Dissaccharide und
Verfahren zu ih-er Herstellung.
Erfindungsgemäß werden 4,6-Dichlor-4,6-didesoxy-ar-D-gaIactopyranosyl- 1,6-dichlor-l ,6-diu'esoxy-/?-D-fructofuranosid

bereitgestellt. Außerdem wurde ö-Chlor-e-desoxy-Ä-D-glucopyranosyl-l.e-dichlor-l.ö-didesoxyvi-D-fructofuranosid

als Zwischenprodukt zur Herstellung der Verbindung gemäß Anspruch 2 hergestellt.
Die Verbindungen gemäß Anspruch 1 und 2 sind Zwischenprodukte und haben den zusätzlichen Vorteil, daß
sie einen sehr süßen Geschmack besitzen, wie in der DE-AS 27 OO 036 beschrieben, so daß sie potentielle
Anwendung als Zuckersubstitut finden.
Die Verbindung gemäß Anspruch 1 kann durch Umsetzung der Saccharose mit Mesitylensulfonylchlorid,
Abtrennen der gewünschten Dimesitylensulfonate von dem Gemisch der substituierten Saccharose, zum Beispiel
durch Chromatographie, und Ersetzen der Mesitylensulfonyloxysubstituenten durch Chloratome unter
Reaktion mit Lithiumchlorid, nachdem die freien Hydroxylgruppen durch Veresterung geschützt worden sind,
hergestellt werden. Diese Veresterung kann beispielsweise mit Acetanhydrid erfolgen.
Aus den oben erwähnten Gemischen der Mesitylensulfonat-substituierten Saccharose kann das r,6,6'-trisub-
stituierte Saccharosederivat, das in verhältnismäßig großem Anteil zugegen ist, abgetrennt werden. Dieses
trisubstituierte Derivat kann in analoger Weise, wie oben angegeben, in das l',6,6'-trichlorsubstituierte Derivat ~
umgewandelt werden. Aus diesem trichlorsubstituierten Derivat kann das gewünschte Tetrachlor-substituierte \,
Derivat in guten Ausbeuten nach Esterspaltung und nachfolgender Umsetzung mit Sulfurylchlorid bei tiefen t
Temperaturen hergestellt werden. $
Die Herstellung der erfindungsgemäßen Verbindungen wird durch die folgenden Beispiele erläutert. si

Saccharose wurde 6 Tage bei —5°C mit Mesitylensulfonylchlorid (3 Mol) in Pyridin behandelt, um als Hauptprodukt
r,6,6'-Tri-O-mesitylensulfonylsaccharose zu erhalten (vgl Carbohydrate Research, 1975,44, C 12 und
C 13). Die Konzentrierung des Reaktionsgemisches und übliche Extraktion des Rückstandes mit Chloroform
ergab 6-O-Mesitylensulfonyl-*-D-glucx>pyranosyl-l,6-di-O-mesityIensulfonyI-/?-D-fructofuranosid (F 135°C unter
Zersetzung). Eine weitere Menge dieser Verbindung wurde aus den Mutterlaugen nach Chromatographie
auf Kieselgel erhalten.
Die Mutterlaugen aus dem Konzentrierungs- und Extraktionsschritt des Beispiels 1 wurden der Chromatographie
auf Kieselgel unterworfen, um ein Gemisch von isomeren Disulfonaten zu erhalten. Nach Zugabe von
2-Propanol oder Äthanol kristallisierte 1 ',ö'-Dimesitylensulfc nat der Formel

zu erhalten, worin OAc eine Acetatgruppe bedeutet.
Diese Verbindung wurde dann entacetyliert durch Behandeln mit methanolischem Natriummethoxid; das
Produkt (1 Mol) wurde in Pyridin gelöst, auf -400C gekühlt und tropfenweise mit einer Lösung von Sulfurylchlorid
(10 MoI) in Chloroform versetzt. Das Reaktionsgemisch wurde auf O0C erwärmen gelassen, 24 Stunden
bei dieser Temperatur gehalten, wonach es in eiskalte 10%ige Schwefelsäure gegossen wurde. Das Gemisch
wurde mit Chloroform extrahiert, der Extrakt wurde mit wäßrigem Natriumbicarbonat gewaschen, dann mit
Wasser gewaschen und mit MgSO getrocknet. Verdampfung des Chloroforms und folgende Chlorsulfatation in
Methanol unter Verwendung von Natriumjodid und Natriumcarbonat (vgl. Carbohydrate Research 1975, 44,
37—44), ergab die rohe l',4,6,6'-Tetrachlorgalactosaccharose. Diese wurde durch Umwandlung in das Tetra-O-acetat
und nachfolgende Säulenchromatographie sowie Des-O-acetylierung gereinigt.
Die gewünschte Tetrachlorverbindung wurde anhand der l3C-NMR-Spektroskopie und Massenspektroskopie
und durch Oberführung in das Tetramethansulfonat [F. 120— 121°C,|>]d + 65,5° (c 1-Chloroform)]charakterisiert.

Das Dimesilrylensulfonat wurde durch Behandlung mit Acetanhydrid acetyliert und dann bei 140° C 18 Stunden
mit Lithiuinchlorid in N1N-Dimethylformamid, das eine Spur Jod enthält, umgesetzt, um das 1 ',6'-DichIorid in
hoher Ausbeute zu erhalten.
Das Ä-D-Glucopyranosyl-l,6-dichlor-l,6-didesoxy-/?-D-fructofuranosid wurde durch 1H-NMR-, Massenspektroskopie
und durch Oberführung in die entsprechenden 1 ',6'-Didesoxyverbindung charakterisiert.
Die rAe'-Tri-O-mesitylensulfonylsaccharose, hergestellt nach Beispiel 1, wurde acetyliert durch Behandlung
mit Acetanhydrid und Umsetzen 13 Stunden bei 1400C mit Lithiumchlorid in Ν,Ν-Dimethylformamid, das eine
