Description
Die vorliegende Erfindung betrifft ein Verfahren zur
Herstellung von Calciumphosphat-Keramik mit einer porösen
Oberfläche.
Keramiken, die auf Calciumphosphat basieren, zeigen neulich
eine verstärkte Verwendung als künstliche Zahnimplantate,
künstliche Knochen usw., da sie eine ausgezeichnete
Affinität zu dem lebenden Körper besitzen.
Bei der Anwendung von Calciumphosphat-Keramik in einem
lebenden Körper als Zahnimplantate, künstliche Knochen
und dergleichen, ist es bekannt, dass die Affinität
verbessert wird, wenn die Oberflächenporosität ansteigt,
und somit die Zirkulation der Körperflüssigkeit gefördert
wird.
Dementsprechend werden verschiedene Herstellungsverfahren
für poröse Calciumphosphat-Keramiken vorgeschlagen. Die
üblicherweise angewandten Verfahren sind die folgenden:
- 1) eine Aufschlämmung von einer auf Calciumphosphat basierenden Verbindung wird durch Hinzufügen eines Aufschäummittels aufgeschäumt, zu einer vorbestimmten Form gebildet, getrocknet und gebrannt (wie in "Sintered hydroxyapatite as a bioceramics", Philips tech. Rev. Bd. 37, S. 234-236, 1977, Nr. 9/10 beschrieben);
- 2) ein Pulver von einer auf Calciumphosphat basierenden Verbindung wird mit einer pyrolytischen, organischen Substanz vermengt, geformt, vorkalziniert und gebrannt (wie in JP-A-62 22 632 beschrieben) (der Ausdruck "JP-A", wie er hier verwendet wird, bedeutet eine nicht geprüfte, offengelegte, japanische Patentanmeldung).
Jedoch tendieren die zuvor genannten Verfahren dazu,
kompliziert zu sein, da sie das Aufschäumen mit
Aufschäummitteln oder das Vermengen mit pyrolytischen,
organischem Material beinhalten. Darüber hinaus ist die
Festigkeit der porösen Keramik beträchtlich niedrig im
Vergleich zu nicht-poröser Keramik, da sie vom Inneren
bis zur Oberfläche vollständig aus porösem Material
zusammengesetzt ist.
Eine Aufgabe der vorliegenden Erfindung besteht darin, ein
Verfahren zur Herstellung von fester Calciumphosphat-Keramik
mit einer porösen Oberfläche bereitzustellen, wobei über
ein einfaches Verfahren Poren in die Oberfläche von
unbehandelter Calciumphosphat-Keramik eingeführt werden.
Eine weitere Aufgabe der vorliegenden Erfindung besteht
darin, ein Verfahren zur Herstellung von Calciumphosphat-
Keramik mit einer ausgezeichneten Affinität zu dem
lebenden Körper zu schaffen.
Weitere Aufgaben und Wirkungen der vorliegenden Erfindung
werden aus der folgenden Beschreibung deutlich.
Die oben genannten Aufgaben der vorliegenden Erfindung
werden durch ein Verfahren zur Herstellung von
Calciumphosphat-Keramik mit einer porösen Oberfläche
gelöst, das die folgenden Schritte umfasst:
- a) Herstellen von unbehandelter Calciumphosphat-Keramik, die eine Mischung von Hydroxyapatit und Tricalciumphosphat umfasst; und
- b) Behandeln der unbehandelten Keramik mit einer sauren Lösung, um das Tricalciumphosphat in der Oberfläche der Keramik selektiv herauszulösen.
Fig. 1 stellt eine Mikroaufnahme mit einem
Vergrösserungsfaktor von 1000 dar, die die
Oberflächentextur der Calciumphosphat-Keramik
mit einer porösen Oberfläche, die gemäss der
vorliegenden Erfindung hergestellt wurde,
zeigt.
Es ist bekannt, dass Calciumphosphat-Keramik sich in
Säuren auflösen kann. Die Löslichkeit unterscheidet
sich von einer Probe zur anderen und die Löslichkeit
von Tricalciumphosphat (Ca3(PO4)2) ist grösser als die
von Hydroxyapatit (Ca10(PO4)6(OH)2). Die vorliegende
Erfindung basiert auf dieser Kenntnis und benutzt den
Unterschied in der Löslichkeit zwischen
Tricalciumphosphat und Hydroxyapatit. Das bedeutet, dass
unbehandelte Calciumphosphat-Keramik, die eine Mischung
von Hydroxyapatit und Tricalciumphosphat umfasst, mit
einer sauren Lösung behandelt wird, um das Tricalciumphosphat
in der Oberfläche selektiv herauszulösen. Damit wird
eine poröse Oberfläche über ein einfaches Verfahren
erhalten.
Die Calciumphosphat-Keramik der vorliegenden Erfindung
zeigt, wenn sie für künstliche Zahnimplantate und künstliche
Knochen eingesetzt wird, eine ausgezeichnete vitale
Affinität, und diese Affinität wird der porösen Oberfläche,
die durch das Verfahren der vorliegenden Erfindung
erhalten wurde, zugeschrieben. Darüber hinaus behält
die Keramik, nachdem sie diesem Verfahren unterzogen wurde,
eine ausreichende Festigkeit, da ihre innere Dichte
beibehalten ist.
In einer Ausführungsform der vorliegenden Erfindung wird
eine Pulvermischung von Hydroxyapatit und
Tricalciumphosphat wie gewünscht geformt, falls notwendig
vorkalziniert, und gebrannt, um unbehandelte
Calciumphosphat-Keramik zu erhalten.
Bekannte Trocken- und Nasssyntheseverfahren
können für die Synthese von
Hydroxyapatit und Tricalciumphosphat, die als
Ausgangsmaterialien verwendet werden, angewandt werden.
In dem Nassverfahren wird Phosphorsäure und/oder ihr
Calciumsalz in Reaktion mit einer Calciumverbindung in
Wasser gebracht. Es ist möglich, die Zusammensetzung des
unbehandelten Produktes zu kontrollieren, indem man das
Ca/P-molare Verhältnis des Ausgangsmaterials anpasst,
um ein gewünschtes Produkt zu erhalten, z.B. ein
unbehandeltes Produkt, das hauptsächlich entweder
Hydroxyapatit oder Tricalciumphosphat umfasst, eine
Mischung von Hydroxyapatit und Tricalciumphosphat, usw.
Das Nassverfahren wird z.B. in Wallaeys, R., Ann. Chim.
(Paris), Bd. 7, 808 und 823 (1952); Moreno, E.C.,
Gregory, T.M., Brown, W.E., J. Res. Nat. Bur. Stand.,
Bd. 72A, 773 (1968); und L.C. Bell, H. Mika, B.J. Kruger,
Archs. Oral. Biol., Bd. 23, 329 bis 336 (1978) beschrieben.
Das Trockenverfahren ist z.B. in Quinaux, N., Arch. Intern.
Physiol. Biochim., Bd. 72, 337 (1964) und Chem. Abstr.,
Bd. 60, 15418a (1964); und Liteanu, C., Macarouci, D.,
Studii Cercetari Chim., Bd. 13, 157 (1962), beschrieben.
In der vorliegenden Erfindung kann Hydroxyapatit und
Tricalciumphosphat entweder getrennt oder zur gleichen
Zeit synthetisiert werden. Wenn die Synthese getrennt
ausgeführt wird, dann werden die Pulver von Hydroxyapatit
und Tricalciumphosphat anschliessend gemischt. Wenn die
Synthese gleichzeitig erfolgt, dann wird eine Mischung
durch das Nassverfahren erhalten, die, falls notwendig,
später pulverisiert werden kann. Die Mischung von
Hydroxyapatit und Tricalciumphosphat enthält vorzugsweise
ungefähr 20 bis 80% der Tricalciumphosphatphase, das
bedeutet, dass ein Ca/P-molares Verhältnis im Bereich
von 1,53/1 bis 1,64/1 erreicht ist, wobei der Bereich
von 1,57/1 bis 1,64/1 besonders bevorzugt ist. Im Prinzip
ist es möglich, die vorliegende Erfindung im gesamten
Bereich der Konzentrationsverhältnisse anzuwenden,
vorausgesetzt, dass sowohl Hydroxyapatit als auch
Tricalciumphosphat vorliegen. Die Porosität des Endproduktes
wird bei Ausgangsmaterialien, die eine grössere Menge an
Tricalciumphosphat enthalten, höher.
Nach der Synthese wird die Mischung von Hydroxyapatit
und Tricalciumphosphat dann wie gewünscht durch
Druckformen, Gussformen usw. geformt. Granulate können
erhalten werden, indem man verschiedene Granularisierungsverfahren
anwendet. In diesem Fall können Binder, wie Wasser und
organische Lösungsmittel, beigegeben werden. Die so
erhaltenen Formen werden, falls notwendig, vorkalziniert
und dann gebrannt. Die Brenntemperatur für die vorliegende
Erfindung liegt vorzugsweise im Bereich von 700 bis 1400°C,
wobei 1000 bis 1300°C besonders bevorzugt sind. Bei diesem
Verfahren ist der Porendurchmesser bei den Proben, die
bei höheren Temperaturen gebrannt wurden, grösser, da
das Kornwachstum mit ansteigender Temperatur gefördert
wird, so dass nach der Säurebehandlung grössere Poren
zurückbleiben.
Die Porosität der porösen Oberfläche der
Calciumphosphat-Keramik der vorliegenden Erfindung liegt
vorzugsweise bei 20 bis 60%. Die Porosität kann kontrolliert
werden, indem man den Mengenanteil von Tricalciumphosphat
in der unbehandelten Calciumphosphat-Keramik ändert,
oder indem man die Kontaktzeit der unbehandelten Keramik
mit der sauren Lösung ändert. Jedoch wird die Porosität
vorzugsweise dadurch kontrolliert, dass man den
Mengenanteil von Tricalciumphosphat ändert.
Salzsäure, Salpetersäure, Schwefelsäure und dergleichen
werden geeigneterweise als saure Lösung verwendet, die
aber darauf nicht beschränkt ist. Die saure Lösung sollte
vorzugsweise einen pH-Wert im Bereich von 1 bis 5,5
besitzen. Wenn der pH-Wert niedriger als 1 liegt, dann
löst sich Hydroxyapatit, und wenn der pH-Wert höher als
5,5 liegt, dann wird Tricalciumphosphat nicht effektiv
genug herausgelöst, um eine poröse Oberfläche zu bilden.
Die Kontaktzeit mit der sauren Lösung unterscheidet sich
gemäss der Säurekonzentration, jedoch sind 1 Sekunde bis
24 Stunden für einen normalen Fall geeignet.
Die Calciumphosphat-Keramik der vorliegenden Erfindung
mit einer porösen Oberfläche wird in geeigneter Weise für
Implantationsmaterialien, wie künstliche Zahnwurzeln und
künstliche Knochen, und auch als Säulenfüllmaterial für
die Trennung von Biopolymeren verwendet.
Die vorliegende Erfindung wird detaillierter mit Bezug
auf das folgende Beispiel beschrieben, wobei sie aber
nicht darauf beschränkt sein soll. Alle Verhältnisse,
Teile, Prozentsätze usw. sind auf das Gewicht bezogen,
sofern nicht anders angegeben.
Die Pulver von Hydroxyapatit und Tricalciumphosphat, die
über bekannte Nasssyntheseverfahren hergestellt wurden,
wurden gemischt, um eine Pulvermischung mit einem
Ca/P-molaren Verhältnis von 1,55/1 zu erhalten. Die
Mischung wurde dann in einer Metallgussform geformt, und
bei 1200°C 4 Stunden lang gebrannt, um eine unbehandelte
Calciumphosphat-Keramik zu schaffen.
Die obige, unbehandelte Calciumphosphat-Keramik wurde
in eine wässrige Salzsäurelösung mit einem pH-Wert von 3
bei Raumtemperatur 10 Sekunden lang eingetaucht und
anschliessend aus der Lösung herausgezogen. Die durch
das Eintauchen hergestellte Keramik besitzt eine poröse
Oberfläche, wobei die poröse Oberfläche eine feine
Unregelmässigkeit aufweist, die der selektiven Herauslösung
von Tricalciumphosphat zugeschrieben wird.
Fig. 1 zeigt eine Mikroaufnahme der behandelten
Calciumphosphat-Keramik, die bei einer Vergrösserung von
1000 aufgenommen wurde. Die Mikroaufnahme zeigt eine
Oberfläche mit feinen Poren. Der Porendurchmesser lag
im Bereich von 500 Å bis 20 µm.
Wie im vorausgehenden festgestellt wurde, kann die
vorliegende Erfindung ein extrem einfaches Verfahren zur
Herstellung von Calciumphosphat-Keramik mit einer
porösen Oberfläche bereitstellen. Dieses Verfahren umfasst
das Behandeln einer Calciumphosphat-Keramik, die eine
Mischung von Hydroxyapatit und Tricalciumphosphat umfasst,
mit einer sauren Lösung, um das Tricalciumphosphat in
der Oberfläche selektiv herauszulösen. Die behandelte
Calciumphosphat-Keramik der vorliegenden Erfindung mit
einer porösen Oberfläche wird geeigneterweise für
Implantationsmaterialien, wie künstliche Zahnwurzeln und
künstliche Knochen, aber auch für Säulenfüllmaterialien,
die für die Trennung von Biopolymeren verwendet werden,
eingesetzt und besitzen eine erhöhte Affinität zu dem
lebenden Körper.
Während die Erfindung im Detail und mit Bezug auf
spezifische Ausführungsformen beschrieben wurde, wird es
einem Fachmann klar sein, dass verschiedene Änderungen
und Modifikationen ausgeführt werden können, ohne vom
Gehalt und Umfang der Erfindung abzuweichen.
Claims (7)
1. Verfahren zur Herstellung von Calciumphosphat-Keramik
mit einer porösen Oberfläche, das die folgenden
Schritte umfasst:
- a) Herstellen einer unbehandelten Calciumphosphat-Keramik, die eine Mischung von Hydroxyapatit und Tricalciumphosphat umfasst; und
- b) Behandeln der unbehandelten Keramik mit einer sauren Lösung, um das Tricalciumphosphat in der Oberfläche der Keramik selektiv herauszulösen.
2. Verfahren zur Herstellung von Calciumphosphat-Keramik
mit einer porösen Oberfläche nach Anspruch 1, wobei
die Mischung von Hydroxyapatit und Tricalciumphosphat
kontrolliert wird, um ein molares Verhältnis (Ca/P)
von Calcium zu Phosphor im Bereich von 1,53/1 bis
1,63/1 zu erreichen.
3. Verfahren zur Herstellung von Calciumphosphat-Keramik
mit einer porösen Oberfläche nach Anspruch 2, wobei
die Mischung von Hydroxyapatit und Tricalciumphosphat
kontrolliert wird, um ein molares Verhältnis (Ca/P)
von Calcium zu Phosphor im Bereich von 1,57/1 bis
1,64/1 zu erhalten.
4. Verfahren zur Herstellung von Calciumphosphat-Keramik
mit einer porösen Oberfläche nach Anspruch 1, wobei
der Schritt der Herstellung von unbehandelter
Calciumphosphat-Keramik einen Brennschritt bei einer
Temperatur von 700 bis 1400°C umfasst.
5. Verfahren zur Herstellung von Calciumphosphat-Keramik
mit einer porösen Oberfläche nach Anspruch 4, wobei
der Schritt der Herstellung der unbehandelten
Calciumphosphat-Keramik einen Brennschritt bei einer
Temperatur von 1000 bis 1300°C umfasst.
6. Verfahren zur Herstellung von Calciumphosphat-Keramik
mit einer porösen Oberfläche nach Anspruch 1, wobei
der Schritt des Behandelns mit einer sauren Lösung
bei einem pH-Wert von 1 bis 5,5 während einer Zeitdauer
von 1 Sekunde bis 24 Stunden ausgeführt wird.
7. Auf Calciumphosphat basierende Keramik mit einer
porösen Oberfläche, die durch das Verfahren von Anspruch
1 hergestellt ist.
