Description
Die Erfindung betrifft embryonale (kardiomyozytäre und
kardiomyoblastäre) Herzmuskelzellen, ihre Herstellung und ihre
Verwendung, insbesondere zur zellvermittelten Gentransplanta
tion. Anwendungsgebiete der Erfindung sind die Medizin und die
Gentechnik.
Es gibt bereits mehrere Arbeiten, die sich mit Herzmuskelzell
kulturen von Säugetieren beschäftigen. Die ersten Untersuchungen
wurden mit Primärkulturen von embryonalem, neonatalem oder
adultem Herzgewebe durchgeführt. Vorteilhafter ist die
Verwendung von permanenten Zellinien von kardiomyozytärem
Gewebe, weil man damit größere Mengen einer homogenen
Zellpopulation auf einem definierten Entwicklungsstand zur
Verfügung stellen kann. Deshalb hat es eine Reihe von Versuchen
zur Immortalisierung von Herzmuskelzellen gegeben (u. a. A. Sen
et al, J. Biol. Chem. 263/1988/, 19132-19136). Alle bisherigen
Zellinien haben jedoch den Nachteil, daß sich bei einer
Langzeitkultivierung funktionelle Defekte bemerkbar machen.
Die Erfindung hat das Ziel, ausgehend von pluripotenten
embryonalen Stammzellen (ESC) oder Primordialen Keimzellen (EGC,
Stewart et al, Dev. Biol. 161/1994/, 626-628) nach deren
Differenzierung in spontan Pulsierende Herzzellen embryonale
kardiomyozytäre bzw. kardiomyoblastäre) Herzmuskelzellen zu
gewinnen, welche weitgehend identische Eigenschaften mit
Herzmuskelgewebe besitzen. Diese Zellen sollen für einen
therapeutischen Einsatz, ggf. nach zusätzlicher gentechnischer
Veränderung, geeignet sein. Der Erfindung liegt die Aufgabe
zugrunde, ein Vektorsystem zur Modifizierung der Stammzellen zu
konstruieren und ein Selektionsverfahren für die transfizierten
Zellen zu entwickeln.
Die Erfindung wird mit modifizierten embryonalen Stammzellen
gemäß Anspruch 1 bis 4, den Vektorsystemen gemäß Anspruch 5 und
6 und den Selektionsverfahren gemäß Anspruch 7 realisiert. Zum
Schutzumfang der Erfindung gehört auch die Verwendung der
modifizierten embryonalen Stammzellen gemäß Anspruch 8 bis 10.
Die erfindungsgemäßen Vektoren bestehen aus folgenden Bestand
teilen:
- a) der regulatorischen, 2,1 kb langen DNA-Sequenz des ventrikel spezifischen Myosin-Leicht-Kette-2 (MLC-2v) als Promotor, dem selektionierbaren Markergen β-Galaktosidase und dem Reportergen Neomycin als Fusionsgen "βgeo" und dem SV40-PolyA-Tail(pAA) und ggf. einer Position zur Aufnahme von immortalisierenden Genen.
- b) der regulatorischen DNA-Sequenz des Herpes-simplex-Virus Thymidinkinase-Promotors (HSV-Tk), dem selektionierbaren Marker gen Hygromycin und dem SV40-PolyA-Tail (pAA).
Mit diesen Vektoren werden pluripotente embryonale Stammzellen
in vitro transfiziert. Die erfolgreich transfizierten Zellen
werden im ersten Schritt mit Hilfe von Hygromycin selektiert.
Hygromycin-resistente Zellen werden dann zu sog. Embryoidkörpern
("embryoid-bodies") differenziert. Danach erfolgt eine Selektion
der Hygromycin-resistenten Embryoidkörper mit dem Zellgift
Geneticin (G418). Die erhaltenen Zellen werden weitergezüchtet
und auf ihre Zusammensetzung (Genexpression, Proteine), ihre
Funktion und ihre kontraktilen Eigenschaften untersucht.
Als Ausgangsmaterial können embryonale Stammzellen (ESC) oder
primordiale Keimzellen (EGC) unterschiedlichster Herkunft, u. a.
von Maus, Ratte, Schwein, Rind, Hund, Kaninchen, Hamster,
einschließlich menschlicher Zellen, eingesetzt werden.
Die erfindungsgemäßen Vektoren und der Ablauf des Zell
selektionsverfahrens sind in Abb. 1 dargestellt.
Weiterer Gegenstand der Erfindung sind Zellen, die neben den
genannten Vektoren a) und b) auch noch therapeutische Gene wie
z. B. die Angiogenesefaktoren VEGF oder bFGF enthalten, welche
durch viralen oder nichtviralen Gentransfer eingebracht wurden.
Die so erhaltenen Zellinien können - mit oder ohne virale
Sequenzen - zur zellvermittelten Gentransplantation, insbeson
dere zum Aufbau gesunden Gewebes und zur Unterstützung
kontraktiler Funktionen, verwendet werden.
Eine weitere wichtige Verwendung der erfindungsgemäßen Zellinien
besteht in der in-vitro-Testung von biologisch aktiven
Substanzen, insbesondere zur Untersuchung pharmakologisch
relevanter Substanzen oder zur Feststellung toxischer Wirkungen
exogener Wirkstoffe an Herzzellen in Kultur. Damit werden
insbesondere in Screeningprogrammen Tierversuche eingespart und
damit dringende Forderungen der Öffentlichkeit nach
Tierersatzmodellen erfüllt.
Die Zellinien können weiterhin als Vesikel für einen lokalen
Gentransfer in das Myokard dienen. Dazu werden die gewünschten
therapeutischen Gene durch ein virales, bevorzugt mit einem
Adenovirus- oder einem Adenovirus-assoziierten-Virus-Shuttle-
Vektor, oder ein nichtvirales Gentransferverfahren transfiziert.
Bevorzugt erfolgt die Verpackung der Gene mit dem AAV-Vektor
pSub201.
Durch die Erfindung wird erstmalig ermöglicht, Herzmuskel
erkrankungen mit Hilfe des zellulären Gentransfers zu behandeln
(Gentherapie). Das bedeutet einen erheblichen medizinischen
Fortschritt, insbesondere können auch Erkrankungen wie
ischämische und kongenitale Kardiomyopathien künftig mit
größeren Erfolgsaussichten therapiert werden.
Die Erfindung soll nachfolgend durch Ausführungsbeispiele näher
erläutert werden.
Zum Aufbau der Vektoren geht man von folgenden Elementen aus:
2.1 kb MLC-2v Promotor (klonierbar mit KpnI und EcoRI), Fusionsgen βgeo (klonierbar mit BamHI), SV40-PolyA (klonierbar mit SacI) und Tk-Hygromycin Fusionsgen im Blueskript KS-Vektor. Wie in Abb. 1 dargestellt, werden aus diesen Elementen 2 Vektoren für die Kotransfektion in pluripotente embryonale Stammzellen erstellt:
2.1 kb MLC-2v Promotor (klonierbar mit KpnI und EcoRI), Fusionsgen βgeo (klonierbar mit BamHI), SV40-PolyA (klonierbar mit SacI) und Tk-Hygromycin Fusionsgen im Blueskript KS-Vektor. Wie in Abb. 1 dargestellt, werden aus diesen Elementen 2 Vektoren für die Kotransfektion in pluripotente embryonale Stammzellen erstellt:
- (A) 2,1 MLC-2v-βgeo
- (B) Tk-Hygromycin.
Als Pluripotente Stammzellinie kann jede ES-Zellinie verwendet
werden, die in Kardiomyozyten differenziert (Wobus et al,
Differentiation 48/1991/, 173-182), z. B. die Linie D3
(Doetschmann et al, J. Embryol. Exp. Morphol. 3/1985/, 27-45).
Die D3-Zellen werden auf gelatinierten Platten mit
standardisiertem Kulturmedium auf feeder-layer oder in
Anwesenheit des rekombinanten "Leukemia-Inhibiting-Factor"(LIF)
kultiviert. Der LIF entspricht dem "Differentiation Inhibiting
Factor", welcher die Differenzierung der ES-Zellen verhindert
und die Zellteilung der pluripotenten ES-Zellen fördert. Die in
Abb. 1 dargestellten DNA-Konstrukte werden durch Elektro
poration in die ES-Zellen eingebracht. Hierfür werden die
Vektoren mittels Restriktionsverdau linearisiert und in einer
Konzentration von 25 µg/ml mittels Elektroporation transfiziert.
Danach werden die Pluripotenten ES-Zellinien im LIF/ES-
Zellmedium expandiert. In den undifferenzierten ES-Zellen ist
nur der Thymidinkinase-Promotor aktiv, was zu einer Expression
des Hygromycin-Resistenzgens führt. Durch Zugabe von Hygromycin
B werden die ES-Zellen auf den Einbau der Fremd-DNA hin
selektioniert (Positiv-Selektion).
Zur Gewinnung von "embryoid bodies" wird das Differenzierungs
system des hängenden Tropfens benutzt. Hierbei wird eine
Zellsuspension, die etwa 400-600 ES-Zellen in 20 µl enthält, auf
die Deckel von Petrischalen pipettiert, die mit einer
physiologischen Pufferlösung gefüllt sind. Die Zellen sammeln
sich im Tropfen und bilden nach zwei- bis dreitägiger Inkubation
"embryoid bodies". Nach 7 Tagen werden die "embryoid bodies" auf
Mikrotestgewebekulturschalen übertragen, wo sie auf dem
Gelatine-beschichteten Substrat anhaften. Während der darauf
folgenden Kultivierung wachsen verschiedene Zelltypen, u. a.
Herzmuskelzellen, aus (Abb. 2). Zwei bis zehn Tage nach
Ausplattierung enthalten etwa 80 bis 90% der ausgewachsenen
"embryoid bodies" Kolonien spontan und synchron kontrahierender
Herzmuskelzellen (Wobus et al, 1991). In diesen embryonalen
Herzmuskelzellen ist der MLC-2v Promotor aktiv. Bei
erfolgreicher Transfektion und Integration des MLC-2v-βgeo
Vektors in das Genom der ES-Zellen kommt es zur Expression des
Fusionsgens β-Galaktosidase/Neomycin. Durch Blaufärbung im β-
Galaktosidase-Assay lassen sich die kardiomyozytären Zellen,
welche klonalen Ursprungs sind, identifizieren.
In einem zweiten Schritt werden die positiven ES-Zellklone,
welche sowohl das Tk-Hygromycin als auch das MLC-2v-βgeo
Fusionsgen enthalten, erneut expandiert und ein Teil unter den
oben beschriebenen Bedingungen zur Differenzierung in "embryoid
bodies" gebracht. Zum Zeitpunkt der kardiomyogenen
Differenzierung im Embryoidkörper wird das Zellgift Geneticin
(G418) in einer Konzentrationen von 300 µg/ml zu den "embryoid
bodies" gegeben. Damit werden die embryonalen (kardiomyozytären
bzw. kardiomyoblastären) Herzmuskelzellen in einem frühen
Stadium selektiert.
Die gewonnenen Zellen werden auf gewebespezifische
Genexpression, funktionelle Eigenschaften mit Hilfe
elektrophysiologischer Techniken und auf ihre kontraktilen
Eigenschaften untersucht, die Zusammensetzung der kontraktilen
Proteine wird mit entsprechenden monoklonalen Antikörpern
charakterisiert. Im Rahmen einer Gentherapie können
teilungsfähige schlagende Zellen anschließend auf adulte und
neonatale kardiomyopathische mdx-Mäuse nach Thorakomie und
direkter Injektion in das Myokard übertragen werden.
Claims (11)
1. Embryonale Herzmuskelzellen,
enthaltend zwei Genkonstrukte aus zwei verschiedenen
regulatorischen DNA-Sequenzen und zwei verschiedenen
selektionierbaren Markergenen.
2. Embryonale Herzmuskelzellen gemäß Anspruch 1,
enthaltend zwei Genkonstrukte aus
- a) regulatorischer, 2,1 kb langer DNA-Sequenz des Ventrikel spezifischen Myosin-Leicht-Kette-2 (MLC-2v) Promotors, dem selek tionierbaren Markergen β-Galaktosidase und dem Reportergen Neomycin,
- b) regulatorischer DNA-Sequenz des Herpes-simplex-Virus-Thymi dinkinase-Promotors und dem selektionierbaren Markergen Hygromycin.
3. Embryonale Herzmuskelzellen gemäß Anspruch 1 und 2,
enthaltend zusätzlich immortalisierende Gene und/oder durch
homologe Rekombination inaktivierte Gene und/oder ein oder
mehrere therapeutische Gene.
4. Embryonale Herzmuskelzellen gemäß Anspruch 1 bis 3,
enthaltend zusätzlich Sequenzen des Adenovirus (Ad) oder des
Adenoassoziierten Virus (AAV).
5. Embryonale Herzmuskelzellen gemäß Anspruch 1 bis 4,
enthaltend zusätzlich einen Angiogenesefaktor, bevorzugt
"vascular endothelial growth factorll, VEGF, oder "basic
fibroblast growth factor", bFGkF, unter der Kontrolle eines in
diesen Zellen aktiven Promotors als therapeutisches Gen.
6. Vektor, bestehend aus MLC-2k Promotor, selektionierbarem
Markergen β-Galactosidase und dem Reportergen Neomycin als
Fusionsgen "βgeo", SV40-Poly A-Tail und ggf. einer Position zur
Aufnahme von immortalisierenden Genen.
7. Vektor, bestehend aus regulatorischer DNA-Sequenz des Herpes
simplex-Virus-Thimidinkinase-Promotors und dem selektio
nierbaren Markergen Hygromycin.
8. Verfahren zur Herstellung von Zellen nach Anspruch 1 bis 5,
dadurch gekennzeichnet, daß embryonale Stammzellen (ESC) oder
primordiale Keimzellen (EGC) mit den Vektoren nach Anspruch 6
und 7 kotransfiziert werden, die Selektion der embryonalen
Stammzellen mit dem Zellgift Hygromycin erfolgt, nach Induktion
der in vitro Kardiogenese anschließend die Selektion der
embryonalen Herzzellen mit dem Zellgift Geneticin (G418)
erfolgt, die erhaltenen Zellen ggf. mit den gewünschten
therapeutischen Genen durch ein virales, bevorzugt mit einem
Adenovirus- oder einem Adenovirus-assoziierten-Virus-Shuttle-
Vektor, oder ein nichtvirales Gen-transferverfahren transfiziert
werden.
9. Verwendung der embryonalen Herzmuskelzellen nach Anspruch 1
bis 4 zur zellvermittelten Gentransplantation, insbesondere zum
Aufbau gesunden Gewebes und zur Unterstützung kontraktiler
Funktionen.
10. Verwendung der embryonalen Herzmuskelzellen nach Anspruch 1
bis 4 zur Untersuchung von Substanzen, insbesondere für
pharmakotoxikologische Untersuchungen.
11. Verwendung der embryonalen Herzmuskelzellen nach Anspruch 1
bis 5 für den Transfer therapeutischer Gene in das Myokard.
