Description
The present invention relates to the production of intermediates in carotenoid synthesis, namely the production of 3-hydroxy-2-cycloalken-1-one derivatives.

kathodisch weiterreduziert. The process according to the invention is characterized in that a compound of the general formula
to a compound of the general formula
cathodically reduced and, if desired, the compound obtained in an acid or neutral medium to a compound of the general formula
further reduced cathodically.
From European Patent Publication No. 0 031 875, the reduction of the compound of formula I above, wherein n is 1, to the corresponding compound of formula III by means of zinc and a carboxylic acid is known. However, this process has the disadvantage that an excess of zinc has to be used and therefore large amounts of zinc salts are obtained.
It has now been found that the process according to the invention is not associated with the disadvantages of the previously known process and nevertheless makes it possible to obtain the desired products in good yield. Since the potentials required for reducing the compounds of the formulas I and II are sufficiently different, either compounds of the formula II or compounds of the formula III can be obtained selectively by this process. The process according to the invention can also be easily carried out as a continuous process.
In the literature, the tautomeric 1,3-diketone is sometimes also mentioned for such compounds. However, it has been shown that the compounds of the formulas II and III are virtually exclusively in the above ketonol forms.
Diaphragma-Materialien, beispielsweise Ton, Keramik, Glas (z. B. Glassinterdiaphragma) oder polymeren Verbindungen (z. B.. NAFIONO [Dupont]) erfolgen kann. The method according to the invention can be carried out in undivided or divided cells. However, a subdivided cell is preferably used, the subdivision using membranes or diaphragms made of conventional membrane or diaphragm materials, for example clay, ceramic, glass (for example glass sintered diaphragm) or polymeric compounds (for example .. NAFION O [ Dupont]) can be done.
The electrodes can have customary shapes. For example, the electrodes can be designed in the form of plates or grids or as expanded metal.
Preferred cathode materials are mercury (for example as a mercury sump electrode), lead and graphite. Depending on the reaction conditions, however, other cathode materials are also possible, for example in strongly acidic solutions cadmium, tin, aluminum or lead or steel or copper networks, in weakly acidic or neutral solutions cadmium, zinc or tin and in basic solutions nickel. Furthermore, nickel, copper, steel, vanadium, silver, cobalt, brass and the like can also be used for the reduction of a compound of formula I to the corresponding compound of formula II in acidic solution.
The anode materials used in the method according to the invention are not critical. Suitable anode materials are, for example, platinum, palladium, silver, gold, graphite or lead. Furthermore, dimensionally stable anodes [such. B. in A. Schmidt, Angewandte Elektrochemie, p. 70, Verlag Chemie (1976) mentioned], also called metal oxide composite anodes, can be used. Such anodes consist of a carrier made of titanium, iron, nickel and the like, which is provided with a metal oxide coating (e.g. lead dioxide, manganese dioxide, ruthenium dioxide or titanium suboxide), an intermediate layer of a carbide or. On the metal surface prior to the application of the metal oxide coating Boride of elements of subgroups IV and V. is applied.
When using an acidic or neutral reaction medium, the electrolysis according to the invention is expediently carried out using a mixture of an electrolyte, an inert organic solvent (cosolvent) and the compound of the formula used as starting material. As the electrolyte, for example, aqueous acids such as sulfuric acid, hydrofluoric acid, hydrochloric acid, phosphoric acid, perchloric acid, aqueous trifluoroacetic acid and the like can be used. Mineral acids, in particular sulfuric acid, have proven to be particularly suitable. It is expedient to work with about 0.1 N to about 18N, preferably about 1 N to about 10N, aqueous acids.
Water-miscible or non-water-miscible inert organic solvents are suitable as co-solvents. When using a water-immiscible cosolvent, such as 1,2-dichloroethane or carbon tetrachloride, a phase transfer catalyst (preferably a tetraalkylammonium salt, for example tetrabutylammonium hydrogen sulfate) is expediently added. However, a water-miscible cosolvent is preferably used, for example an alcohol such as methanol, ethanol or t-butanol, a cyclic ether such as tetrahydrofuran or 1,4-dioxane, a nitrile such as acetonitrile, an amide such as N, N-dimethylformamide or Hexamethylphosphoric triamide, a carboxylic acid such as formic acid or acetic acid, or propylene carbonate, dimethyl sulfoxide or acetone. Preferred such cosolvents are methanol, ethanol, t-butanol, tetrahydrofuran and 1,4-dioxane.
If a carboxylic acid is used as the cosolvent, an aqueous solution of a carboxylate (for example sodium formate, sodium acetate) can also be used as the electrolyte. On the other hand, it is also possible to use an aqueous solution of a carboxylic acid, such as formic acid, as the electrolyte. To improve the conductivity, a conductive salt (for example sodium formate) is expediently added and an alcohol such as methanol, for example, is used as the cosolvent.
The volume ratio of electrolyte / cosolvent is expediently about 5: 1 to about 1: 5 and preferably about 1: 1 when using a water-miscible co-solvent. On the other hand, when using a water-immiscible co-solvent, only as much co-solvent is generally added as is necessary to ensure a sufficient To ensure solubility of the educt and the product.
Furthermore, an aqueous solution of a base, for example about 0.1N ― 5N sodium hydroxide solution or potassium hydroxide solution, can also be used as the electrolyte. Since the educt and product are sufficiently soluble in such electrolytes, the use of a cosolvent is not necessary. However, the potentials required for reduction are more negative in basic reaction media than in acidic or neutral media. While the reduction of the compounds of the formula I to compounds of the formula II in basic solutions is still possible without problems, it is generally no longer possible to further reduce the compounds of the formula II to compounds of the formula III, since the negative potentials required for this already exist Decomposition of the solvent occurs.
The reduction of the compounds of formula II is therefore advantageously carried out in an acidic or neutral reaction medium. However, such reaction media are also preferred for the inventive reduction of the compounds of the formula I to compounds of the formula II. Acidic reaction media are particularly preferred.
I.In the case of reduction of the compound of the formula II, in which n is zero, it has proven expedient to use a phase transfer catalyst, for example a tetraalkylammonium salt, such as tetramethylammonium tetrafluoroborate, tetraethylammonium tosylate, tetraethylammonium perchlorate, tetrabutylammonium tetraammonium tetrammonium tetrammonium tetrammonium tetrammonium tetramate, Add hexadecyl-trimethylammonium bromide or preferably tetrabutylammonium bromide to the reaction mixture. The concentration of the phase transfer catalyst, based on the total mixture, is preferably about 0.05 mol / l to about 0.8 mol / l.
The concentration (w / v) of the starting material in the reaction mixture used can, in the case of the compounds of the formulas 1 and II, in which n = 0, generally between about 0.3% and 15%, preferably about 5% and 15% vary. For the compounds of the formulas I and II, in which n = 1, it is advantageously about 0.3% to about 10% and preferably about 5%.
The temperature used in carrying out the method according to the invention is not critical. However, its upper limit is limited by the boiling point of the reaction mixture. The process is preferably carried out between room temperature and about 65 ° C. Das erfindungsgemäße Verfahren kann galvanostatisch oder potentiostatisch durchgeführt werden. Das potentiostatische Verfahren ist bevorzugt. The method according to the invention can be carried out galvanostatically or potentiostatically. The potentiostatic method is preferred.
- a) For the reduction of the compound of formula I, where n =, to the corresponding compound of formula II, when using a lead cathode in 10N sulfuric acid / dioxane 1: 1, a potential of about -750 mV (against saturated calomel electrode SCE) and when using a nickel cathode in 2N sodium hydroxide solution a potential of about -1300 mV (against SCE) was applied.
- b) For the reduction of the compound of formula I or 11, in which n = 1, to the corresponding compound of formula III, when using a lead cathode in 1 N sulfuric acid / ethanol 1: 1, a potential of approximately -1200 mV (against SCE) applied.
- c) For the reduction of the compound of formula I, where n = 0, to the corresponding compound of formula II, when using a mercury sump cathode in 3N sulfuric acid / dioxane 1: 1, a potential of about -650 mV (against saturated silver / silver chloride) is expedient -Electrode SSE) and in 3N sodium hydroxide solution a potential of about - 1500 mV (against SCE) was applied.
- d) For the reduction of the compound of the formula I or 11, in which n = 0, to the corresponding compound of the formula III, when using a mercury sump cathode in 3N sulfuric acid / dioxane 1: 1 and adding tetrabutylammonium hydrogen sulfate, a potential of approximately -1200 is expedient mV (against SSE) applied.
The current density used in carrying out the method according to the invention is not critical. In general, it can vary between about 4 and 70 mA / cm 2 for the 5-ring connections (n = 0) and between about 4 and 40 mA / cm 2 for the 6-ring connections (n = 1). The preferred range is about 5-15 mA / cm 2.
Voltage and current intensity depend on the reaction medium used, the size of the cell, the current density used and the like.

It is then left to stand at -20 ° C. overnight, filtered off with suction and washed twice with 20 ml of diisopropyl ether (cooled to -20 ° C.) and dried to constant weight at 50 ° C. in a water jet vacuum. 9.6 g (70%) of a product with a melting point of 170 ° C., which is a tautomer mixture of 3-hydroxy-2,5,5-trimethyl-2-cyclopenten-1-one and 3-hydroxy-2, 4,4-trimethyl-2-cyclopenten-1-one.
a) Als Reaktionsgefäß dient ein zweiteiliges Glasgefäß (H-Zelle). Man erhält 17,6 g weiße Kristalle. The anode compartment is separated from the cathode compartment by a round glass sintered diaphragm (diameter: 3.5 cm). In the cathode compartment there is a mercury sump electrode (surface: approx. 28 cm 2 ) and a saturated silver / silver chloride reference electrode (SSE). A lead sheet (3.5 cm x 3.5 cm) is used as the anode. Both electrode compartments are each equipped with a gas inlet tube and the cathode compartment with a magnetic stirrer. While stirring and introducing nitrogen gas, 160 ml of dioxane and 3N aqueous sulfuric acid are added to the cathode compartment and 90 ml of dioxane and 3N aqueous sulfuric acid are added to the anode compartment. After 15.4 g of the initially incompletely dissolved 4-hydroxy-2,2,5-trimethyl-4-cyclopentene-1,3-dione have been added to the cathode compartment, a cathode potential of -650 mV (against SSE) electrolyzed at room temperature while stirring and introducing nitrogen, whereupon a current of approx. 200 mA is established. After the current has dropped to 5 mA and 19,300 coulombs, which corresponds to 100% of the theoretically required amount of current, have flowed through the cell, the electrolysis is stopped. The combined catholyte and anolyte solutions are placed in a 2 liter separating funnel S i with 500 ml of saturated saline. Two further 1 liter separating funnels S 2 and Ss are charged with 200 ml saline solution. One portion of 300 ml and nine portions of 100 ml of methylene chloride are passed through the three separating funnels S 1 ―S 3 . The organic phases are dried over 50 g of sodium sulfate and filtered. The combined filtrates are concentrated to constant weight in a rotary evaporator in a water jet vacuum at 40 ° C. bath temperature. 17.6 g of white crystals are obtained. The crude product is dissolved hot in a mixture consisting of 50 ml of diisopropyl ether and acetone. After cooling to room temperature, the first white crystals precipitate out. Then it is left overnight at ―20 ° C, suction filtered and washed twice with 20 ml of diisopropyl ether (cooled to -20 ° C) and dried at 50 ° C in a water jet vacuum to constant weight. 9.5 g (61%) of a product with a melting point of 147-148 ° C., which contains a tautomer mixture of 3,4-dihydroxy-2,5,5-trimethyl-2-cyclopenten-1-one and 3, 5-Dihydroxy-2,4,4-trimethyl-2-cyclopenten-1-one.
Nach der Umkristallisation aus einem Gemisch, bestehend aus je 50 ml Diisopropyläther und Aceton, werden 9,9 g (71%) eines Produkts mit einem Schmelzpunkt von 169-179°C erhalten, welches ein Tautomerengemisch von 3-Hydroxy-2,5,5-trimethyl-2-cyclopenten-1-on und 3-Hydroxy-2,4,4-trimethyl-2-cyclopenten-1-on darstellt.b) In addition to 45 ml of dioxane and 3N aqueous sulfuric acid and 3.05 g of tetrabutylammonium hydrogen sulfate, 15.6 g of the product obtained in paragraph a) are added to the cathode compartment for further reduction at a cathode potential of -1200 mV (against SSE). The anode compartment, each with 50 ml of dioxane and 3N aqueous sulfuric acid and a platinum wire anode, is separated from the cathode compartment by a round polymer membrane (diameter: 4 cm). The other test conditions are not changed. After the current has dropped from initially 150 mA to 70 mA and 27 100 coulombs, which corresponds to 140% of the theoretically required amount of current, have flowed through the cell, the electrolysis is stopped. The catholyte solution is placed in a 500 ml separating funnel Si with 150 ml of saturated saline. Two further 500 ml separating funnels S 2 and Ss are each charged with 150 ml of saturated saline. Through the three separating funnels S 1 ―S 3 , nine portions of 150 ml methylene chloride are sent in sequence. The organic phases are dried over 50 g of sodium sulfate and filtered. The combined filtrates are concentrated to constant weight in a rotary evaporator in a water jet vacuum at 40 ° C. bath temperature. 14.9 g of a yellow crystalline residue are obtained which, after purification (removal of the polar tetrabutylammonium hydrogen sulfate) on a chromatography column (silica gel / diethyl ether), gave 11.9 g of white-yellow crystals. After recrystallization from a mixture consisting of 50 ml of diisopropyl ether and acetone, 9.9 g (71%) of a product with a melting point of 169-179 ° C. are obtained, which has a tautomer mixture of 3-hydroxy-2,5,5-trimethyl-2-cyclopenten-1-one and 3-hydroxy-2,4,4-trimethyl-2-cyclopenten-1-one.
The anode compartment is separated from the cathode compartment by a round glass sintered diaphragm (diameter: 2.5 cm). A graphite electrode (3.5 cm x 3.5 cm) and a saturated silver / silver chloride reference electrode (SSE) are located in the cathode compartment. A titanium anode coated with lead dioxide is used as the anode. Both electrode compartments are each equipped with a gas inlet tube and the cathode compartment is also equipped with a magnetic stirrer. While stirring and introducing argon gas, 35 ml each of ethanol and 1N aqueous sulfuric acid are added to the cathode compartment and 35 ml each of ethanol and 1 N aqueous sulfuric acid are added to the anode compartment. After 0.7 g of 2-hydroxy-3,5,5-trimethyl-2-cyclohexene-1,4-dione have been added to the cathode compartment, the mixture is stirred at a constant cathode potential of -1250 mV (against SSE) and argon gas is introduced electrolyzed at room temperature, whereupon a current of approx. 306 mA is established. After 2600 coulombs, which corresponds to 162% of the theoretically required amount of electricity, have flowed through the cell, the electrolysis is stopped. The catholyte solution is extracted five times with 50 ml of chloroform. The combined extracts are dried over sodium sulfate, filtered and concentrated. According to gas chromatographic analysis, the concentrated solution contains 0.45 g (70%) of the tautomer mixture of 3-hydroxy-2,6,6-trimethyl-2-cyclohexen-1-one and 3-hydroxy-2,4,4-trimethy) -2-cyc) ohexen-1-one and 0.022 g (3%) of the tautomer mixture of 3,4-dihydroxy-2,6,6-trimethyl-2-cyclohexen-1-one and 3,6-dihydroxy-2, 4,4-trimethyl-2-cyclohexen-1-one.
