明 細 書 . 発明の名称 酵素センサ 技術分野 本発明は、 酵素セ ンサ、 特にポテン シ ョメ ト リ ックの応答で 生体基質の濃度を測定する酵素センサに関する。 背景技術 従来、 グルコース, 尿素. 尿酸などの酵素セ ンサが知られて いる。 それらは酵素反応で生成する過酸化水素 ( H 2 0 2 )濃度 あるいは反応で消費される酸素 ( 0 2 )濃度を酸素セ ンサや過酸 化水素センサを用いて電流法で測定したのち、 基質濃度を求め る酵素セ ンサである。 こ のため、 一般に小型化が困難であつ た。 また、 酸素の消費や過酸化水素の生成を伴わない酵素反応 には利用できないという欠点があった。 この問題の解決法と し て酵素反応の進行に伴う P H変化を測定して基質濃度を求める セ ンサがある。
最近は、 I S F E T (ィ才ン選択性電-界効果ト ラ ン ジスタ ) の p Hセンサを利用して、 小型の酵素センサを作る試みがなざ
れているが、 I S F E Tのゲー卜絶縁膜 (例 : S i 3 N 4 膜、 A 1 2 0 3 膜、 T a 2 0 5 膜など) の表面と酵素膜との間の被 着性が悪く、 その結果、 感度がやや低い、 ド リ フ トが大きい、 寿命が短い等の欠点があった。
5 発日月の開示 本発明の巨的は、 小型で生体の基賞濃度をボテンショメ ト リ ヅクに測定する酵素センサであって、 高感度でドリ フ トが小さ く、 寿命の長い酵素センサを提供するこ とにある。
. この問題点を解決するための一手段と して、 本発明の酵素セ 0 ンサは、 絶縁性基板と、 該絶縁性基板上に被覆された導電性層 と、 該導電性層の表面に被覆された酸化還元機能を有する酸化 ' 還元機能層と、 該酸化還元機能層の表面に被覆された酵素が固 定化された酵素固定化層とを傭える。
又、 M 0 S F E T と、 該 M 0 S F E Tのゲート絶縁膜上に被 5 覆された導電性層と、 該導電性層の表面に被覆された酸化還元 機能を有する酸化還元機能層と、 該酸化還元機能層の表面に被 ' 覆された酵素が固定化された酵素固定化層とを備える。
かかる構成において、 酵素固定化層によって生体基 Kの濃度 を対応する水素イオン濃度に変換し、 酸化還元機能層により水 0 素ィォ-ン濃度を対応する電界に変換する。 発生した電界は導電 性層により基準極との電位差として測定される。
又、 発生した電界は導電性層により M 0 S F E Tのゲート絶
緑膜上に伝達されて、 M O S F E Tによって測定される。
本-発明により、 小型で生体の基質濃度をポテンショメ 卜 リ ツ クに測定する酵素セ ンサであって、 高感度で ド リ フ トが小ざ く、 寿命の長い酵素センサを提供できる。
更に詳細には、 本発明の酵素センサは、
( 1 ) I S F E . Tゲー ト部に構成されるので、 微小化および 酵素マルチ化センサを提供できる。
( 2 ) 酸化還元機能層を P H感応膜と して用いるので感度が高 レ 0
( 3 ) 電解重合法を用いて膜形成を行う ため、 被着性がよく導 電性基材の凹凸部にも固着性がよく膜形成ができるため、 耐久 性が良く長寿命である。
( 4 ) 電解重合法を用いて酵素を固定化するため酵素の固定化 が容易であり、 被着性が良く耐久性の良い酵素固定化膜が得ら れる。.
( 5 ) ポテンショメ ト リ ックな測定を行うため、 酸素の関与し ない酵素反応を利用した酵素センサを構成するこ とができる。
( 6 ) ポテンショメ ト リ ックな測定を行うため、 生体や測定系 の電気的なリークが少ないため安全である。
( 7 ) 電解重合法による膜形成のため、 膜厚の制御が容易であ る。 図面の簡単な説明

第 3図は実施例 2の酵素センサによる測定結果を示す図、 第 4図は実施例 3の酵素センサによる測定結果を示す図、 第 5図は比较例 1 の酵素センサによる 定結果を示す図、 第 6図 ( a ) , ( b ) は実施例 4の F E Tを利用する酵素セ ンサの断面模式図である。 発曰且を実施するための最良の形態 以下、 図面を参照しながら末癸明の実施例を詳細に説明す る。 . 本実施例でほ、 導電性層 (酸化還元反応発現する酸化イ リ ジ ゥム、 I T 0 , 白金, パラジウムオキサイ ド, 銀など) を絶縁 性基板 (サファイ ア, ダイヤモン ド, S i 0 2 , S i 3 N 4 , Τ a 2 0 5 など) 上に被着した基体の上に酸化還元機能層を直 接被覆 (例 :電解重合法) し、 その上に酵素固定化層 (従来の 酵素固定化法、 または酵素固定化膜を固定する反応膜 : 例 : 1 , 2ジァミ ンベンゼン, ピロール共存中で、 電解反応法を利 用して酵素固定化反応を行うこ とによ り作成) を被覆した酵素 電極を作成し、 この酵素電極を作甩極として基準電極 (飽和ナ ト リ ウム力 σメル電極) との電位差を測定して上記構成の層
に よ る電気的特性と層の物理的特性等をテス ト した結果、 I S F E T上に酵素固定化層を塗布して酵素センサと して使用 した場合の問題点 (ゲー ト絶縁膜と酵素固定化層との間の 剝離, 感度低下, ド リ フ トが大きい, 寿命が短い等の諸問題) を解決出来た。 その代表的特性であるネルンス ト式の傾きが 5 8 raVZpH以上 ( 3 2 : : 理論的 6 0 . 5 4 6 mV/pH) であり 理論式に近似している。 又、 1 , 2ジァミノベンゼン併用時の 酵素固定化では 7 3 . S 7 mV/pHと高感度である。 これらに比 ベ酸化重合膜 (酸化還元機能層) 無しに直接導電性層 (例 : 酸 化イ リ ジウム) 上に酵素を被覆して作製した酵素センサのそれ では 3 5. 4 0 raVZ p Hで感度が非常に低い。
(実施例 1 )
本実施例で作製 し た酵素セ ン サの構成摸式図を第 1 図 ( a ) , ( b ) に示す。
( 1 ) 酸化ィ リ ジゥム電極
2 0 m m x l 8 m m , 厚さ 1 . 5 m mのサファイア基板 1上 に、 3 m m X 1 2 m m , 厚さ 1000Aとなる酸化イ リ ジウム層 2 を 3 . 2 m mの間隔で 3 ケ所にスパッタ蒸着した。 この酸化ィ リ ジゥム層 2の末端から 3 m mの所に導電性接着剤 5 (サイコ ロン B、 厚木中央研究所 (株) 製) でリー ド線 4を接続した。 次にその接続部の上にエポキシ系接着剤 3 (ァラルダイ ド、 チ バガイギーネ土製) を被覆し、 外部と絶縁した。 また、 隣接した 酸化ィ リ ジゥム層の間にも同接着剤で壁 3 aを作り、 相互の影 響を防止した。 これを酸化イ リ ジウム電極とする。
( 2 ) 酸化還元機能層
上記酸化ィ リジゥム電極表面に電解重合法により酸化還元機 能層 6を被着した。 電解は酸化イ リ ジウム電極を作用極、 銀 / 塩化銀電極を基準極、 白金巻線を対極とする 3電極セルを用い て行った。

温度— 2 0 、 窒素雰囲気で 0 Vから + 1 . 5 V (対 A g /A g C 1 ) まで 3回電位掃引 (掃 引速度 5 0 mV/sec ) した後、 + 1 . 5 Vで 1 0 分間定電位電解を行い、 ポリ ( 2 , 6 —ジ メチルフエノ一ル) 膜 (約 3 Ο μ πι層) を形 成した。
( 3 ) 酵素固定化層
上記酸化還元機能層 S上に酵素固定化層 7 として下記に示す 手順でグルコースォキシダーゼ膜を被着した。 該膜の被着はグ ルタルアルデヒ ドを架撟剤とする架橋法によった。
( 液) 11 8 . 0 4 リ ン酸塩緩衝液に 1 5重量パーセン ト と なる牛血漿アルブミンを溶かし、 さらにその溶液 5 m l にグルコースォキシダーゼ 0 . 5 gを溶かす。
( B液) 2 5パーセン ト グルタルアルデヒ ド水溶液
( C液) 1 0 ノぺーセン ト グリ シン水溶液
酸化ィ リ ジゥム電極 1個につき約 S i 1 の A液をマイクロシ リ ンジを用いて酸化還元機能層 6上にのせ約 1分間乾燥した。 さらに、 同量の B液を滴下し 1分間乾燥させた後、 C液に約 1 分間浸すこ と によって未反応のグルコースォキシダーゼを取り 除き、 グルコース才キシダーゼ層を酵素固定化層 7 と して被着 した。
(実施例 2 )
( 1 ) 酸化ィ リ ジゥム電極
実施例 1 と同様の酸化ィ リ ジゥム電極を作成した。
( 2 ) 酸化還元機能層
実施例 1 と同様な方法で、 酸化ィ リ ジゥム電極表面上に酸化 還元機能層 5を被着した。
( 3 ) 酵素層
以下に述べる方法により、 1 , 2 —ジァミノベンゼンの電解 重合膜中にグルコースォキシダーゼを取り込み、 酸化還元機能 層 6上に酵素固定化層 7を被着した。
グルコース才キシダーゼを含む 1 , 2 —ジァミ ノベンゼン水 溶液中で、 上記酸化還元機能層 6を被 Sした酸化イ リ ジウム電 極を作用極、 銀 塩化銀電極を基準極、 白金巻線を対極と して 電解重合を行う と、 1 , 2 —ジァミノベンゼンの電解重合膜が 形成されると同時に、 共存するグルコースォキシダーゼが該膜 中に取り込まれ、 ダルコースォキシダーゼ膜が被着されるこ と になる。

ぐ電解重合条件〉
窒素雰囲気で 0 Vから + 1 . 5 V (対 A g ZA g C l ) まで 3回電位掃引 (掃引速度 5 0 mVZsec ) した後、 + 1 . 5 V で 3 0分間定電位電解した。
(実施例 3 )
( 1 ) 酸化ィ リジゥム電極
実施例 1 と同様の酸化イリジウム電極を作製した。
( 2 ) 酸化還元機能層
実施例 1 と同様な方法で酸化イ リ ジウム電極表面上に、 酸化 還元機能層 sを被着した。
( 3 ) 酵素固定化膜
1 , 2—ジァミノベンゼンの代わり にビロールを用い、 実施 例 2 と同様の方法により酸化還元機能層 6上に酵素固定化層 7 としてグルコースォキシダーゼ膜を被着した。
(実験例 1 )
実施例 1 で作製した酵素センサを用いて基準電極 (飽和ナ ト リ ウムカロメル電極) との間の電位差を測定する方法により、 被検液のグルコース濃度の変化に対する応答を謌ベた。 なお、 被検液の温度ほ 3 2でに設定し、 p Hはリ ン酸塩緩衝液により 6 . 8 6に調整した。 1 0 mgZdlのグルコース水溶'液に 5 0 0
nigZdlのグルコース水溶液を滴化して濃度を変え、 電位変化を 測定した。 ただし、 測定は滴下後 1 0分間経過し、 電位が安定 した時点で行った。
得られた結果を第 2図 ( a ) に示し、 その時のグルコース濃 度の対数に対する電極電位のプロッ トを第 2図 ( b ) に示す。 このよう に、 グルコース濃度の対数と酵素センサの電位との間 には良い直線関係が得られ、 その近似式は次のよう になった。

(実験例 2 )
実施例 2で作製した酵素センサを用いて、 実験例 1 と同様の 測定を行なった。 その結果を第 3図に示す。 直線の近似式は、

比較例と して、 酸化イ リ ジウム電極を直接酵素固定化層で被 覆した以外は実施例 1 と同じ酵素センサを作製し、 実験例 1 と 同様の測定を行った結果、 第 5図に示すよ う にグルコース濃度 の対数と酵素センサの電位応答との間にほ、 ほぼ直線関係が得 られるが、 その近似式ほ、

M 0 S F E T 1 1 を半導体プロセス技術を利甩して形成したの ち、 M 0 S F E T 1 1 よりやや離れた部分に酸化ィ リジゥムを 反応性スパッタ法を用いて蒸着して分離ゲート部 1 2を形成し たのち、 実施例 1 〜 3 と同様に酸化還元機能層 1 3 および酵素 固定化層 1 4を形成し、 絶縁材 1 5で分離ゲート部 1 2以外を 覆った。 このよう にして作製した酵素センサほ実験例 1〜 3 と 同様の高感度 6 0〜 7 0 mV/decadeの応答を示した。 また、 第 6図 ( b ) に示すよ う に、 M 0 S F E T 2 0 のゲート絶縁膜 2 1 ( S i 02 / S i 3 N 4 膜) の表面に導電性層 2 2 (酸化 イ リジウム等) を形成し、 更に酸化還元機能層 2 3 と酵素固定 化層 2 4を形成して、 絶緣材 2 5で覆つたものもできる。 本実 施例では、 導電性層と して酸化ィ リ ジゥムについて説明した が、 I T 0、 白金、 ノ ラジウムオキサイ ド、 銀についても同様 の結杲が得られた。
尚、 本実施例ではサフアイャ基板上に作成された酵素電極と M 0 S F E Tのゲート絶籙膜上を各層で覆った F E Tセンサに ついて説明したが、 本実施例で示された如く導電性層 ·酸化還 元機能層 ' 酵素固定化層で覆う ことにより高感度でド リフ トが 小さく、 寿命の長い酵素センサを作成でき、 生体基質の濃度に
対応して発生した電界の測定方法ほ本例に限らない
Claims
請 求 の 範 囲' ( 1 ) 絶縁性基板と、 該絶縁性基板上に被覆された導電性層 と、 該導電性層の表面に被覆された酸化還元機能を有する酸化 還元機能層と、 該酸化還元機能層の表面に被覆ざれた酵素が固 定化された酵素固定化層とを備えるこ とを特徴とする酵素セン サ。
( 2 ) 絶縁性基板は、 サファイア, ダイヤモン ド, S i 02 , S i 3 N 4 , T a 2 05 などの絶縁性を有する物質から選ばれ るこ とを特徴とする請求の範囲第 1項記載の酵素センサ。
( 3 ) 導電性層ほ、 酸化イ リ ジウム、 I T O (イ ンジウム * ス ズ酸化物) , P t , P d O , A gなどの酸化還元反応を発現す る物 Kから選ばれるこ とを特徵とする請求の範囲第 1項記載の 酵素センサ。
( ) 酸化還元機能層が水素イオン濃度に感応する層であつ て、 電解酸化重合反応によって形成されたものであるこ とを特 徵とする請求の範囲第 1項記載の酵素センサ。
( 5 ) 酵素層が電解 (酸化) 重合法によって形成されたもので あるこ とを特徴とする請求の範囲第 1項記載の酵素センサ。
( 6 ) 酵素固定化層が架撟法あるいは包括法等によって形成さ れたものであることを特徴とする請求の範囲第 1項記載の酵素 センサ。
( 7 ) M O S F E T と、 該 M O S F E Tのゲ一卜絶縁膜上に被 覆された導電性層と、 該導電性層の表面に被覆された酸化還元
機能を有する酸化還元機能層と、 該酸化還元機能層の表面に被 覆された酵素が固定化された酵素固定化層とを備えるこ とを特 徴とする酵素センサ。.
( 8 ) 導電性層は酸化ィ リ ジ ゥム、 I T 0 (イ ン ジ ウム ' スズ 酸化物) , P t , P d 0 , A gなどの酸化還元反応を発現する 物質から選ばれるこ とを特徴とする請求の範囲第 7項記載の酵 素セ ンサ。
' ( 9 ) 導電性層が F E T部よりわずかに離れて形成される分離 ゲートであるこ とを特徴とする請求の範囲第 7項記載の酵素セ ンサ。
( 1 0 ) 酸化還元機能層が水素ィォン濃度に感応する層であつ て、 電解酸化重合反応によって形成されたものであるこ とを特 徴とする請求の範囲第 7項記載の酵素センサ。
( 1 1 ) 酵素固定化層が電解 (酸化) 重合法によって形成され たものである .こ とを特徴とする請求の範囲第 7項記載の酵素セ ンサ。
( 1 2 ) 酵素固定化層が架撟法あるいは包括法等によって形成 されたものである こ とを特徴とする請求の範囲第 7項記載の酵 素セ ンサ。
