Abstract

A method for promoting hair growth by topically applying to the scalp of a subject a hair growth enhancing agent which includes at least one benzoic acid ester of primary alcohols having from 12 to 15 carbon atoms. Pharmaceutical kits containing a composition including the hair growth enhancing agent are also contemplated.

Description

Pharmaceutical composition containing benzole acid ester and kit
Benzoic acid esters of primary alcohols are known to be useful as diluents, solvents, plasticizers, liquid carriers and the like. Compositions containing benzoic acid esters of primary alcohols have been used for these purposes in the toiletry and cosmetic industries, for example. DETAILED DESCRIPTION OF THE INVENTION The first aspect of the present invention involves a method for promoting hair growth which comprises topically applying to the scalp of a subject a hair growth enhancing agent comprising at least one benzoic acid ester of primary aliphatic alcohols having from 12 to 15 carbon atoms and a biologically acceptable excipient which does not substantially interfere with the hair growth promotion.
The benzoic acid esters of the present invention can be prepared by reacting benzoic acid with the primary linear alcohol(s) under reaction conditions conducive to benzoate formation. The hair growth enhancing agents of the present invention may be produced as described in U.S. Patent No. 4,275,222 to Scala.
As the biologically acceptable excipient, there is contemplated any non-toxic compound capable of acting as an additional active ingredient, a filler, other carriers or vehicles which impart the desired flow characteristics to the topical composition containing the hair growth enhancing agent and the like without substantially interfering with the activity of the hair growth enhancing agent. As additional active ingredients, there are contemplated compounds which are also capable of promoting the growth of hair. Also as additional^ active ingredients, there are contemplated any compounds capable of treating other scalp conditions. Exemplary of' such compounds are anti-seborrhea and anti-psoriasis agents such as chloroxine, sodium sulfacetamide/sulfur and anthralin.
As filler, there is contemplated any pharmaceutically inert compound which is added to the composition to provide bulk. Exemplary of such compounds are inert clay compounds and talc.
The hair growth enhancing agent of this invention may be applied to the scalp of a patient or subject in any dosage form capable of permitting intimate contact of the hair growth enhancing agent with the scalp. For example, the hair growth enhancing agent may be administered in lotion, cream or ointment dosage form. The disparate flow characteristics of dosage forms is a result of the properties of the compounds contained in the carrier or vehicle.
In cream compositions, acceptable excipients include water, stearic acid, stearic alcohol, propylparaben , methylparaben, glycerin, carbitol, microcrystalline cellulose, polyethylene glycol and the like. In lotion dosage forms, acceptable excipients include water, alkylaryl sulfonic acid salts, hydroxyethylcellulose, propylene glycol, silicon emulsions
(e.g. simethicone and cyclomethicone) , xantham gum and the like. In ointment compositions, acceptable excipients include anhydrous lanolin, dimethicone, petrolatum, polyethylene glycol, white wax and the like.
The benzoic acid ester hair growth enhancing agent is present in the dosage form in an amount sufficient to impart hair growth promoting activity to the dosage form at a reasonable dosage without disturbing the homogeneity of the dosage form. Thus, the present invention contemplates the presence of the benzoic acid ester hair growth enhancing agent in an amount from about 0.5 to about 25 percent by weight of the pharmaceutical topical agent. Preferably, the hair growth enhancing agent is present in an amount ' from about 1.0 to about 10 percent by weight of the pharmaceutical topical agent. Still more preferably, the hair growth enhancing agent is present in an amount from about 2.5 to about 5.0 percent of the pharmaceutical topical agent.
The dosage form of the present invention is applied in such an amount as to provide a sufficient amount of the hair growth enhancing agent to the scalp area to promote the growth of hair. Dosages contemplated by the present invention range from about 0.05 to about 10 ml of the dosage form per day. Preferred dosages range from about 0.1 to about 1.0 ml of the dosage form per day. Such a dosage may be applied once daily or, if desired, in divided dosages.
One preferred embodiment of the first aspect of the present invention provides a hair growth enhancing agent comprising from about 10 to about 35 percent by weight of the C}_2 alcohol benzoate. Another preferred embodiment of this aspect provides a hair growth enhancing agent comprising' from about 25 to about 45 percent by weight of the C13 alcohol benzoate. A third preferred embodiment of this aspect provides a hair growth enhancing agent comprising from about 15 to about 30 percent by weight of the C14 alcohol benzoate. Another preferred embodiment provides a hair growth enhancing agent comprising from about 15 to about 25 percent by weight of the C15 alcohol benzoate. A further preferred embodiment of the first aspect of the present invention involves a method for promoting hair growth which comprises topically applying to the scalp of a subject a hair growth enhancing agent comprising at least two benzoic acid esters of primary alcohols having from 12 to 15 carbon atoms as said at least one benzoic acid ester. One more preferred form of this embodiment of the present invention provides a hair growth enhancing agent comprising from about 10 to about 35 percent by weight of the C^ alcohol benzoate. Another more preferred form of this embodiment provides a hair growth enhancing agent comprising from about 25 to about 45 percent by weight of the Cι_3 alcohol benzoate. A third more preferred form provides a hair growth enhancing agent comprising from about 15 to about 30 percent by weight of the C14 alcohol benzoate. Another more preferred form provides a hair growth enhancing agent comprising from about 15 to about 25 percent by weight of the C^ζ alcohol benzoate.
A further preferred embodiment of the present invention includes a method for promoting hair growth which comprises topically applying to the scalp of a subject a hair growth enhancing agent comprising at least three benzoic acid esters of primary alcohols having from 12 to 15 carbon atoms as said at least one benzoic acid ester. A more preferred form of this embodiment of the present invention provides a hair growth enhancing agent comprising from about 10 to about 35 percent by weight of the C12 alcohol benzoate. Another more preferred form of this embodiment provides a hair growth enhancing agent comprising from about 25 to about 45 percent by weight of the Cτ_3 alcohol benzoate. A third more preferred form provides a hair growth enhancing agent comprising from about 15 to about 30 percent by weight of the C14 alcohol benzoate. Another more preferred form provides a hair growth enhancing agent comprising from about 15 to about 25 percent by weight of the C]_5 alcohol benzoate.
The final embodiment of the first aspect of-the present invention provides a method for promoting hair growth which comprises topically applying to the scalp of a "subject a hair growth enhancing agent comprising all four benzoic acid esters of primary alcohols having from 12 to 15 carbon atoms as said at least one benzoic acid ester. A more preferred form of this embodiment of the present invention provides a hair growth enhancing agent comprising from about 10 to about 35 percent by weight of the C^ alcohol benzoate. Another more preferred form of this embodiment provides a hair growth enhancing agent comprising from about 25 to about 45 percent by weight of the 0^3 alcohol benzoate. A third more preferred form provides a hair growth enhancing agent comprising from about 15 to about 30 percent by weight of the C^4 alcohol benzoate. Another more preferred form provides a hair growth enhancing agent comprising from about 15 to about 25 percent by weight of the C15 alcohol benzoate.
The second aspect of the present invention provides a kit containing a topical medication suitable for promoting hair growth on the scalp of a subject which comprises a package containing: (a) a container including a topical medication comprising a hair growth enhancing agent comprising at least one benzoic acid ester of primary alcohols having from 12 to 15 carbon atoms and containing a biologically acceptable excipient which does not substantially interfere with the hair growth promotion for topical administration to the scalp of said subject; and (b) directions for administration of said topical medication to said scalp for the promotion of hair growth said topical medication upon continued application to said scalp promoting the growth of hair thereon. The container may be a standard pharmaceutical container such as a bottle, tube or jar. Directions are provided explaining that the topical medication contains benzoic acid ester(s) which constitute the active ingredient in the kit, and that the topical medication is to be topically administered to the scalp of a patient wishing to promote hair growth directly to those areas where growth is desired. Such directions are set forth on a label which is attached directly to the pharmaceutical container. Alternatively, the container may be a box or other cardboard, plastic or similar container having therein both a package insert with instructions on how to use the topical medication as a topical hair growth promoting agent together with an inner container housing the topical medication.
One preferred embodiment of the second aspect of the present invention provides a hair growth enhancing agent comprising from about 10 to about 35 percent by weight of the C^ alcohol benzoate. Another preferred embodiment of this aspect provides a hair growth enhancing agent comprising from about 25 to about 45 percent by weight of the C]_3 alcohol benzoate. A third preferred embodiment of this aspect provides a hair growth enhancing agent comprising from about 15 to about 30 percent by weight of the C-j_ alcohol benzoate. Another preferred embodiment provides a hair growth enhancing agent comprising from about 15 to about 25 percent by weight of the C15 alcohol benzoate.
The following are representative of the present invention.
EXAMPLE 1 A hair growth enhancing agent comprising 25% by weight of the benzoic acid ester of the C12 primary linear alcohol (1-dodecanol) ,. 40% by weight of the benzoic acid ester of the C13 primary linear alcohol (tridecanol) , 20% by weight of the benzoic acid ester of the C14 primary linear alcohol (tetradecanol) and 15% by weight of the benzoic acid ester of the C15 primary linear alcohol (pentadecanol) is prepared according to the process disclosed in U.S. Patent 4,275,222. 10 ml of the resultant hair growth enhancing agent is admixed with 240 ml of si ethicone and 50 ml of cyclomethicone to form a pharmaceutically acceptable lotion. EXAMPLE 2
0.30 ml of the lotion of Example 1 is applied to the exposed scalp of a patient once daily in a therapeutic program to promote hair growth.
EXAMPLE 3 A hair growth enhancing agent comprising 30% by weight of the benzoic acid ester of the C12 primary linear alcohol (1-dodecanol) , 35% by weight of the benzoic acid ester of the C13 primary linear alcohol (tridecanol) , 20% by weight of the benzoic acid ester of the C14 primary linear alcohol (tetradecanol) and 15% by weight of the benzoic acid ester of the C]_5 primary linear alcohol (pentadecanol) is prepared according to the process disclosed in U.S. Patent 4,275,222. 10 ml of the resultant hair growth enhancing agent is admixed with 120 ml of anhydrous lanolin, 80 ml of polyethylene glycol, 30 ml of mineral oil and 20 ml of white wax to form a pharmaceutically acceptable ointment.
EXAMPLE 4 0.35 ml of the ointment of Example 3 is applied to the exposed scalp of a patient once daily in a therapeutic program to promote hair growth.
EXAMPLE 5 A hair growth enhancing agent comprising 25% by weight of the benzoic acid ester of the C12 primary linear alcohol (1-dodecanol) , 30% by weight of the benzoic acid ester of the C^3 primary linear alcohol (tridecanol) , 25% by weight of the benzoic acid ester of the C^4 primary linear alcohol (tetradecanol) and 20% by weight of the benzoic acid ester of the C15 primary linear alcohol (pentadecanol) is prepared according to the process disclosed in U.S. Patent 4,275,222. 10 ml of the resultant hair growth enhancing agent is admixed with 90 ml methylparaben, 80 ml stearyl alcohol, 30 ml propylparaben 20 ml beeswax and 40 ml water to form a pharmaceutically acceptable cream. EXAMPLE 6
0.25 ml of the cream of Example 5 is applied to the exposed scalp of a patient once daily in a therapeutic program to promote hair growth.
EXAMPLE 7 250 ml of the lotion of Example 1 is placed in a bottle having affixed thereto a label stating directions explaining that the bottle contains a topical medication which includes benzoic acid ester compounds as the active ingredient, and that the topical medication is to be topically administered to the scalp of a patient to promote hair growth of the patient.
EXAMPLE 8 250 ml of the lotion of Example 1 is placed in a bottle which is housed inside a box. A package insert stating directions explaining that the bottle contains a topical medication which includes benzoic acid ester compounds as the active ingredient, and that the topical medication is to be topically administered to the scalp of a patient to promote hair growth of the patient is also placed inside a box to complete the pharmaceutical kit.

Claims

WHAT IS CLAIMED IS:
1. A method for promoting hair growth which comprises topically applying to the scalp of a subject a hair growth enhancing agent comprising at least one benzoic acid ester of a primary alcohol having from 12 to 15 carbon atoms and containing a biologically acceptable excipient which does not substantially interfere with the hair growth promotion.
2. A method of claim 1, wherein said hair growth enhancing agent contains from about 10 to about 35 percent by weight of the C12 alcohol benzoate.
3. A method of claim 1, wherein said hair growth enhancing agent contains from about 25 to about 45 percent by weight of the C13 alcohol benzoate.
4. A method of claim 1, wherein said hair growth enhancing agent contains from about 15 to about 30 percent by weight of the C14 alcohol benzoate.
5. A method of claim 1, wherein said hair growth enhancing agent contains from about 15 to about 25 percent by weight of the C15 alcohol benzoate.
6. A method for promoting hair growth of claim 1 which comprises topically applying to the scalp of a subject a hair growth enhancing agent comprising at least two benzoic acid esters of primary alcohols having from 12 to 15 carbon atoms.
7. A method of claim 6, wherein said hair growth enhancing agent contains from about 10 to about 35 percent by weight of the C12 alcohol benzoate.
8. A method of claim 6, wherein said hair growth enhancing agent contains from about 25 to about 45 percent by weight of the C^ alcohol benzoate.
9. A method of claim 6, wherein said hair growth enhancing agent contains from about 15 to about 30 percent by weight of the C.4 alcohol benzoate.
10. A method of claim 6, wherein said hair growth enhancing agent contains from about 15 to about 25 percent by weight of the C]_5 alcohol benzoate.
11. A method for promoting hair growth of claim 1 which comprises topically applying to the scalp of a subject a hair growth enhancing agent comprising at least three benzoic acid esters of primary alcohols having from 12 to 15 carbon atoms.
12. A method of claim 11, wherein said hair growth enhancing agent contains from about 10 to about 35 percent by weight of the C12 alcohol benzoate.
13. A method of claim 11, wherein said hair growth enhancing agent contains from about 25 to about 45 percent by weight of the C]_3 alcohol benzoate.
14. A method of claim 11, wherein said hair growth enhancing agent contains from about 15 to about 30 percent by weight of the C^ alcohol benzoate.
15. A method of claim 11, wherein said hair ' growth enhancing agent contains from about 15 to about 25 percent by weight of the C^5 alcohol benzoate.
16. A method for promoting hair growth of claim 1 which comprises topically applying to the scalp of a subject a hair growth enhancing agent comprising all four benzoic acid esters of the primary alcohols having from 12 to 15 carbon atoms.
17. A method of claim 16, wherein said hair growth enhancing agent contains from about 10 to about 35 percent by weight of the C^2 alcohol benzoate.
18. A method of claim 16, wherein said hair growth enhancing agent contains from about 25 to about 45 percent by weight of the C13 alcohol benzoate.
19. A method of claim 16, wherein said hair growth enhancing agent contains from about 15 to about 30 percent by weight of the C14 alcohol benzoate.
20. A method of claim 16, wherein said hair growth enhancing agent contains from about 15 to about 25 percent by weight of the C15 alcohol benzoate.
21. A kit containing a topical medication suitable for promoting hair growth on the scalp of a subject which comprises a package containing:
(a) a container including a topical medication comprising a hair growth enhancing agent comprising at least one benzoic acid ester of primary alcohols having from 12 to 15 carbon atoms and containing a biologically acceptable excipient which does not substantially interfere with the hair growth promotion for topical administration to the scalp of said subject; and
(b) directions for administration of said topical medication to said scalp for the promotion of hair growth said topical medication upon continued application to said scalp promoting the growth of hair thereon.
22. A kit of claim 21, wherein said topical medication contains from about 10 to about 35 percent by weight of the C^2 alcohol benzoate.
23. A kit of claim 21, wherein said topical medication contains from about 25 to about 45 percent by weight of the
C13 alcohol benzoate.
24. A kit of claim 21, wherein said topical medication contains from about 15 to about 30 percent by weight of the C]_4 alcohol benzoate.
25. A kit of claim 21, wherein said topical medication contains from about 15 to about 25^percent by weight of the C]_5 alcohol benzoate.
