DESCRIPTIONDESCRIPTION
The present invention relates to a method for depositing an analyte solution on a support, a device for carrying out the method, a solid support produced according to this process and a diagnostic kit using such a support.
In recent years, numerous methods have been developed for the attachment of analytes, in particular nucleic acids or peptides, to solid supports, in particular for the manufacture of biochips.
A first type of method consists in carrying out in situ the synthesis of the analytes to manufacture a biochip. To this end, US-A-5,744,305 describes a method of assembling oligonucleotides by photolithography from nucleotides protected by a photolάbile group. Patent application WO-A-95 / 25,116 thus describes the ink jet technique for bringing the reagents for synthesis of oligonucleotides onto the support. These methods make it possible to prepare biochips with a high density of analytes on the support but problems of purity of the analytes can arise, linked in particular to the limits of the yields of synthesis of the analytes.
A second type of method consists in preparing in advance the different analytes which can be thus purified, before distributing them on the solid support in discrete positions. To this end, US-A-6,110,426 proposes a method for producing such biochips, using a capillary which is brought directly into contact on a solid surface to deliver a controlled volume of liquid. Effective contact between the end of the capillary and the solid support is then necessary for the drop to be deposited by capillarity. Likewise, US-A-6,083,763 describes a method and an apparatus for the production of biochips, by a set of capillaries sliding in a device so as to compensate for the differences in height of each of them. The deposition of specific oligonucleotides is carried out by capillarity, by contact between the capillaries and the planar surface of the biochip.
Another technique can consist in inducing the deposit of liquid by a mechanical shock when the device containing the liquid comes into contact with the support. Thus, in patent application WO-A-00/25923, the deposition of a small volume of liquid is induced by the deceleration observed during contact between the device containing the liquid and the support intended to receive the liquid.
These contact methods, whether by capillarity or during a mechanical shock, imply great precision in the movement of the capillary and perfectly flat solid supports. In addition, the device allowing the deposit of the drop is always in contact with the support receiving the drop, which poses contamination problems, particularly in the field of biology, problems which it is desired to avoid.
Other techniques solve this problem of contact such as for example the patent US-A-6,083,762 which proposes a drop distribution system, comprising a microdispenser coupled to a piezoelectric transducer to eject drop volumes lower than the nanoliter on a solid surface. A similar result is obtained by applying a hot source on the wall of a capillary to form a bubble which ejects a defined volume of solution (see T. Okamoto et al., Nature Biotechnology, 18, p. 438-441, 2000) .
However, these techniques require complex equipment, and therefore a very high manufacturing cost. In addition, it is not necessary to have a deposit volume as low as the picoliter and a volume of the order of nanoliter is sufficient. Although the results obtained in the prior art are satisfactory and allow the deposition of very small quantities of solution of the order of a picolitre, there is a great need for simple techniques to allow the deposition of analyte solutions on support, without risk of contamination.
Selon un deuxième mode de réalisation, l'axe est :To this end, the present invention describes a method, which solves the problems of the prior art mentioned above, by proposing a method of depositing a solution containing at least one analyte on a solid support V which comprises the following steps: a) loading the solution into at least one reservoir of a device, each reservoir being provided at one end with an outlet orifice of dimensions adapted so that the solution does not flow by gravity, when the tank is at rest. b) cause the displacement of the reservoir between two positions, a so-called starting position and a so-called arrival position, along an axis, and c) stop the displacement when said reservoir is in the arrival position, without contact between the reservoir and the solid support, to create a mechanical shock on the reservoir and deposit, via the outlet orifice, a drop of solution on the solid support, said drop moving in the air before being deposited on the solid support. According to a first embodiment, the axis is in a plane different from the horizontal, preferably substantially vertical and the starting position is a high position and the arriving position a low position. According to a variant of this first embodiment, the reservoir comprises at least one vent, and / or is at controlled pressure. According to a second embodiment, the axis is:
• horizontal, ou• horizontal, or
In a plane different from the horizontal, preferably substantially vertical, with the starting position in the low position and the arrival position in the high position,
According to a variant of this second embodiment, the reservoir comprises at least one venting, said reservoir having internal dimensions allowing the solution to be maintained by capillarity, and / or is at controlled pressure.
The displacement of the reservoir can be caused by any means such as gravity, a spring or any type of known actuator. The position of arrival of the tank is determined using a stop means. The function of the stop means is to stop the movement of the tank with or without prior braking, and therefore to create a mechanical shock suddenly. In particular, there is no deformation of the tank during the mechanical shock. The tank set in motion between the starting position and the arrival position undergoes upon impact on the stop a negative acceleration whose value depends on the initial conditions of the movement, the rigidity of the supporting structure and the materials in contact at the level of the stop during impact.
By “analyte” is meant any compound whose detection and / or identification and / or quantification is of interest, in particular the analyte has at least one recognition site allowing it to react with a target molecule d 'biological interest. By way of example, there may be mentioned as analytes the polynucleotides, the antigens, the antibodies, the peptides, the proteins, the haptens, the sugars, the ions, the metals. In a first variant, the analytes can be placed directly in solution. In a second variant, the analytes can be fixed on particles such as latexes or colloids (see for example the patent or the patent applications of the applicant FR-B-2,773,416 or FR-A-00/00799 or WO -A- 00/39587 or WO-A-99/67641, provided that the particles are dispersed in an aqueous medium).
Des matériaux naturels, de synthèse, modifiés ou non chimiquement, peuvent être utilisés comme support solide, notamment :The term "solid support" as used in this invention includes all materials on which an analyte can be immobilized. Natural materials, synthetic, modified or not chemically, can be used as solid support, in particular:
• des polymères, tels que polychlorure de vinyle, polyéthylène, polystyrène, polyacrylate, polyamide, ou copolymères à base de monomères vinyle aromatiques, alkylesters d'acides - ou β-insaturés, esters d'acides carboxyliques insaturés, chlorure de vinylidène, diènes ou composés présentant des fonctions nitriles (acrylonitrile) ; des polymères de chlorure de vinyle et de propylène, polymère de chlorure de vinyle et acétate de vinyle ; copolymères à base de styrènes ou dérivés substitués du styrène ; • des fibres synthétiques, telles que le nylon, la nitrocellulose ;• polymers, such as polyvinyl chloride, polyethylene, polystyrene, polyacrylate, polyamide, or copolymers based on aromatic vinyl monomers, alkyl esters of - or β-unsaturated acids, esters of unsaturated carboxylic acids, vinylidene chloride, or dienes compounds with nitrile functions (acrylonitrile); polymers of vinyl chloride and propylene, polymer of vinyl chloride and vinyl acetate; copolymers based on styrenes or substituted derivatives of styrene; • synthetic fibers, such as nylon, nitrocellulose;
• des matériaux inorganiques, tels que la silice, le verre, la céramique, le quartz ;• inorganic materials, such as silica, glass, ceramic, quartz;
• des latex, des particules magnétiques ;• latexes, magnetic particles;
The solid support according to the invention can be, without limitation, in the form of a microtiter plate, a sheet, a tube, a well, particles or a flat support such as a wafer of silica or silicon. Preferably, at least one part of the support is flat like a silicon wafer " or the bottom of a well of a microtiter plate. The solid support is hydrophilic and / or hydrophobic depending on the applications and the nature of the solution containing the analyte (s) For example, hydrophilic zones are used for deposition, said hydrophilic zones being delimited by hydrophobic zones, which allows better control of the diameter of the spots.
In particular, the tank has an elongated shape to allow a set of tanks to be associated in a reduced space. For example, reservoirs, as described in patents US-A-6,083,763, ' US-A-6,110,426 or in the patent application
EP-A-0.955.084 sont utilisables.EP-A-0.955.084 can be used.
When there are several reservoirs, in other words at least two reservoirs, this variant makes it possible in particular to manufacture a biochip, that is to say a solid support comprising several spots, in other words at least two spots each containing a different analyte attached to the solid support, said support having a generally smaller than 1 cm 2, and the analytes are distributed over the solid support in discrete positions. The reservoir also allows, by its capacity, the successive deposition of several drops.
The outlet orifice of said reservoir is dimensioned so that the liquid does not flow by gravity but flows by the action of a mechanical shock on the reservoir. The dimensioning of the orifice also makes it possible to control the volume of the drop ejected from the reservoir, that is to say it makes it possible to calibrate the volume of solution leaving the reservoir. Of course, the dimension of the outlet orifice is a function of various parameters, such as the density of the analyte solution, its viscosity, the hydrophilicity or the hydrophobicity of said solution and that of the material used for the manufacture of the tank and the outlet, negative acceleration (deceleration) when the tank stops in its arrival position, and finally the variation in height of the liquid column in the tank.
In an alternative embodiment, the height of liquid in the reservoir is limited to 20 mm, advantageously is between 10 and 15 mm in the case where the top of the column of liquid is at atmospheric pressure. If the tank is blocked, it is possible to increase the height of liquid in the tank, possibly with temperature and / or pressure control.
In another variant of the invention, the reservoir comprises, in addition to its outlet orifice, an air vent limiting the phenomena of suction which could take place during the impact. This venting therefore favors the filling of the reservoir and the deposition of the drop.
Still according to a variant of the invention, the reservoir is at controlled pressure.
In all cases, there is no contact between the device and the solid support on which we want to deposit the solution. Likewise, there is no contact between the volume of solution to be dispensed, that is to say the drop, when the latter is released from the reservoir, and the solid support, which avoids the capillarity problems of the state of the art, for depositing the drop on the solid support. The drop therefore performs a movement in the air present between the space separating the outlet orifice of the reservoir, when the latter is in the arrival position, and the solid support. By "air" is meant any gaseous medium which may be air but also a gas or a mixture of inert gases such as nitrogen and / or argon.
In an alternative embodiment of the invention, the movement of the reservoir is substantially vertical with respect to the horizontal plane of the support and the liquid moves by the combined effect of mechanical shock and gravity. Preferably, the displacement is vertical.
'The orifice can be of any shape, provided that the analyte solution does not flow by gravity when the reservoir is at rest or when the reservoir is moved from the arrival position to the departure position ( in the case of resetting the device). It is this complete definition that should be given to the expression "when the tank is at rest". For example, the orifice is a hole whose cross section is a diameter or a quadrilateral, preferably a square, of dimension (diameter or sides) between 10 and 500 micrometers (μm), preferably between 25 and 250 μm . '
In a preferred embodiment of the method, the movement acts on at least two tanks and the movement of the tanks is simultaneous.
Preferably, each reservoir contains different analytes.
In a particular embodiment of the method, the method comprises an additional step d), which consists in moving the reservoir (s) (1) from the arrival position to a departure position, identical or different from the previous one, without the solution flows by gravity.
In another particular embodiment of the method, the method comprises an additional step e), which consists in repeating steps a) to c) at least once, and / or steps b) and c) steps a) to d), and / or steps b) to d).
In addition, before, during or after operation e), each reservoir and the support are displaced relative to each other, so that the displacement generates the deposit of drop (s) whose position is independent. and without contact with any other drop previously deposited. The method thus makes it possible to deposit a set of drops on a surface. Thus in the process, the spots, generated by the drops after the solutions have dried on the solid support, do not touch. For example, the device is positioned offset from the previous position, this offset being equal to at least once the diameter of a spot on the solid support.
In a first variant, the method makes it possible to deposit the same analyte at discrete positions on the solid support in order to carry out a multiplicity of tests using the same analyte.
In a second variant, the method makes it possible to deposit different analytes in discrete and different positions on the solid support in order to carry out, for example, a multidetection test. An example of a multidetection test is given for example in the patent application of the applicant WO-A-93/02213 for HLA typing with a multiplicity of analytes which are capture oligonucleotides. Other examples of tests are given in the field of genetics such as patent application EP-A-0.412.883 or in the field of infectious with patent applications WO-A-96/00298, WO-A -97/40193, WO-A-97/27332. In a third variant of the method, the device is positioned above a spot previously deposited and the deposition of a second drop makes it possible to increase the density of analytes on the surface.
In a fourth variant, the method makes it possible to deposit at least two different analytes on the same spot from at least two different solutions. The drops deposited on the solid support are for example dried either in air or in a vacuum box according to methods known to those skilled in the art to allow the preservation of the analytes.
The loading of the reservoir 'with analyte solution is performed extemporaneously or predictive manner, in order to have as many containers as dispensing or deposition configuration. The solutions are loaded manually with, for example, a micropipette or automatically. The reservoirs thus prepared are stored under conditions of stability of the analytes before use.
Dans un mode de réalisation particulièrement intéressant de réalisation, l'étape c), ci-dessus mentionnée s'effectue selon deux processus successifs distincts :In a particularly interesting embodiment, step c), mentioned above, is carried out according to two distinct successive processes:
• tout d'abord freiner le déplacement du ou des réservoirs, sans contact entre le réservoir et le support solide,• first of all slow down the movement of the tank or tanks, without contact between the tank and the solid support,
The present invention also relates to a biochip manufactured according to the method of the invention, that is to say a solid support on which are immobilized at least two different analytes in discrete positions, advantageously at least four different analytes, preferably between four and one hundred different analytes, as well as a diagnostic kit comprising at least one biochip manufactured according to the method.
US- A-4,981,783 ; US-A-5,700,637 ; US-A-5,445,934 ; US-A- 5,744,305 ; US-A-5,807,522).An example of non-limiting application of biochips manufactured according to the method of the present invention relates to diagnosis such as for example immunoassays, whatever their format, by direct analysis or by competition, or indeed detection and / or the quantification of nucleic acids from a "random sample containing target nucleic acids. In a diagnostic test, the biochip is preferably used as a capture device (see the applicant's patent FR-B-2,707. 010 or US-A-4,981,783; US-A-5,700,637; US-A-5,445,934; US-A-5,744,305; US-A-5,807,522).
La présente invention concerne également un dispositif permettant la mise en œuvre du procédé, ci-dessus mentionné, ledit dispositif comprenant :The present invention also relates to a device allowing the implementation of the method, mentioned above, said device comprising:
At least one reservoir containing an analyte solution, each reservoir being provided at one end with an outlet orifice of dimensions adapted so that the analyte solution does not flow by gravity, when the reservoir is at rest,
• au moins un axe le long duquel (ou desquels) chaque réservoir est mobile entre deux positions différentes par rapport à cet axe, une position dite de départ et une position dite d'arrivée, au moins un moyen de butée permettant de stopper le mouvement du ou des réservoirs en position d'arrivée tout en permettant à ladite solution d'analyte de s'écouler sous la forme d'une goutte via l'orifice de sortie, sans contact entre le réservoir et le support solide pouvant recevoir la solution d'analyte.• at least one axis along which (or which) each tank is movable between two different positions relative to this axis, a so-called starting position and a so-called arrival position, at least one stop means making it possible to stop the movement of the reservoir (s) in the arrival position while allowing the said analyte solution to flow in the form of a drop via the outlet orifice, without contact between the reservoir and the solid support capable of receiving the solution analyte.
According to a first alternative embodiment, the device comprises a single axis and at least two tanks, preferably four tanks, which are positioned symmetrically with respect to the axis.
According to a second alternative embodiment, the device comprises at least two axes connected to each other by a bracket, the bracket acting as a support for at least one tank, preferably at least two tanks, and even more preferably at least four tanks. Preferably, the axis or axes are substantially vertical and the starting position is a high position and the arrival position is a low position relative to a vertical frame of reference. The mechanical shock creates a sudden deceleration on the tank which releases a volume of preset solution, such as a drop. The solution flows substantially along the axis defined by gravity. Note that in the device reset movement, when the tank is moved manually or automatically from its low position to its high position, it undergoes an acceleration sufficiently low so that the liquid does not flow by gravity. Likewise, the deceleration suffered by the tank in the high position during resetting is sufficiently low so that the solution does not flow.
The section of the outlet orifice can be of any shape provided that the analyte solution does not flow by gravity and that the volume of solution is calibrated. For example, the orifice is a round or quadrilateral hole, preferably square, with a size between 10 and 500 μm, preferably between 25 and 250 μm, and even more preferably between 40 and 100 μm. For circular orifices, the dimension concerned is the diameter. For non-circular orifices, the dimension concerned is represented by the largest diagonal. In this configuration, the volume of solution per drop is between 0.5 ni and 1 μl, more preferably between 1 ni and 200 ni. The stop means makes it possible to maintain a space between the solid support and the orifice of the reservoir in the arrival position, said space being greater than twice the diameter or the largest diagonal of the orifice. Advantageously, the space is adjustable and between two and ten times the diameter of the orifice. When the tank is moved from the starting position to the arriving position, the movement is abruptly stopped by a stop means.
In an embodiment where the movement to create the mechanical shock is vertical, the height of fall of the tank between the high position and the low position can be several centimeters, but a fall height of a few millimeters is sufficient to create a mechanical shock allowing the release of the drops from the reservoir, if the negative acceleration on the reservoir caused by said shock is sufficient.
According to an alternative embodiment, the device contains at least two reservoirs which have the same positions, high and low, relative to the frame of reference constituted by the vertical axis. The simultaneous movement of the reservoirs makes it possible to deposit a set of drops on the solid support. Preferably, the tanks are integral with each other. According to a first alternative embodiment, the stop means can be carried by the axis or axes.
According to a second variant embodiment, which may be complementary to the first variant, in which the device allows the distribution of drops of solution on a solid support, which consists of the bottom of a container, such as a plate microplate, tube, or other, in this case, the means of abutment is constituted by the edges or the container.
According to a third alternative embodiment, the stop means making it possible to stop the movement of the reservoir or reservoirs is produced by two distinct successive means:
First of all, a means of braking the movement of the tank or tanks, without contact between the tank and the solid support,
According to the third alternative embodiment, the braking means is constituted by a stop which drifts under the action of the movement of the reservoir (s), and the stopping means is constituted by a stop which does not dodge under the action of the movement of said tank or tanks.
As previously mentioned, the present invention also relates to a solid support, on which at least two analytes are immobilized, such a support being produced according to the method described above. The invention also relates to a diagnostic kit comprising such a solid support.

Enfin, la figure 8 représente un diagramme montrant l'amortissement des réservoir en position basse pour un système de butée classique de notre invention, tel que représenté aux figures la à 2c, et pour un système de butée amortie, tel que représenté à la figure 7.Figures 6a and 6b show a photograph of the solution deposition as a function of the size of the orifice drilled in the reservoir according to an embodiment of the method according to the invention. 7 shows a schematic view of another embodiment of the device consisting, on the one hand, of a bracket carrying ten tanks according to Figures la to 2c, and on the other hand, a stop means for stopping the movement of the tanks by two separate successive means: a braking means and a means of stopping the movement. Finally, FIG. 8 represents a diagram showing the damping of the tanks in the low position for a conventional stop system of our invention, as shown in FIGS. 1a to 2c, and for a damped stop system, as shown in FIG. 7.
Figure la represents a simplified device illustrating the operation of the invention. The reservoir 1 consists of a tube, for example plastic. An orifice 3 is made at the bottom of the tube. The upper part of the reservoir 1 is provided with a collar which extends radially and perpendicularly to the vertical wall of the tube. This flange acts as a stop 5, as will be explained below. Figure lb describes a container II, which can cooperate with a tank 1, according to Figure la. This container 11 is constituted by a flat-bottomed tube. The bottom and the side wall of said container 11 respectively constitute a solid support 6 and a flange 17. In the case presented in FIG. 2a, the cooperation between the container 1, presented in FIG. 1a, and the container 11 is better understood. shown in Figure lb. In this figure, the cooperation is in the starting position, which is a high position in the present case but it is not compulsory. Said container 11 has the shape of a microtiter plate well and the stop 5 is opposite the rim 17. The reservoir 1 is then filled with a solution 2, containing for example an analyte of interest that the user wants to deposit on the support 6, In this figure 2b, it is clear that said stop 5 comes into contact with the upper rim 17 of the well. The deposition is carried out by applying a force F to move the reservoir F. The mechanical shock is created by this contact between stop 5 and rim 17. In this embodiment, the lower part of the reservoir 1, on which the orifice is located outlet 3 does not touch the solid support 6, that is to say that a drop 7 will be able to transit via a fluid different from the solution 2 from the orifice 3 to the support 6. It therefore exists, like this is well represented in FIG. 2c, a spacing 13 between said orifice 3 and said support 6, which is of sufficient size to allow the drop of the drop 7 to drop. In the latter figures, the cooperation is in the arrival position, which is a low position in this case but it is not mandatory.
Note that the tank 1 can return to the high position, according to Figure 2a. For this, any device known to a person skilled in the art, allowing a return to the starting position, such as a manual action or an automated action, such as an action provided by a spring, a jack, an elastic, a cam, can be considered.
Le trou a un diamètre de 50 μm.Many alternative embodiments are possible without departing from the scope of the present invention. The only principle to be observed is that the outlet 3 is dimensioned so that the liquid 2 does not flow by gravity, but by the action of a mechanical shock on the tank 1 and that said outlet 3 is sized so that the volume of solution ejected is calibrated. In any case, there is never contact between the liquid 2, still located in the reservoir 1, and the solid support 6, on which we want to deposit the drop of solution 7. Said drop 7 therefore moves in the air. The spacing 13 between the edge of the orifice of the reservoir 3 and the solid support 6 is at least greater than twice the diameter of the orifice. FIG. 3a shows another embodiment of the invention to allow the deposition on a solid support 6 of four different solutions 2 containing analytes of interest. In this figure, the set of tanks 1, called the dispensing head 20, is in the high position, while in FIG. 3b, it is in the low position that the head 20 is found, following its fall, along C, by simple gravity. The device comprises four cylindrical tanks 1 mounted vertically around a guide axis 4 and movable along this axis 4. The equidistant positioning of the four tanks is best shown in FIG. 4a, which is a sectional view along AA of the figure 3b. Each reservoir 1 substantially resembles a cylinder whose end, lower in these figures, is closed by a nozzle 8, which is constituted by a ruby 8, pierced with a hole 3, which is located at the end of said reservoir 1 The hole has a diameter of 50 μm.
Comme cela sera expliqué ultérieurement, le moyen de butée 5 est solidaire directement ou indirectement du dispositif,A stop 5 is present at the end of the axis to block the vertical movement of the tanks 1. The operation of the stop 5 appears in FIGS. 3b and 4b, the latter being a detail B taken from FIG. 3b. A counterbore 22, well represented in FIG. 4b, is machined in the lower part of the reservoir 1 which abuts against the means 5. As will be explained later, the abutment means 5 is directly or indirectly secured to the device,
A finger 9 for immobilizing the dispensing head 20 in rotation, carrying the reservoirs 1, is fixed, like the axis 4, on the upper part of the frame 21 of the device. Note that a bearing 23 is also present, to facilitate the movement of this finger 9 and possibly of said axis 4.
In addition, well represented in FIG. 4a, a first notch 26 produced in the upper part of the head 20, where we see the openings allowing the filling of the reservoirs 1, makes it possible to block the rotation relative to the finger 9, and therefore to the frame 21. However, in the lower position of said head 20, as shown in FIG. 3b, the finger can be lifted along S, by everything, in order to allow the head to pivot. 20 along R of FIG. 4a, said finger 9 is then lowered again to come to block said dispensing head 20 again with respect to another notch 26. Note that in this FIG. 4a, it is the notches 26 two in number which are movable and not the finger 9 which is unique is always remains in the same position. The dotted representation of finger 9 is only there to explain the position that it 9 can take after rotation R, of the head 20 relative to the frame 21. These two notches 26 are positioned at an angle of 45 ° l relative to each other in relation to axis 4, that is to say that for the same position of said axis 4, the head 20 can undergo two mechanical shocks in different positions, and thus allow the realization eight spots 29 on the support 6. It is of course obvious that the number and the position of the notches 26 may vary. For example there could be three separated by an angle of 30 °, or four separated by an angle of 22.5 °. However, these angles may not be constant and the arrangement of the orifices 3, relative to the center of the axis 4, may not be identical.
A more complete device is shown in FIG. 5. In this case, the four tanks 1 are held in the high position by a locking means, such as a pawl 10. The set of tanks 1 in the high position is shown in dotted lines on this Fig. When this means 10 is released by a control lever 12, the head 20 falls by the action of gravity to a height of a few millimeters along the axis 4 and the movement of the tanks 1 is blocked in the low position by the stop 5 thus creating a sudden deceleration and therefore a mechanical shock. The locking means 10 and the control lever 12 are integral with one another and are movable about a hinge pin 24 carried by the frame 21. The free end of said lever 12 comprises a stud 27 which can cooperate with a blind hole 28 carried by said frame 21, in order to limit the movement E of the lever 12. The tanks 1, machined from metal, together have a movement of sufficient speed, along the axis 4, to create a mechanical shock on contact with the stop 5, which is also machined from metal. The materials of the reservoir 1 and of the stop 5 as well as the rigidity of the support structure contribute to the intensity of the expected shock. Other materials can be used such as plastic or glass. In the event that the momentum of the tank or the tank assembly (i.e. the product of mass by speed) is insufficient by itself, a means of acceleration such as a spring can be added to the device. The height of movement of the reservoirs 1 is an additional means for creating this increase in the amount of movement, for example using a rack 14, and the precise conditions are determined by a person skilled in the art on a case-by-case basis. The height of fall of the reservoir is therefore variable and adjustable by means of this rack 14. A spacing 13 of 500 μm is present between the flat solid support 6, which receives the drops of solution, and the orifice 3 of the reservoir, not shown in FIG. 5. The four drops 7 of solution 2 fall simultaneously on said solid support 6. This spacing 13 of 500 μm is provided by conical feet 15 which are positioned in the wells 11 of a microtiter plate 25 shown in this figure 5.
To verify the reproducibility of the deposit, the four reservoirs of the device in FIG. 5 are filled with an orange solution G at 5 mg / ml in a 4x PBS buffer (200 mM sodium phosphate, pH 7.0, 600 mM NaCl ). The fall height is set at 10 mm. The solid support 6 consists of the bottom of the wells 11 of a microtiter plate 25 NUNC reference 439454 made of polystyrene. The deposition is repeated eight times on the bottom of a well of the plate. The spot size, measured under a microscope, is between 149 and 179 μm with a coefficient of variation of less than 7% and a standard deviation of less than 10 μm.
It is possible to distribute on a 4 mm diameter disc, delimited in the bottom of a microtiter plate well, sixteen spots of 160 μm in diameter separated by a distance of between 600 and 700 μm without risk of contamination.
With sixteen different analytes on each spot 29, like sixteen capture probes for HLA typing, it is possible to perform typing in a single well 11 by a very simple and inexpensive method. Therefore, it is necessary to put different solutions 2 in the different tanks 1, in order to simultaneously deposit a drop of several different solutions.
A solution of PBS tween colored with orange ink G is introduced into an eppendorf tube (poly lab reference 35341) pierced at its bottom with a hole of variable diameter 50 μm (Figure 6a) or 160 μm (Figure 6b). The tube 1 is impacted against a fixed element, such as the frame of the device, not shown in FIGS. 6a and 6b, via its stop 5, as shown in Figure la, to create a mechanical shock and release drops of solution on a sheet of photographic paper. These figures show that the deposition system is reproducible, for a given orifice diameter 3, even under manual conditions and that the diameter of said orifice 3 makes it possible to vary the volume of solution 2 distributed in the form of a drop 7 and the size of each spot 29. The scale represented in FIGS. 6a to 6b is graduated by values expressed in mm.
It is therefore possible to deposit the nanovolume of solution 2 containing an analyte on a small surface in order to produce a biochip with, for example, different oligonucleotides which allow molecular diagnosis.
La figure 7 montre un dispositif de distribution de gouttes 7, qui est plus perfectionné que les trois dispositifs précédents représentés :FIG. 7 shows a device for dispensing drops 7, which is more sophisticated than the three previous devices shown:
• aux figures la à 2c pour le premier dispositif,• in Figures la to 2c for the first device,
.• Figures 3a to 4b for the second device, and • Figure 5 for the third device, since it includes two innovations. .
Si l'on utilisait des têtes de distribution 20, telles que représentées précédemment, cela ferait quarante spotsThe first improvement relates to the number of tanks 1, or of the dispensing head, implemented during a single mechanical shock. In the present case, there are therefore ten reservoirs 1 and therefore ten spots 29 per impact on the support 6. If we used distribution heads 20, as represented previously, this would make forty spots
29 réalisés lors d'un choc mécanique.29 made during a mechanical shock.
In fact, all of the ten tanks 1 are carried by a bracket 16 which is essentially constituted by:
The lateral uprights 30 allow the physical connection between the lower structure 31 and the upper structure 32. In addition, they are perpendicular to the upper structure 32. In the present case, the height of these uprights 30 substantially defines the clearance height D of the bracket 16. This bracket 16 is moreover movable along two axes 4 located on either side of the latter 16. Each uprights 30 cooperating with an axis 4, acting as a guide, that the mechanical shock in the arrival position of the ten tanks 1 is identical from one tank 1 to the other 1.
The upper structure 32 is substantially horizontal and is pierced with a number of bores, not referenced in the figures, capable of receiving ten reservoirs 1, which are symmetrically arranged. It is of course obvious that for those skilled in the art, any means of fixing the reservoirs 1 at the bores is usable, preferably a means which is removable, such as for example a nut, a pin, a pawl. The lower structure 31 makes it possible to transmit the kinetic energy of the bracket to the frame 21 of the device. This transmission movement will be developed below, in connection with the second improvement.
This second improvement relates to the abutment means 5 which is in two successive parts for damping and blocking in the arrival position the tanks 1, in the low position in this case, it is a "pure shock" which can be defined as a shock without rebound and without vibration of said mobile tanks 1. The two parts, which constitute the stop means 5, allow:
La partie pour freiner 18 est constituée par :• first of all to brake 18 the movement of the reservoir (s) 1, without contact between the reservoir 1 and the solid support 6, • then stop 19 the movement of the said reservoir (s) 1, always without contact between the reservoir 1 and the said support solid 6, braking or stopping creating a mechanical shock on the reservoir (s) and depositing a drop 7 of solution 2 on the solid support 6, said drop moving in the air before depositing on the solid support 6. The part to brake 18 consists of:
• un longeron 33, mobile en pivotement selon P, sous l'action du débattement D de la potence 16,A beam 33, pivotally movable along P, under the action of the movement D of the bracket 16,
• un axe d'articulation 34 reliant le longeron 33 au bâti 21,A hinge pin 34 connecting the beam 33 to the frame 21,
A shock absorber 36 carried by the beam 33, and • a magnet 37 carried by the frame 21, the assembly slipping under the action of the fall or the deflection of said bracket 16. The part for stopping 19 is constituted by a stop integral with the frame 21, which does not not dodge. Finally, FIG. 8 highlights the difference in kinematics between a device provided with a conventional stop (fatty and dotted curve), according to the first three embodiments mentioned, and a device provided with an improved stop (line curve end and continuous), according to the embodiment of FIG. 7.
In the case of the conventional embodiment, the start of the curve is constituted by a straight line with a single slope, while in the improved embodiment, this curve consists of two half straight lines having two different slopes. The first slope is identical to, while the second slope is less inclined than that of the conventional embodiment. Between these two half straight lines, a point PC is noted which corresponds to the point of contact between the lower structure 31 of the bracket 16 and the spherical support 35 of the braking means 18. The right and the second half right then bend to descend .. The tangential point, at the level of the highest points of the two curves, makes it possible to define:
• le temps (ToC) mis pour obtenir un vitesse nulle, ainsi que cette vitesse nulle (NoC), pour le mode de réalisation classique, et • le temps (ToP) mis pour obtenir un vitesse nulle, ainsi que cette vitesse nulle (NoP), pour le mode de réalisation perfectionné.• the time (ToC) taken to obtain a zero speed, as well as this zero speed (NoC), for the conventional embodiment, and • the time (ToP) taken to obtain a zero speed, as well as this zero speed (NoP ), for the improved embodiment.
Les exemples ci-joints sont donnés à titre explicatif , et n'ont aucun caractère limitatif Ils permettront de mieux comprendre l'invention.(top of the rising curves) is stronger for the classic embodiment than for the improved embodiment. This difference in DD deceleration is also clearly visible in this figure. Similarly, after the ToC and ToP, the acceleration speed (top of the descending curves) is stronger once again for the conventional embodiment, which is much less damped than for the improved mode. Moreover, the ripple (an ascent and a descent) which follows the ToC and ToP, is of a greater amplitude for the conventional embodiment than for the improved mode. The examples attached are given for explanatory purposes, and are in no way limiting. They will allow a better understanding of the invention.
This example presents a sandwich test between a recognition molecule fixed on a support, a target molecule which one wishes to highlight, which is specifically fixed on the recognition molecule, and a detection molecule making it possible to visualize the fixation of the target molecule on the recognition molecule. The recognition molecules are deposited on the support according to the invention.
Thus, the solution of recognition molecules is a solution of recombinant P24 proteins (lot pmR RK24H, 14-1-99, bioMérieux, Marcy l'Etoile, France), which is true for the other P24 which will be mentioned later, free in solution, initially prepared at a concentration of 100 mg / ml in PBS buffer. Another solution of recognition molecules, in which the proteins (protein P24) are immobilized on a polymer of maleic anhydride / methyl vinyl ether (AMNE), according to the method described in patent application PCT / FR98 / 01299, is also prepared. The device according to the invention, as presented in FIG. 5, is used to deposit a drop 7 of these two solutions 2 of recognition molecules. Thus, two of the four reservoirs 1, provided with a ruby 8 pierced with a hole 3 with a diameter of 200 μm, are filled with 20 μl of the free protein solution. The other two reservoirs 7 are filled with 20 μl of the protein solution immobilized on an AMNE polymer.
Les -puits sont ensuite rincés en PBS tween 0,05.The following steps are as usually carried out by a person skilled in the art. The wells 11 are thus rinsed in PBS tween 0.05% (v / v), then saturated with a PBS tween 20 buffer at 0.1%, enriched with 10% goat serum and 0.2% milk, for 30 minutes at 37 ° C (200 μl per microplate). The wells are then rinsed in PBS tween 0.05.
Les microplaques 25 sont ensuite incubées lh à 37°C, et rincés en PBS tween 0,05.Five sera from FflN positive patients and five sera from HIN negative patients, diluted to 1/500 in PBS tween 0.05 buffer, constituting the solution 2 of target molecule which it is desired to detect, are deposited in the various wells 11. The microplates 25 are then incubated for 1 h at 37 ° C., and rinsed in PBS tween 0.05.
In each well 11, 100 μl of the solution of detection molecules (solution of antihuman antibodies labeled with peroxidase (1 mg / ml), diluted to 1/10000 in reaction buffer) are added. Incubation takes place during lh at 37 ° C. The wells are then rinsed in PBS tween. To each well 11, 100 μl of colorimetric precipitating substrate for HRP (TMB, bioFX) is then added. The spots 29 in contact with the HIN positive sera are colored blue, while the HIN negative sera by ELOSA do not give any detectable coloration. The spots 29 are analyzed under a white light microscope, using software (Image Pro +), the full scale of which is used, which ranges from 0 (black) to 250 (white) arbitrary units, making it possible to obtain a black and white image of the background of the plate, and determine the average gray level on a window defined by the user.

The prior immobilization of the recombinant P24 on the AMVE polymer tends to improve in 60% of the cases the sensitivity of the reaction thanks to a better accessibility of the protein vis-à-vis the serum antibodies.

Note that the sequences in bold underlined in dotted line correspond to the hybridization of a part of the recognition molecule on a part of the target molecule, while the sequences in bold underlined in solid line correspond to hybridization of the detection molecule to a part, ditlérente the previous one, the, target molecule.
La microplaque est incubée une nuit à 4°C puis lavée deux fois par 300 μl de PBS Tween (0,15 M NaCl, 0,05 M de phosphate de sodium, pH 7 et 0,5% de Tween 20 (Merck)).Initially, the recognition molecules, in solution at a concentration of 1 μM in a PBS PBS 3X buffer medium (0.45 M NaCl, 0.15 M sodium phosphate, pH 7), are discreetly deposited in the bottom of a well of a 96-well microplate (NUNC reference maxisorb) according to the invention. Thus, the device, as presented in FIG. 5, is used to deposit a drop 7 of this solution 2 of recognition molecules. The microplate is incubated overnight at 4 ° C. and then washed twice with 300 μl of PBS Tween (0.15 M NaCl, 0.05 M sodium phosphate, pH 7 and 0.5% Tween 20 (Merck)) .
In a second step, the target molecules, in solution in a PBS salmon buffer medium (PBS 3X + DNA of salmon sperm 10 mg / ml, SIGMA) are added.
Dans un troisième temps, l,5μl d'une solution de molécules de détection (conjugués oligonucléotides-peroxydase) sont ajoutés à une concentration de 50 pmol/ml dans un tampon phosphate pH 7 (Na2HPO4 KH2PO4, BSA 0, 1%)Thirdly, 1.5 μl of a solution of detection molecules (oligonucleotide-peroxidase conjugates) are added at a concentration of 50 pmol / ml in a phosphate buffer pH 7 (Na 2 HPO 4 KH 2 PO 4 , BSA 0 , 1%)
La microplaque est incubée 1 heure à 37°C et lavée par deux fois 300 μl de PBSThe microplate is incubated for 1 hour at 37 ° C. and washed twice with 300 μl of PBS
Tween, puis, 100 μl de substrat colorimétrique et précipitant TMB (BioFx Laboraries,Tween, then, 100 μl of colorimetric substrate and TMB precipitator (BioFx Laboraries,
Rotterdam, Netherlands) are added. After 10 minutes of reaction the reading is carried out on a microscope (ZEISS axioskop II, Germany) equipped with a high definition camera SPOT RT (Diagnostic Instruments, Burroughs, United States of America), and an objective of magnification X4 allowing an image to be made of the entire bottom of the well.
A well serving as a negative control is also produced according to the same protocol, but using as in place of the target molecule, an oligonucleotide of sequence of twenty (20) nucleotides:
SEQ ID No. 4: 5'-TATGAAACTT ATGGGGATAC-3 ', not complementary to the recognition molecule, and of course of the target and detection molecules. The results show that the spots made with the target molecule have a strong blue coloration coming from the enzymatic activity of the peroxidase, while the spots made with the non-complementary oligonucleotide do not have any coloration. The system is therefore specific and does not generate background noise.
